UV-induced matrix metalloproteinases (MMPs) in skin fibroblast lead to the breakdown of collagen and other extracellular matrix proteins and which is related to photoaging of human skin. This study investigated the effect of fucoidan on UV-induced MMP-1 expression in human skin fibroblasts. The cells were pretreated with fucoidan at doses of 1 ㎍/mL, 10 ㎍/mL, 100 ㎍/mL for 24 h. After UVB irradiation, they were then further harvested. Expression of MMP-1 and ERK/JNK/p38 protein were determined by western blotting and that of MMP-1 mRNA by RT-PCR. The cells were pretreated with fucoidan and harvested 72 h later. They were inhibited in the expression of MMP-1 protein dose-dependently, compared to the UVB alone irradiated group. And also, the expression of ERK, JNK and p38 protein was inhibited. Moreover, inhibitions of ERK and JNK by fucoidan resulted in the decrease of c-fos, c-jun and Elk phosphorylation /expression. Also, we studied the effect of fucoidan on UVB-induced MMP-1 expression in mRNA level. The cells pretreated with fucoidan were inhibited in the expression of MMP-1 at mRNA levels in the following order; 50 ㎍/mL> 25 ㎍/mL>10 ㎍/mL. Therefore, these results suggest that fucoidan has anti-photoaging effect in HS68 cells by suppression of ERK/JNK signal pathway.
