영문초록 : Collagen was isolated and characterized from velvet antler of Elk (Cervus canadensis). The total collagen yield extracted was as similar as 12.1% on the basis of lyophilized dry weight and that was as similar as compared to other vertebrates. Velvet antler collagen had similar electrophoretic pattern of typical type I , having at least two α chains: α1, and α2 and α3. It showed that the large amount of type I collagen having heterotrimer subunit composition (α1α2α3). The SDS-PAGE pattern using 6.5% gel, both velvet antler and porcine skin collagen contained inter and intra molecular cross-linked components; β (dimmers) and γ (trimers). Moreover , the denaturation temperature was 39℃, which was about 2℃ higher than that of collagen from porcine skin. Glycine was the major amino acid and that was nearly 42.85% of total amino acids, and the degree of hydroxylation of proline and hydroxyproline of velvet antler collagen was calculated to be 33.42% and 14.93%, respectively. Hydroxylation of proline was fairly higher than those of other collagen. Peptide mapping of velvet antler in elk collagen. was similar to that of porcine skin collagen on electrophoretic pattern . As a result, the primary structure of velvet antler collagen was identical to that of porcine skin collagen. Therefore, there is a possibility to use velvet antler collagen in elk for industrial purposes and subsequently it may maximize the economical value of the animal source.
