MTT분석법을 이용한 자궁경부암 및 난소암 세포주의 성장인자에 관한 연구 = The growth factor study of uterine cervical and ovarian cancer cell lines using MTT assay

국문 초록 (Abstract)

MTT 분석법을 이용하여 자궁경부암 세포주인 HeLa, SNUH-17과 난소암 세포주인 SNU-8에 대하여 12종류의 성장인자들의 영향을 측정하여 다음과 같은 결론은 얻었다. 1. 수립된 세포주를 사용하여, 성장인자의 영향을 측정하는데 있어 MTT 분석법은 빠르고 객 관적인 결과를 얻을 수 있는 검사방법이었다 2. 세 종류 세포주의 성장에 transferrine, insulin그리고 sodium pyruvate는 필수적이었다. 3. 자궁경부암 세포주인 HeLa, SNUG-17에 있어 sodium pyruvate가 세포주의 성장을 가장 촉진하는 성장인자임이 밝혀졌다. 4. 난소암 세포주인 SNU-8에서는 sodium pyruvate의 역할이 미약한 것으로 분석되었다. 이상의 결과에서 향후 부인암세포주들에서 sodium pyruvate의 역할에 대한 고찰이 필요할겠 고 이들 부인암 세포주의 수립 및 유지를 위한 규정배지의 개발에는 sodium pyruvate. transferrin insulin등 3종류의 성장인자들은 필수적으로 포함되어야 한다고 사료된다.

다국어 초록 (Multilingual Abstract)

The establishment of cell lines plays a important role for the study on cellular and molecular biology of gynecologic malignancies. It is well known that defined media is very useful to the establishment of cancer cell lines, so that we must develop effective defined media containing essential growth factors to achieve higher success rate in the establishment of gynecologic cancer cell lines. The effects of various growth factors on gynecologic cancer cell lines were tested using MTT assay. The cell lined tested were SNUG-17 HeLa from human cervical cancer and SNU-8 form human ovarian cancer. The growth factors tested were transferrin, insulin, BSA selenuim, EGF hydrocotrisone, B-estradiol, Triidothyronine, ethanolamine/phosphoryl establishment , soduim pyruvate. The amout of formazan product generated and measured after solubilization in DMSO at 540nm is proportional to cell number although the absolute absorbance for a given cell number vaires between cell lines,. The growth rates of each cell line in microtitier was wells were determined using MTT assay. For each cell line the optimal seeding concentration was determined. This results suggest that the defined media containing sodium pyruvate, transferrin, and insulin are essential to the establishment and culture of gynecologic cancer cell lines. More detailed study is necessary to identify the role of sodium pyruvate as a growth factor in gynecologic cancer cell lines.
