Flavonoids are suggested to possess both anti-oxidant and chelating ability. Effects of rutin and quercetin on the degradation of collagen and depolymerization of hyaluronic acid by Cu^(2+) and H₂O₂ were investigated. Cu^(2+) plus H₂O₂-induced collagen degradation was prevented by the oxygen radical scavengers SOD, catalase, DMSO and DABCO and by the metal chelators EDTA and penicillamine. Rutin and quercetin markedly prevented collagen degradation by Cu^(2+) plus H₂O₂. The decrease of viscosity of hyaluronic acid by H₂O₂ was restored by rutin but not by quercetin. Rutin and quercetin decomposed H₂O₂. These results suggest that rutin and quercetin may prevent the damages of collagen and hyaluronic acid by Cu^(2+) and H₂O₂ by their scavenging action on oxygen radicals and chelating action on Cu^(2+).
