      Tryophagus putrescentiae induces inflammatory cytokine secretions from the human cell lines THP-1 and EoL-1 cells

      The present study aimed to identify the roles of the storage mite Tyrophagus putrescentiae (Tp) in relation to human skin diseases, such as atopic dermatitis. We examined the effects of T. putrescentiae extracts (TpE) on the secretion of inflammatory cytokines and their mRNA expression in human immune cells. Human monocytic THP-1 and eosinophilic EoL-1 cells were challenged with TpE and the inflammatory cytokines, interleukin-6 (IL-6), interleukin-8 (IL-8), and monocyte chemotactic protein-1 (MCP-1) secreted in culture supernatant were analyzed using an enzyme-linked immunosorbent assay (ELISA). The mRNA levels in the cell cultures were also measured via reverse transcriptase-polymerase chain reaction (RT-PCR). In the THP-1 cells, the TpE significantly induced the secretion of IL-6, IL-8, and MCP-1 in a time- and dose-dependent manner. It also stimulated increases in the expression levels of the IL-6, IL-8, and MCP-1 mRNA. However, while the TpE induced IL-8 and MCP-1 mRNA levels in the EoL-1 cells, it did not induce IL-6 expression. Using Western blot analysis, we confirmed the activation of extracellular signal-regulated kinase (ERK), a member of the mitogen-activated protein kinases (MAPKs), in response to TpE challenge of the THP-1 cells.
      Collectively, our results show that TpE challenge elicits inflammatory response in the human monocytic THP-1 and eosinophilic EoL-1 cells, and it induces increased production of the inflammatory cytokines IL-6, IL-8, and MCP-1.
