        Effect of Transgenic Rhizobacteria Overexpressing Citrobacter braakii appA on Phytate-P Availability to Mung Bean Plants

        Rhizosphere microorganisms possessing phytase activity are considered important for rendering phytate-phosphorus (P) available to plants. In the present study, the Citrobacter braakii phytase gene (appA) was overexpressed in rhizobacteria possessing plant growth promoting (PGP) traits, for increasing their potential as bioinoculants. AppA was cloned under the lac promoter in the broadhost- range expression vector pBBR1MCS-2. Transformation of the recombinant construct pCBappA resulted in high constitutive phytase activity in all of the eight rhizobacterial strains belonging to genera Pantoea, Citrobacter, Enterobacter, Pseudomonas (two strains), Rhizobium (two strains), and Ensifer that were studied. Transgenic rhizobacterial strains were found to display varying levels of phytase activity, ranging from 10-folds to 538-folds higher than the corresponding control strains. The transgenic derivative of Pseudomonas fluorescens CHA0, a well-characterized plant growth promoting rhizobacterium, showed the highest expression of phytase (~8 U/mg) activity in crude extracts. Although all transformants showed high phytase activity, rhizobacteria having the ability to secrete organic acid showed significantly higher release of P from Caphytate in buffered minimal media. AppA overexpressing rhizobacteria showed increased P content, and dry weight (shoot) or shoot/ root ratio of mung bean (Vigna radiata) plants, to different extents, when grown in semisolid agar (SSA) medium containing Na-phytate or Ca-phytate as the P sources. This is the first report of the overexpression of phytase in rhizobacterial strains and its exploitation for plant growth enhancement.

        On the Prediction of Grain Refinement Mechanism in Direct Chill Casting of Aluminum and Its Alloys Under Low Degree Mechanical Forced Convection

        The present investigation focuses on grain refinement in pure aluminum and its alloy under mechanical stirring with a verylow degree of forced convection of 800 rpm as compared to reported values of up to 8000 rpm during direct chill castingprocess (DC). Commercially pure aluminum (99.7%) and Al6082 alloy billets were fabricated in an in-house designed DCcasting simulator. Mechanical forced convection (MFC) was applied to the melt during casting by a common four-bladedimpeller. The microstructural properties of the as-cast billets have been analysed to study the effect of MFC during casting andsolidification. The structure of both Al and its alloy is found to be refined in presence of MFC as compared to conventionalDC. A combination of simulation, experimental and theoretical approaches has been used to determine the effect of MFCin melt-temperature distribution to propose the mechanism of grain refinement. It has been observed that MFC prominentlyreduces the temperature gradient and extends undercooling time, resulting in fine grain structure.

        Some approximation properties of modified Szasz-Mirakjan-Baskakov operators

        The generalization of well known Száasz-Mirakjan operators was introduction by G C Jain, Approximation of functions by a new class of linear operators, Journal of the Australian Mathematical Society, 13(3):271-276, 1972. In P Patel and V N Mishra, Jain-Baskakov op- erators and its different generalization, Acta Mathematica Vietnamica, 40:715-733, 2015 introduced the integral modification of Jain operators and discussed its different generalization. In this manuscript, we extend the study of the operators introduced by Patel & Mishra and discussed some direct results in ordinary approximation for this operators.         Age-related changes in hematological and biochemical profiles of Wistar rats

        Background: Wistar rats are extensively used as the model for assessing toxicity and efficacy in preclinical research. Hematological and biochemical laboratory data are essential for evaluating specific variations in the physiological and functional profile of a laboratory animal. Establishing hematological and biochemical reference values for Wistar (han) rats at various age intervals was the goal of this work. Male and female Wistar rats (n = 660) of ages 6–8 weeks, 10–14 weeks and > 6 months were used in the experiment. Blood and serum were collected from these rats under fasting conditions. Results: We observed that the majority of hematological and biochemical parameters were significantly influenced by sex and age. Hematological changes were significantly correlated to aging were increased red blood cells, hemoglobin, hematocrit, neutrophils, monocytes and eosinophils in both sexes, as well as decreased platelet, mean corpuscular volume, mean corpuscular hemoglobin and lymphocytes in both sexes. White blood cells of male rats were considerably higher than those of female rats in all age ranges. For biochemistry, increase in glucose, total protein and creatinine were seen in both sexes, along with increases in urea in females and alanine aminotransferase in males. Age was significantly associated with decreased alkaline phosphatase in both sexes. Conclusions: When using Wistar rats as a model, these reference values may be useful in evaluating the results.

        Hematological and biochemical reference intervals of wild-caught and inhouse adult Indian rhesus macaques (Macaca mulatta)

        Background: Nonhuman primates are used for research purposes such as studying diseases and drug discovery and development programs. Various clinical pathology parameters are used as biomarkers of disease conditions in biomedical research. Detailed reports of these parameters are not available for Indian-origin rhesus macaques. To meet the increasing need for information, we conducted this study on 121 adult Indian rhesus macaques (57 wild-sourced and 64 inhouse animals, aged 3–7 years). A total of 18 hematology and 18 biochemistry parameters were evaluated and reported in this study. Data from these parameters were statistically evaluated for significance amongst inhouse and wild-born animals and for differences amongst sexes. The reference range was calculated according to C28-A3 guidelines for reporting reference intervals of clinical laboratory parameters. Results: Source of the animals and sex appeared to have statistically significant effects on reference values and range. Wild-born animals reported higher WBC, platelets, neutrophils, RBC, hemoglobin, HCT, MCV, and total protein values in comparison to inhouse monkeys. Sex-based differences were observed for parameters such as RBCs, hemoglobin, HCT, creatinine, calcium, phosphorus, albumin, and total protein amongst others. Conclusions: Through this study, we have established a comprehensive data set of reference values and intervals for certain hematological and biochemical parameters which will help researchers in planning, conducting, and interpreting various aspects of biomedical research employing Indian-origin rhesus monkeys.

        Regardless of advances in treatment modalities with the invention of newer therapies, breast cancer remains a major health problem with respect to its diagnosis, treatment and management. This female malignancy with its tremendous heterogeneous nature is linked to high incidence and mortality rates, especially in developing region of the world. It is the malignancy composed of distinct biological subtypes with diverse clinical, pathological, molecular and genetic features as well as different therapeutic responsiveness and outcomes. This inconsistency can be partially overcome by finding novel molecular markers with biological significance. In recent years, newer technologies help us to indentify distinct biomarkers and increase our understanding of the molecular basis of breast cancer. However, certain issues need to be resolved that limit the application of gene expression profiling to current clinical practice. Despite the complex nature of gene expression patterns of cDNAs in microarrays, there are some innovative regulatory molecules and functional pathways that allow us to predict breast cancer behavior in the clinic and provide new targets for breast cancer treatment. This review describes the landscape of different molecular markers with particular spotlight on vitamin D signaling pathway and apoptotic specific protein of p53 (ASPP) family members in breast cancer.

        Adaptive smith predictor controller for total intravenous anesthesia automation

        Anesthetic agent propofol needs to be administered at an appropriate rate to prevent hypotension and postoperative adversereactions. To comprehend more suitable anesthetic drug rate during surgery is a crucial aspect. The main objective of thisproposal is to design robust automated control system that work effi ciently in most of the patients with smooth BIS andminimum variations of propofol during surgery to avoid adverse post reactions and instability of anesthetic parameters. And also, to design advanced computer control system that improves the health of patient with short recovery time and lessclinical expenditures. Unlike existing research work, this system administrates propofol as a hypnotic drug to regulate BIS,with fast bolus infusion in induction phase and slow continuous infusion in maintenance phase of anesthesia. The novelty ofthe paper lies in possibility to simplify the drug sensitivity-based adaption with infusion delay approach to achieve closedloopcontrol of hypnosis during surgery. Proposed work uses a brain concentration as a feedback signal in place of the BISsignal. Regression model based estimated sensitivity parameters are used for adaption to avoid BIS signal based frequentadaption procedure and large off set error. Adaptive smith predictor with lead–lag fi lter approach is applied on 22 diff erentpatients’ model identifi ed by actual clinical data. The actual BIS and propofol infusion signals recorded during clinical trialswere used to estimate patient’s sensitivity parameters EC 50 and λ . Simulation results indicate that patient’s drug sensitivityparameters based adaptive strategy facilitates optimal controller performance in most of the patients. Results are obtainedwith proposed scheme having less settling time, BIS oscillations and small off set error leads to adequate depth of anesthesia. A comparison with manual control mode and previously reported system shows that proposed system achieves reductionin the total variations of the propofol dose. Proposed adaptive scheme provides better performance with less oscillation inspite of computation delay, surgical stimulations and patient variability. Proposed scheme also provides improvement inrobustness and may be suitable for clinical practices.

      • Association between p53 Gene Variants and Oral Cancer Susceptibility in Population from Gujarat, West India

        Background: p53 gene variants i.e. 16 bp duplication in intron 3, Arg72Pro in exon 4 and G>A in intron 6 have been reported to modulate susceptibility to various malignancies. Therefore, the present study evaluated the role of these p53 polymorphisms in oral cancer susceptibility in a population from Gujarat, West India. Method: Genotype frequencies at the three p53 loci in 110 controls and 79 oral cancer cases were determined by the PCR-RFLP method. Results: Heterozygous individuals at exon 4 showed protection from developing oral cancer. Homozygous wild and heterozygous individuals at intron 3 and those heterozygous at exon 4 in combination appeared to be at lowered risk. Furthermore, carriers of the 16 bp duplication allele at intron 3, proline allele at exon 4 and G allele at intron 6 were protected from oral cancer development. Conclusion: p53 polymorphisms, especially Arg72Pro in exon 4 could significantly modify the risk of oral cancer development in Gujarat, West Indian population.

        Concomitant variations of the tibialis anterior, and extensor hallucis longus, and extensor hallucis brevis muscles

        Tibialis anterior (TA) muscle originates from the lateral surface of tibia and its tendon attaches to the medial cuneiform and base of the first metatarsal. The TA muscle is responsible for both dorsiflexion and inversion of the foot. We present a case of bilateral TA muscle variations that diverge slightly from the current classification systems of this muscle. Recognizing variations such as these may be important for anatomists, surgeons, podiatrists, and physicians. Following routine dissection, an accessory tendon of the TA muscle was found on both sides. Accessory tendons of the extensor hallucis longus and extensor hallucis brevis joined to form a common tendon on both sides. We believe that this unique case will help further the classification systems for the tendons of the TA and also be informative for clinical anatomists as well as physicians treating patients with pathology in this region.
