        갈대 생육지에서 질소 시비가 사초의 생육과 수량 및 사료가치에 미치는 영향

        본 연구는 질소(N) 시비가 부존 조사료자원인 갈대(Phragmites communis)의 생육과 수량 및 사료가치에 미치는 영향을 구명하고자 2012년에 충남 천안과 경기 안산지역에서 실시되었다. 천안 국립축산과학원 갈대 재배지에서는 N 무시비구, N 50 kg/ha구, N 100 kg구 등 3처리를 두고 4월 30일 시비, 6월 21일 수확하였으며, 안산 시화간척지 갈대 자생지에서는 N 무시비구, N 60 kg/ha구 등 2처리를 두고 5월 1일 시비, 8월 1일 수확하였다. 갈대의 초장과 엽색은 N 시비구에서, 그리고 N 시비수준이 높을 때 양호하였다. 건물률은 N 시비구에서 약간 낮아지는 경향으로 천안지역은 무시비구 39.8%, N 시비구 37.6%, 36.7%, 안산지역은 무시비구 39.4%, N 시비구 37.6%였다. 천안지역의 건물수량, 조단백질 수량 및 가소화 건물수량은 무시비구에서 ha당 각각 4,026 kg, 235 kg, 1,850 kg, N 50 kg 시비구는 각각 4,658 kg, 306 kg, 2,388 kg, N 100 kg 시비구는 각각 5,622 kg, 446 kg, 3,143 kg으로 유의적으로 증가하였으며 (p<0.05), 안산지역은 무시비구에서 각각 2,802 kg, 177 kg, 1,288 kg, N 60 kg 시비구는 각각 3,876 kg, 294 kg, 1,853 kg으로 유의적으로 증가하였다 (p<0.05). 천안지역의 조단백질 함량, 건물 소화율 및 상대사료가치 (RFV)는 무시비구에서 각각 5.85%, 45.96%, 64.0 (품질 5등급), N 50 kg 시비구는 각각 6.58%, 51.27%, 72.3 (5등급), N 100 kg 시비구는 각각 7.94%, 55.91%, 72.7 (5등급)로 높아졌으며, 안산지역도 무시비구에서 각각 6.30%, 45.98%, 70.2 (5등급), N 60 kg 시비구는 각각 7.59%, 47.80%, 78.3 (4등급)으로 높아지는 경향이었다. 이상의 결과를 종합하여 볼 때, 갈대 생육지는 시비가 가능한 조건이라면(하천변이나 강변 등 수질오염이 우려되는 지역이 아닌), ha당 N 60~100 kg을 생육기에 시용해 주는 것이 생산량과 사료가치를 크게 개선시켜줄 수 있어 바람직하였다. This study was carried out to investigate the effect of nitrogen (N) fertilization on the forage growth, production and quality of native reed (Phragmites communis) grasses. Field experiments were conducted in Cheonan and in Ansan, 2012. Treatments were control (no N fertilization), 50 kg/ha and 100 kg/ha in Cheonan plots (fertilization on April $30^{th}$, and harvest on June $21^{st}$). Treatments in Ansan plots were control (no N fertilization) and 60 kg/ha (fertilization on May first, and harvest on August first). Forage growth and leaf colors were improved in N fertilized plots. However, the drymatter (DM) percentage was slightly decreased with N fertilization. Forage yields, in terms of DM, crude protein (CP) and digestible DM (DDM), were significantly increased with N fertilization in both sites. In Cheonan, DM, CP and DDM yields per ha were 4,026 kg, 235 kg and 1,850 kg, respectively, in the control plot, and were 4,658 kg, 306 kg and 2,388 kg, respectively, in the N 50 kg plot, and 5,622 kg, 446 kg and 3,143 kg, respectively, in the N 100 kg plot. In Ansan, DM, CP and DDM yields per ha were 2,802 kg, 177 kg and 1,288 kg, respectively, in the control plot, and were 3,876 kg, 294 kg and 1,853 kg, respectively, in the N 60 kg plot. Forage quality in terms of CP content, in vitro DM digestibility (IVDMD) and relative feed value (RFV) were also increased with N fertilization in both sites. In Cheonan, the CP content, IVDMD and RFV were 5.85%, 45.96% and 64.5 (grade 5), respectively, in the control plot, 6.58%, 51.27% and 72.3 (grade 5), respectively, in the N 50 kg plot, and 7.94%, 55.91% and 72.7 (grade 5), respectively, in the N 100 kg plot. In Ansan, the CP content, IVDMD and RFV were 6.30%, 45.98% and 70.2 (grade 5), respectively, in the control plot, and 7.59%, 47.80% and 78.3 (grade 4), respectively, in the N 60 kg plot. In conclusion, N fertilization of 60~100 kg/ha was desirable for greater forage production, with a higher quality of native Phragmites communis achievable. This should only be applied if the fertilization area is not located at a riverside/streamside or in riparian land where there is a high risk of water pollution by fertilization.

        돈분뇨 액비 시용이 양파의 생육, 양분흡수량 및 토양 화학성에 미치는 영향

        벼와 양파 2모작 논토양에서 효율적인 돈분뇨 액비 시용을 위하여 액비 시용에 따른 양파의 생육, 양분흡수량 변화와 토양의 이화학적 특성을 조사하였다. 사용된 액비의 질소, 인산 및 칼륨 함량은 각각 4.2, 0.34, 1.2g kg⁻¹ 등이었다. 추비용 액비는 시용 10일 전에 물로 2배 희석하여 월 2회씩 4회와 6회로 나누어 시용하였다. 기비는 질소표준시비량 기준으로 19,280kg ha⁻¹로 시용하였으며 추비에 사용된 액비량은 101,910kg ha⁻¹이었다. 양파의 초장은 정식 136일 후부터 186일까지 급격히 증가하였으며 화학비료구, 액비기비+화학비료 추비구 및 액비기비+액비 4회 추비구 간에는 차이가 없었으나 무비구와 액비전량 기비구는 양분부족으로 생육이 저조하였다. 구 비대초기의 구경은 무비구에서 가장 컸으나 그 이후 화학비료구와 액비시용구에서 크게 증가하였다. 엽 생체중의 변화는 초장과 유사한 경향을 보인 반면 구 생체중은 구경과 비슷한 경향을 나타내었다. 초장과 엽중은 화학비료구와 액비전량 기비구 및 액비기비+액비 6회 추비구간 다소 작았으나 구경, 구중 및 엽수는 처리간 차이가 뚜렷하지 않았다. 양파 구와 잎의 양분흡수량은 전 생육기간에 시그모이드 형을 나타내었으며 잎과 구의 양분흡수량은 각각 잎과 구의 생육이 진전됨에 따라 증가하는 경향이었다. 화학비료구와 액비기비+화학비료 추비구 및 액비 4회 추비구간에 양분흡수량의 차이는 없었고 액비전량 기비구에서 잎의 질소와 칼륨흡수량이, 구의 질소흡수량이 화학비료구보다 적었다. 무비구는 질소, 칼륨 및 마그네슘 흡수량이 다른 처리구보다 적었다. 액비기비+액비 4회 추비구에서 질소와 인산이용률이 각각 23.5%, 13.2%로 가장 높았으나 액비기비+액비 4회 추비구에서 칼륨이용률이 높았고 상품 수량도 55.0Mg ha⁻¹로 가장 많았다. 구의 질소, 인산 및 칼륨함량은 생육이 진전됨에 감소하는 경향이었고 잎의 질소함량은 생육초기에 가장 많은 반면 칼륨함량은 구비대 초기에 가장 많았다. 수확 후 토양 pH, 질산태 질소, 교환성 칼륨 및 EC 농도는 화학비료구보다 액비시용구에서 약간 증가하였다. 본 연구 결과를 종합해 볼 때 돈분뇨 액비를 기비로 시용하고 3배 희석한 액비를 2월과 3월에 4회 추비하면 균형적인 양분공급이 가능하고 적정 수량도 확보할 수있었다. This study was carried out to evaluate the effect of liquid pig manure (LPM) on the nutrient uptake, yield of onion and soil properties. The LPM applied to onion field contained 4.2 g kg⁻¹ N, 0.34 g kg⁻¹ P, 1.2 g kg⁻¹ K and it was diluted with water by 1:2 for top-dressing, which was applied separately 4 or 6 times from February to April. The LPM application rate for basal fertilization was 19,280 kg ha⁻¹ which included 80 kg N, 6.6 kg P, and 22 kg K. The application rate for top-dressing was 101,910 kg ha⁻¹ which included 160 kg N, 13 kg P, and 53 kg K. The plant height was not almost different among treatments until 136 days after planting. Thereafter, it enlarged abruptly until 186 days, on May 4. The significant difference among CF and LPM + top-dressing by CF or LPM I was not shown, but no fertilizer and LPM only (it was applied totally as basal fertilization) made plant height of onion lessen at the maximum growth stage because of insufficient nutrients. The plant diameter increased after 136 days, and there was a significant difference between CF or LPM and no fertilizer from 186 days after planting. The leaf weight was similar tendency to plant height while bulb weight was similar to plant diameter. The amount of nutrient uptake in leaf coincided with leaf growth and the nutrient uptake in bulb swelled simultaneously with bulb growth. The difference between CF and LPM + top-dressing by CF or LPM I was not significant in nutrient uptake of onion leaf and bulb. But LPM only decreased in nitrogen and potassium of leaf and in nitrogen of bulb. And no fertilizer made a remarkable difference to others in nitrogen, potassium and magnesium of leaf and bulb. The utilization of nitrogen and phosphorus in LPM + top-dressing by CF were 23.5%, 13.2% respectively, more than the others. The LPM + top-dressing by LPM I was most available with potassium, and it leaded the most abundant marketable yield by 55.0 t ha⁻¹ as well. The nitrogen, phosphorus, and potassium contents in bulb were inclined to decrease with progress of onion growth. The nitrogen contents in leaf maximized at the initial growth stage while potassium contents increased at the initial bulbing stage. The LPM application increased pH, nitrate nitrogen, potassium and EC in soil more than chemical fertilizer (CF) did.

        Effect of Non-animal-Derived Nitrogen Sources on the Production of Hyaluronic Acid by Streptococcus sp. KL0188

        Hyaluronic acid (HA) is a linear high-molecular-weight polysaccharide with useful biomedical applications. Streptococcus zooepidemicus, a typical HA-producing bacterium, requires an animal-derived nitrogen source such as tryptone, peptone or sheep blood as a nutrient. Sixteen non-animal-derived (NAD) nitrogen sources were tested as a replacement for the expensive animal-derived nitrogen sources, which may have safety issues. Among the sixteen tested NAD nitrogen sources, a yeast-derived nitrogen source (YE 0251) showed the highest HA productivity, which was equivalent to the control HA production medium containing tryptone in a 5-L jar and in 3,000-L industrial fermentations. In the 3,000-L fermentation, YE 0251 increased cell mass (dry cell weight) and HA production by 11% and 8%, respectively, compared with the control HA production medium. The fmal specific volumetric productivity (0.41 g/L h) was improved by about 70% after reducing the fermentation time from 20 h to 12 h, compared to the conventional production medium.

        Nanohybrid magnetic liposome functionalized with hyaluronic acid for enhanced cellular uptake and near-infrared-triggered drug release

        <P><B>Abstract</B></P> <P>The aim of this work is to prepare and evaluate a novel lipid-polymer hybrid liposomal nanoplatform (hyaluronic acid-magnetic nanoparticle-liposomes, HA-MNP-LPs) as a vehicle for targeted delivery and triggered release of an anticancer drug (docetaxel, DTX) in human breast cancer cells. We first synthesize an amphiphilic hyaluronic acid hexadecylamine polymer (HA-C<SUB>16</SUB>) to enhance the targeting ability of the hybrid liposome. Next, HA-MNP-LPs are constructed to achieve an average size of 189.93±2.74nm in diameter. In addition, citric acid-coated magnetic nanoparticles (MNPs) are prepared and embedded in the aqueous cores while DTX is encapsulated in the hydrophobic bilayers of the liposomes. Experiments with coumarin 6 loaded hybrid liposomes (C6/HA-MNP-LPs) show that the hybrid liposomes have superior cellular uptake in comparison with the conventional non-targeting liposomes (C6/MNP-LPs), and the result is further confirmed by Prussian blue staining. Under near-infrared laser irradiation (NIR, 808nm), the HA-MNP-LPs aqueous solution can reach 46.7°C in 10min, and the hybrid liposomes released over 20% more drug than the non-irradiated liposomes. Using a combination of photothermal irradiation and chemotherapy, the DTX-loaded hybrid liposomes (DTX/HA-MNP-LPs) significantly enhance therapeutic efficacy, with the IC<SUB>50</SUB> value of 0.69±0.10μg/mL, which is much lower than the values for DTX monotherapy.         Survey of Cultivation and Status of Gerbera Smart Farms

        본 연구는 거베라 주산지인 경북과 경남 지역의 농가 재배 및 스마트팜 현황을 조사하고 분석하고자 수행하였다. 조사방법은 2020년 81 농가를 대상으로 대면 설문조사를 실시하였다. 거베라 경영주 연령은 50∼60대가 71.6%로 가장 많았고, 30∼ 40대가 16%, 70세 이상이 9.9%, 30세 이하는 2.5% 순이었다. 거베라 재배 경력은 5년 이하가 37%였으며, 6∼10년 25.9%, 11∼20년 29.6%, 21년 이상 농가가 7.4%였다. 재배 면적은 0.3 ∼0.6ha가 61.7%로 가장 많았고, 0.3ha 미만이 30.9%, 0.6ha 이상이 7.4%였으며, 전체 평균재배 면적은 0.37ha였다. 거베라 재배 농가당 평균 인력은 자가 노동력 2.1명, 고용 노동력 0.8명 이었다. 재배 방식에서 토경 88.9%, 양액재배 농가는 11.1%였다. 국산품종을 전체 또는 일부라도 재배하고 있는 농가는 61.7%였고 외국산 거베라 품종만 재배하고 있는 농가는 38.3% 였다. 거베라 병해충 발생조사에서 병 발생이 많았다고 응답한 농가는 91.4%였으며, 충 발생이 많았다고 응답한 농가는 98.8% 였다. 스마트팜 시스템을 설치한 농가는 12.3%였으며, 설치하지 않은 농가에서 향후 설치할 의향이 있다고 응답한 농가는 76.9% 였다. 향후 경영계획을 묻는 질문에서 현상유지 80.2%, 축소 8.6%, 경영확대 11.1%였으며, 경영 확대를 대답한 농가는 경북 지역이 8농가로 경남지역 1농가 보다 많았다. This study was conducted to investigate and analyze gerbera cultivation and the status of smart farms in Korea. The survey was conducted in person for 81 farms, in 2020. About 71.6% managers of the gerbera farms were in their 50s and 60s, 16% in their 30s and 40s, 9.9% were over 70, and 2.5% under 30. About 37% of the farmers have cultivation experience of less than five years; 25.9%, 6–10 years; 29.6%, 11–20 years; and 7.4%, over 21 years. Around 61.7% of the area under gerbera cultivation was 0.3–0.6 ha, 30.9% was less than 0.3 ha, and 7.4% was over 0.6 ha; and the average area under cultivation was 0.37 ha. The average family labor of the gerbera farms was 2.1 people and the employed labor was 0.8 people. The cultivation methods included about 88.9% soil and 11.1% nutrient cultivation farms. However, 61.7% of the farmers cultivated all or part of the domestic gerbera varieties, whereas 38.3% only cultivated foreign varieties. In the pest survey, around 91.4% of the farm managers responded to have observed different diseases, and around 98.8% farms exhibited various pests. The smart farm system has been installed by 12.3% of the farming households. However, 76.9% of farming households with no prior smart farms are planning to install them in the future. Future plans include the status quo of 80.2%, reduction in total area by 8.6% and expansion in total area by 11.1%, and around eight farms in Gyeongbuk and one farm in Gyeongnam area are planning to expand their management.

      • 人間의 c-Ha-ras發癌性 蛋白質에 結合되어 있는 GDP와 GTP의 形態 比較

        A truncated human c-Ha-ras gene product 〔ras(1-17)protein〕in which glutamine-61 was replaced by leucine 〔ras(L61/1-171)protein〕, was prepared and ligand was exchanged from GDP to GTPγS 〔Guanosine 5'(3-0-thio) triphosphate〕. A mass production of ras protein bound with GTPγS was possible using ultra-filtration with chealating agent. GTPγS bound to c-Ha-ras protein was clearly confirmed by ultra-violet spectroscopy and phosphorus nuclear magnetic resonance. The conformations of a bound GDP and a bound GTP to a ras(L61/1-171)protein were compared by time dependent nuclear Overhauser effect (NOE) spectroscopy. The dependences of the H1' and H8 proton resonance intensities on duration of time-dependent NOE profiles, and the conformation of the protein-bound GDP and GTPγS were elu??dated ; as for the bound GDP, the guanosine moiety takes the antiform about N-glycosidic bond with the dihedral angle of χ=-124±2° and the riose ring takes the C2'-endo form(Ha. 1990). Whereas the bound GTPγS, the guanosine moiety, takes the anti form about N-glycosidic bond with the dihedral angle of χ=-80±2°and the ribose ring takes the C3'-endo form The conformations around effector region are of the regular structure in the GDP-bound state, and largely distorted in the GTPγS-bound state, whichis probably related to the conformational activation of the effector region of ras proteins by ligand substitution of GDP with GTPγS. The role of the interaction between the bound guanine nucleotide and effector region is discussed with regard to the mechanism of the ras protein functions.

      • 人間의 c-Ha-ras 發癌性 蛋白質에 結合되어 있는 GDP의 形態

        ¹H-NMR spectra were analyzed of a GDP/GTP-binding domain of human c-Ha-ras gene product(residues 1-171) in which glutarmine-61 was replaced by leucine [ras(L61/1-171) protein]. By one-dimensional and two-dimensional homonuclear Hartmann-Hah spectroscopy and nuclear Overhauser effect (NOE) spectroscopy of the complex of the ras(L61/1-171) protein and GDP, the ribose H1', H2', H3' and H4' proton resonances of the bound GDP were identified. The guanine H8 proton resonance of the bound GDP was identified by substitution [8-²H]GDP for GDP. The dependences of the H1' and H8 proton resonance intensities in the duration of irradiation of the H1', H2', H3' and H8 protons were measured. By numerical simulation of these time-dependent NOE profiles, the conformation of the protein-bound GDP was elucidated ; the guanosine moiety takes the anti form about the N-glycosidic bond with the dihedral angle of χ=-124±2。 and the ribes ring takes the C2'-endo form. Such an analysis of the conformation of a guanine nucleotide as bound to a GTP-binding protein will be useful for further studies on the molecular mechanism of the conformational activation of ras proteins on ligand substitution of GDP with GTP.

      • 안정 동위체 표지 다차원 NMR 법에 의한 c-Ha-Ras 단백질의 구조해석

        In order to elucidate the structural change between the GDP-bound and GTP-bound forms of the Ras protein, isotope-aided 2D) and 3D NMR experiments were performed for uniformly ^(15)N-^(13)C labeled sample. First, most of the backbone resonances (N, NH, C_(α), CO) were assigned for the GDP-bound form by HNCO, HNCA and HCACO experiments. Next, HCCH-TOCSY and CT-HCCH-COSY experiments were performed for observation of side chain resonances of the Ras protein. For the GTP-bound form, the resonances due to G10-G13, I21, Y32-S39, and D57-Y64 were not identified clearly due to line broadening. Strength of the magnetic field strongly influenced the line shape of the resonances of residues near these regions of the GTP-bound form in ^(1)H-^(15)N HSQC spectrum, otherwise such an effect was not observed for the GDP-bound form. These results suggested that chemical exchange between multiple conformations occurred in the GTP-bound form.

        목적 : 메티실린 내성 황색 포도알균에 의한 연조직및 골 감염의 치료 시 glycopeptide약물의 정주 요법에 이은 경구 항생제로의 처방 전환이 요구되는 경우가 많으나 마땅한 경구 약제의 선택이 어려운 실정이다. 이에 경구 rifampin을 메티실린 내성 황색 포도알균에 의한 연조직 및 골 감염에 사용하여 치료한 경우를 분석하여 그 유용성 정도와 실패한 경우 실패의 위험인자를 알아보고자 하였다. 재료 및 방법 : 2001년 1월부터 2004년 9월까지 메티실린 내성 황색 포도알균에 의한 연조직 및 골 감염 환자를 대상으로 rifampin을 그 치료 목적으로 사용한 44명의 환자를 의무 기록과 X선 사진을 통해 후향적으로 분석하였다. 기초 자료를 분석하였고 감염이 재발하여 치료 실패로 판정된 환자들에 대해 치료 실패의 원인이 될 만한 주요 변수에 대한 Chi-square 검증과 로지스틱 회귀분석을 시행하였다. 결과 : 치료 후 재발은 20.5% (8/39)였으며 재발 예와 사망에 대한 위험요소에 대한 단변량 분석에서 고관절 보형물이 있는 경우 흔하였으나 통계적 차이에는 미치지 못하였다(P=0.065). 고관절 보형물 외에 동반된 질환들, 쇼크의 동반, 균혈증의 동반, 경구 시 ciprofloxacin과 rifampin의 병합 복용 기간 등을 혼란 변수로 사용한 다변량 로지스틱 회귀분석에서는 고관절 보형물 만이 통계적으로 유의하였다(P=0.044). 결론 : 메티실린 내성 황색 포도알균에 의한 연조직및 골 감염에 rifampin과 다른 약제를 병합하여 치료하는 경우 재발은 적었다. 치료 실패의 경우는 고관절 부위 감염과 같이 보형물을 쉽게 제거하기 어려운 상황과 관계되는 것을 보여준다. Background : Rifampin is sometimes used in combination with other anti-staphylococcal agents to treat methicillin-resistant Staphylococcus aureus (MRSA) infections but few reports are available about the usefulness of rifampin against MRSA infections, especially in soft tissue and bone infections. Thus, we evaluated the efficacy of rifampin as switch therapy for soft tissue and bone infections caused by MRSA and assessed the risk factors for treatment failure. Materials and Methods : We retrospectively studied 44 patients who had soft tissue and bone infections caused by MRSA and were treated with rifampin in combination or as monotherapy as switch therapy from January 2001 to September 2004. Results : The mean age of the subjects was 50.2 years and the number of male and female were 34 and 10, respectively. Median duration of rifampin use was 32 days and 25% of the patients had artificial prostheses. Thirty one patients (79.5%, 31/39) were cured with parenteral glycopeptide followed by rifampin in combination or as monotherapy. Eight patients (8/39) recurred after the completion of treatment. The presence of hip prosthesis was the only significant risk factor (P= 0.027) in multivariate logistic regression test. Rifampin was tolerable in all cases. Conclusion : Rifampin as switch therapy for soft tissue and bone infection caused by MRSA was effective in 79.5% (31/39) of sujects. The treatment failure seems to be associated with unremovable infected prosthesis.
