        금낭화(Dicentra spectabilis)의 배우체 형성 및 접합자배의 발달

        본 연구는 금낭화 배우체의 형성과 배 발달특성에 관한 기초 자료로 활용하기 위해 수행되었다. 시원세포로부터 발달된 소포자 모세포는 화뢰의 길이가 1 mm일 때 감수분열을 하여 4면체형(tetrahedral) 4분자가 형성되었다. 4개의 소포자는 분리되어 각각 웅성배우체로 발달하였다. 대포자 모세포는 화뢰의 길이 $4{\sim}5\;mm$ 수식 이미지에서 관찰되었다. 대포자의 발달유형은 정상형(polygonum)이었으며, 배낭의 형태는 굽어있는 곡생배주(amphitropous)였다. 3개의 뚜렷한 반족세포는 배낭이 성숙한 후에도 퇴화하지 않고 남아있었다. 개화 전 자웅배우체는 충분히 성숙하였다. 개화 시에 수술과 암술의 길이는 거의 비슷하거나 수술이 0.5 mm 짧아서 자화수정에 적당한 구조를 가지는 것으로 나타냈다. 수정 후 배는 구형, 심장형을 거쳐 자엽배까지 발달하였으며, 종자 산포시 초기 자엽배를 가지고 있음을 알 수 있었다. This study was carried out to obtain the basic informations on the characteristics of gametophytes formation and embryo developement in Dicentra spectabilis. Microspore mother cells developed from archesporial cells, start meiosis when flower bud length reaches around 1 mm, formed tetrahedral type tetrad. The 4 microspores were separated. They were developed to male gametophytes, respectively. Megaspore mother cells were observed when flower bud length was 4~5mm. The developemental type of megaspore was polygonum and embryo sac was amphitropous. Three large and distinctive antipodals did not degenerated and remained after embryo sac was developed. When the male and female gametophytes was fully developed, the length of stamen and style was very similar or stamen was shorter about 0.5 mm than that of style. This result indicates that self-fertilization can be occurred in this species. After fertilization, developing zygotic embryos showed various stages of development from globular to cotyledonary embryos, and zygotic embryo in seed scattering time seemed to have an early cotyledonary stage.

        현탁배양을 통한 금낭화(Dicentra spectabilis L. Lemaire)의 대량증식

        금낭화 (Dicentra spectabilis L.)는 개발가치가 있는 국내 자생화 중의 하나로서 우수한 품종의 대량생산이 절실히 요구되는 실정이다. 본 연구는 현탁배양을 통한 배발생세포의 대량증식과 체세포배 발생 및 식물체재생의 적정조건을 구명하여 효율적인 기내묘목의 대량생산 체계를 확립하고자 시도되었다. 1.0mg/L 2,4-D SH고체배지에서 유도된 배발생캘러스를 현탁배지에서 증식시킨 결과, 1.0mg/L 2,4-D를 포함한 SH배지에서 MS배지에 비해 증식율이 높게 나타났으며, 체세포배 발생율은 1/2배 SH기본배지에서 가장 높게 나타났다. 250$m\ell$ 삼각플라스크에서 5,000개 이상의 고빈도로 체세포배가 생산되었다. 체세포배로부터 발아유도시에 GA$_3$는 정상적인 식물체로의 재생을 억제하는 효과를 나타내었으며, 정상적인 식물체 재생은 1/2희석 SH배지에서 양호하였다, 신초와 뿌리를 갖는 유식물체를 모래와 원예용상토(5:1, vol)가 혼합된 토양이 함유된 포트에 이식하여 1개월 후 58%의 생존율을 나타내었다. 이상의 결과를 활용하면 체세포배로부터 금낭화의 묘목을 대량생산 할 수 있을 것으로 생각된다. Bleeding heart (Dicentra spectabilis L. Lemaire) is one of the most valuable wild flower in Korea. This work was conducted for the mass production of somatic embryos through suspension culture and more effective plant regeneration system in Dicentra spectabilis. High-frequency embryogenic callus proliferation was achieved in SH liquid medium supplemented with 1 mg/L 2,4-D. Half-strength SH medium was suitable concentration for somatic embryo induction and germination. About 5,000 embryos were produced per 250$m\ell$ flask after 4 weeks of culture. Germination rate of somatic embryos was decreased when GA$_3$ was added in medium. The plantlets showed a 58% survival rate when transferred to pots after 1 month of culture. The results indicate that micropropagation procedure via somatic embryogenesis can be applied for an efficient mass propagation of Dicentra spectabilis.

        Prognostic value of metabolic tumor volume on PET?/?CT in primary gastrointestinal diffuse large B cell lymphoma.

        <P>Primary gastrointestinal (PGI) diffuse large B cell lymphoma (DLBCL) is a relatively common disease. Recent studies indicate that measurement of maximum standardized uptake value (SUV(max)) on pretreatment for (18)F-fluorodeoxyglucose PET is an important prognostic factor in PGI DLBCL. However, there is still an association between initial tumor burden and prognosis. Thus, in the present study, we investigated whether tumor volume by PET could have a potential prognostic value to predict the outcome. From 2006 to 2009, 165 Stage I E/II E PGI DLBCL patients were enrolled in the study. One hundred and five patients received cyclophosphamide, doxorubicin, vincristine, prednisone, and rituximab (R-CHOP) only, whereas 60 patients underwent surgery plus R-CHOP. Metabolic tumor volume (MTV) was defined initial tumor burden as target GI lesion above SUV, 2.5 by PET as a contouring border. Over a median follow-up period of 36.6 months, receiver operating characteristic (ROC) analysis indicated that the best cut-off values for MTV and SUV(max) were 160.1 cm(3) and 12.0, respectively. The estimated area under the ROC curve was higher for MTV than SUV(max). Thus, MTV was a better predictor for survival than SUV(max). In patients with a low MTV (<160.1 cm(3)), there were no significant differences in survival between patients undergoing R-CHOP alone and surgery plus R-CHOP (P = 0.347 for progression-free survival [PFS]; P = 0.148 for overall survival [OS]). Conversely, in patients with a high MTV (>160.1 cm(3)), survival was longer in those who underwent surgery plus R-CHOP than in those treated with R-CHOP alone (P < 0.001 for PFS; P < 0.001 for OS). Multivariate analysis revealed that high MTV is an independent factor for predicting survival. Even in the era of rituximab, treatment of PGI DLBCL is not easy in patients with a high MTV.</P>

      • Prognostic value of Waldeyer's ring involvement of diffuse large B-cell lymphoma treated with R-CHOP.

        <P>The objective of present study was to compare the prognosis of diffuse large B-cell lymphoma (DLBCL) with Waldeyer's ring involvement (WR-DLBCL) to that of nodal DLBCL (N-DLBCL) without WR involvement. Between July 2004 and October 2009, medical records and metabolic tumor volume (MTV) measured by 2-[fluorine-18]-fluoro-2-deoxy-D-glucose ((18)F-FDG) positron emission tomography /computed tomography were collected from patients confirmed with DLBCL. All patients received six or eight cycles of R-CHOP therapy. One hundred and eight patients were included and categorized into WR-DLBCL and N-DLBCL groups. WR involvement and other prognostic factors were analyzed for survivals. High international prognostic index score and high MTV were independent poor prognostic factors. However, WR involvement was a good prognostic factor only in univariate analysis. This study suggests that tumor burden appeared to be more important than specific organ involvement for the prognosis of DLBCL.</P>

        Clinical significance of metabolic tumor volume by PET/CT in stages II and III of diffuse large B cell lymphoma without extranodal site involvement

        <P>The objective of this study was to investigate whether metabolic tumor volume (MTV) by positron emission tomography (PET) can be a potential prognostic tool when compared with Ann Arbor stage, in stages II and III nodal diffuse large B cell lymphoma (DLBCL). We evaluated 169 patients with nodal stages II and III DLBCL who underwent measurements with PET prior to rituximab combined with cyclophosphamide, adriamycin, vincristine, and prednisone (R-CHOP). Cutoff point of MTV was measured using the receiver operating characteristic (ROC) curve. During a median period of 36 months, stage II was 59.2% and III was 40.8%. Using the ROC curve, the MTV of 220 cm<SUP>3</SUP> was the cutoff value. The low MTV group (<220 cm<SUP>3</SUP>) had longer progression-free survival (PFS) and overall survival (OS), compared with the high MTV group (≥220 cm<SUP>3</SUP>) (<I>p</I> < 0.001, <I>p</I> < 0.001). Stage II patients had longer survival than those in stage III (PFS, <I>p</I> = 0.011; OS, <I>p</I> = 0.001). The high MTV group had lower PFS and OS patterns, regardless of stage, compared with the low MTV group (<I>p</I> < 0.001, <I>p</I> < 0.001). Multivariate analysis revealed an association of the high MTV group with lower PFS and OS (PFS, hazard ratio (HR) = 5.300, <I>p</I> < 0.001; OS, HR = 7.009, <I>p</I> < 0.001), but not stage III (PFS, <I>p</I> = 0.187; OS, <I>p</I> = 0.054). Assessment of MTV by PET had more potential predictive power than Ann Arbor stage in the patients that received R-CHOP.</P>

        가시없는 블랙베리(Rubus fruticosus L. cv. BB21)의 근맹아를 이용한 다경유도와 절간배양을 통한 식물체 증식조건의 확립

        본 연구에 사용한 식물체는 Rubus fruticosus L. cv. BB21은 Rubus fruticosus L. ${\times}$ R. parvifolius L. 교잡 블랙베리 V3품종에서 방사선을 처리하여 돌연변이로 선발 육종된 V9개체 중 다시 방사선을 처리하여 선발육종 된 품종이다. 이러한 우량계통의 신속한 증식을 이루기 위하여 조직 배양을 통한 효과적인 방법을 구명하고자 배지 및 생장조절물질 등의 처리에 따른 안정적인 생산을 위해 본 연구가 수행되었다. 땅속에서 채집된 근맹아 절편체를 0.1, 0.5, 1.0, 2.0 mg/L BAP가 첨가된 MS와 1/2MS 배지에 치상하여 6주간 배양한 후, 다경의 형성률을 조사하였다. 형성된 다경으로부터 산업적으로 이용하기 위한 안정된 생산을 위하여, 신초의 잎을 제거한 후 절간배양을 수행하여 다수의 소식물체를 증식하고자 여러종류의 배지 즉, AD (Anderson 1980), B5 (Gamborg 1968), MS (Murashige and Skoog 1962), SH (Schenk and Hildebrandt 1972), QL (Quoirin and Lepoivre 1977), WPM (Lloyd and McCown 1981)배지와 상기의 각각의 동일배지의 무기염류의 수준을 절반으로 한 1/2배지에 배양하였다. 초기의 생육(30일간 배양 시)에 있어서 생장은 배지 간에 차이를 나타내어 1/2AD, 1/2QL 배지에서 가장 양호한 초장을 나타내었으며, 절간수도 약 3.8, 4.4개를 나타내었으며, 뿌리, 생중량 다경유도 모두 양호하였다. 90일간 배양한 경우에서는 MS배지가 초장이 가장 양호하였으며 AD, SH 순으로 양호한 결과를 나타내었다. 식물체의 토양 순화는 신초와 뿌리가 잘 발달된 정상적인 식물체를 모래, 토양, 그리고 버뮤큘라이트가 혼합된 토양(1:1:1, v/v)에 각각 직접 이식하여 실시한 결과, 식물체의 발근율과 생존률(약98%)을 나타내었다. 이상의 결과를 통하여 블랙베리 묘목의 대량생산이 가능하여 농가에 원활한 공급이 가능할 것으로 예상된다. This study was conducted to develop a simple, rapid, and reliable method for in vitro propagation of disease-free and true-to-type clones from sucker explants of thornless blackberry (Rubus fruticosus L. ${\times}$ R. parvifolius L.). To induce multiple shoots, the sucker explants were sterilized in 1% NaOCl solution, and then were aseptically cultured on the full and 1/2 MS solid medium supplemented with BAP (0.1, 0.5, 1.0, 2.0 mg/L). After six weeks of culture, the highest frequency (85.4%) of shoot formation from sucker explants was obtained on the full-strength MS medium with 1.0 mg/L BAP. Node explants obtained from multiple shoots were cultured on the various media of full- or half-strength of AD, B5, MS, SH, QL, WPM media, respectively. After 30 days of culture, plant growth was good on the half-AD, half-QL medium. After 90 days of culture, plant growth was good on the full MS and full SH medium. The survival rate of the plantlets after transfer to plastic pots containing soil mixture (sand: soil: vermiculite was 1:1:1, vol.) in the greenhouse was 98%. The results indicate that a multiple-shoot procedure can be applied for an efficient mass propagation of Rubus fruticosus L. ${\times}$ R. parvifolius L.

        오가피(Acanthopanax sessiliflorus)의 체세포배로부터 식물체 재생에 미치는 GAa<sub>3</sub>와 Charcoal의 영향

        To establish the optimum condition for plant regeneration from somatic embryos of Acanthopanax sessiliflorus Rupr. et Maxim, a medicinal plant, somatic embryos were induced from zygotic embryo-derived embryogenic callus in hormoen-free MS medium. To induce plantlet conversion, cotyledonary somatic embryos were cultured on MS solid medium with GA$_3$at various concentrations (0~10 mg/L) for three weeks. Plantlets were transferred to 1/3 MS solid medium with 0.5% charcoal for 7 weeks. Stem length was increased proportionally to the concentration and treatment period of GA$_3$. Also, the highest leaf width (8.9 mm) and leaf number (2.84) of plantlet were obtained when plantlets were converted on 5,10 mg/L GA$_3$pretreatments, respectively. The highest plant conversion frequency (66.7%) was obtained when the somatic embryos were cultured on medium containing 5 mg/L GA$_3$ for 3 weeks and then were transferred to 1/3 MS medium with 0.5% charcoal. The highest survival rate of soil transfer was 90% when plantlets were regenerated on medium with 5 mg/L GA$_3$ for 3 weeks and then transferred to plastic pots containing vermiculite and sand mixture for 4 weeks.

        블랙베리(Rubus fruticosus L.)의 맹아절편체로부터 다경유도를 통한 식물체 재분화

        블랙베리의 다경유도를 통한 식물체 재생을 위하여, 노지에서 생육되고 있는 맹아를 채취하여 1.2% NaOCl 용액에 침지하여 표면 살균한 후, kinetin과 BA가 각각 포함된 MS 배지에서 4주간 배양하여 절편당 형성된 신초의 수를 조사하여 BA가 효과적임을 밝혔다. 동일방식으로 맹아를 IBA와 BA를 첨가한 MS 고체배지에서 4주간 배양하여 신초를 유도하였는데 1.0 mg/L BA가 첨가된 배지에서 신초유도는 100% 이루어졌다. 기내배양한 신초의 절편체를 1.0 mg/L BA가 첨가된 MS 배지에 치상하여 형성된 신초의 수는 절편체당 5.3개로 최고를 나타내었다. 약 10주 정도 배양을 더 오래하였을 때, 다경 형성율이 더 높아질 뿐 아니라 절편체당 신초 수도 많아졌다. 형성된 신초로부터 뿌리 발생을 위하여 glycine의 농도를 달리한 (0~2.0mg/L) MS 기본배지에서 4주간 배양하였을 때, 0.5 mg/L glycine을 첨가한 배지에서 고빈도 (85%의 뿌리발생을 관찰 할 수 있었다. 재생된 식물체는 모래 : 토양 : 버뮤큘라이트 (1:1:1, vol.) 혼합토양에서 순화시켜, 토양에 이식하였을 때 95%의 식물체 생존율을 나타내었다. This study was carried out to induce plant regeneration via shoot formation from sucker explants of Rubus fruticosus L. To induce adventitious shoots, sucker explants were sterilized in 1.2% NaOCl solution, and cultured on the MS solid medium supplemented with kinetin (0.5, 1.0, 3.0 mg/L) and BA (0.5, 1.0, 3.0 mg/L), respectively. As above, to induce adventitious shoots, sucker explants were cultured on the MS solid medium supplemented with IBA (0, 0.1, 1.0 mg/L) and BA (0, 0.1, 1.0, 2.0 mg/L). After 4 weeks of culture, the highest frquency (100% of shoot formation from sucker explants was obtained from the medium with 1.0 mg/L BA. The highest shoot number per explant from in vitro shoot explants was 5.3. After 10 weeks of culture, the number of shoot per explant was increased. The highest frequency (85% of root formation was obtained at 0.5 mg/L glycine medium, when the explant with shoot were cultured on the MS medium containing glycine at various concentrations from 0 to 2.0 mg/L. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1:1:1, vol.) was 95%. The results indicate that micropropagation procedure can be applied for an efficient mass propagation of Rubus fruticosus.

        임신 제 1삼분기에 진단된 재발성 멕켈 그루버 증후군

        Meckel Gruber syndrome consisting of an occipital encephalocele, polycystic kidney disease and polydactyly is a rare autosomal recessive disorder with a recurrence risk of 25%. Targeted ultrasonography in late embryonic or early fetal stages of pregnancy has great importance in diagnosis and management of affected pregnancy in high risk groups due to incomplete genetic mapping of meckel syndrome gene (MKS). We present a case of prenatal diagnosis at 14 weeks` gestational age of Meckel Gruber syndrome in a woman, who experienced same disorder in her previous pregnancy.
