Objective Vaginal atrophy, the second most common complication of menopause, can lead to sexual dysfunction. This study evaluated the effect of a vitamin D vaginal suppository on sexual functioning in postmenopausal women. Methods This three-arm randomized controlled trial was conducted between August 2019 and August 2020. The sample comprised 105 postmenopausal women who were referred to comprehensive health service centers to receive postmenopausal care. The inclusion criteria were as follows: (i) being menopausal for at least 1 year, (ii) being married, (iii) being sexually active, and (iv) having sexual desire. Participants were randomly assigned to three groups for 8 weeks of treatment: intervention (vaginal suppository containing 1,000 units of vitamin D3), placebo (vaginal suppository placebo), or control (no treatment). The main outcome measure was sexual functioning, which was assessed using the Female Sexual Function Scale (FSFI) 4 times during the study (i.e., 1 month before the intervention, immediately after the intervention, 1 month after the intervention, and 2 months after the intervention). Results Immediately and 1 month after the trial, the intervention group had the highest FSFI score, followed by the placebo group, both of which were significantly higher than those of the control group (P<0.05). At the 2-month follow-up, the intervention and placebo groups had similar FSFI scores (P=0.08), both of which were significantly higher than those in the control group (P=0.001 and P=0.03, respectively). Conclusion Vitamin D vaginal suppositories were more effective at improving sexual functioning among postmenopausal women in the short-term and appeared to prevent aging-related sexual functioning decline in the long term.

Background: In recent decades, consumption of simple sugars has increased dramatically, which contributes to health problems including insulin resistance and obesity. In this study, we investigated the effects of high concentrations of white sugar (WS) and brown sugar (BS) on serum concentration of brain-derived neurotrophic factor (BDNF), insulin resistance, and body weight in albino rats. Methods: Twenty-four male Wistar rats were randomly divided into three groups: control, a group treated with 15% WS, and a group treated with 15% BS. Rats were given WS and BS by gavage (daily) for 42 days. At the end of the intervention period, serum level of BDNF, insulin resistance, and body weight were measured. Results: Body weight and insulin resistance were significantly increased and serum BDNF level was decreased in both WS and BS groups compared to the control group (P<0.05). In the WS-treated rats, the amount of changes in the insulin resistance, body weight, and serum BDNF level was greater compared to that in BS-treated (P< 0.05). Conclusion: Due to the adverse effects of consuming high levels of WS and BS on serum level of BDNF, insulin resistance, and body weight, high intakes of these sweeteners are not recommended.

Fructooligosaccharides have been mainly produced by microbial fructosyltransferases (FTase) enzymes. The present work focuses on the optimization of medium composition and cultivation parameters affecting FTase produced by Penicillium aurantiogriseum AUMC 5605 in shake flask cultivation. FTase production was optimized in two steps using DeMeo’s fractional factorial design. A 1.46-fold increase in FTase production (105.4 U/mL) was achieved using the optimized culture medium consisting of (g/L): sucrose, 600; yeast extract, 10; K2HPO4, 5; MgSO4·7H2O, 0.5; (NH4) 2SO4, 1.0 and KCl, 0.5. The obtained results showed that the maximum FTase enzyme activity was produced at initial cultivation pH values ranging from 6.0-6.5, at agitation speed of 200 rpm and using vegetative fungal cells as inoculum. Moreover, results showed that optimization of medium composition and some cultivation parameters resulted in an increase of about 93.7% in the enzyme activity than the nonoptimized cultivation conditions after 96 h of cultivation. Additionally, maximum production and specific production rates recorded 2340 U/L/h and 102 U/L/h/g cells, respectively.
