        MiR-374b Promotes Proliferation and Inhibits Apoptosis of Human GIST Cells by Inhibiting PTEN through Activation of the PI3K/Akt Pathway

        Gastrointestinal stromal tumours (GIST) are the most common mesenchymal tumors of the gastrointestinal (GI) tract. In order to investigate a new treatment fot GIST, we hypothesized the effect of miR-374b targeting PTEN gene-mediated PI3K/Akt signal transduction pathway on proliferation and apoptosis of human gastrointestinal stromal tumor (GIST) cells. We obtained GIST tissues and adjacent normal tissues from 143 patients with GIST to measure the levels of miR-374b, PTEN, PI3K, Akt, caspase9, Bax, MMP2, MMP9, ki67, PCNA, P53 and cyclinD1. Finally, cell viability, cell cycle and apoptosis were detected. According to the KFGG analysis of DEGs, PTEN was involved in a variety of signaling pathways and miRs were associated with cancer development. The results showed that MiR-374b was highly expressed, while PTEN was downregulated in the GIST tissues. The levels of miR-374b, PI3K, AKT and PTEN were related to tumor diameter and pathological stage. Additionally, miR-374b increased the mRNA and protein levels of PI3K, Akt, MMP2, MMP9, P53 and cyclinD1, suggesting that miR-374b activates PI3K/Akt signaling pathway in GIST-T1 cells. Moreover, MiR-374b promoted cell viability, migration, invasion, and cell cycle entry, and inhibited apoptosis in GIST cells. Taken together, the results indicated that miR-374b pro-motes viability and inhibits apoptosis of human GIST cells by targeting PTEN gene through the PI3K/Akt signaling pathway. Thus, this study provides a new potential target for GIST treatment.

        논문 : 한국어 원어민 화자의 표현 형태 양상 조사 연구

        This study aims to survey and analysis the expressional forms of Korean native speakers so that it can be used as the base of Korean language study material. Even if a Korean language learner masters the exact grammatical forms, the learner would have trouble in real communication without understanding actual functions of the forms. To learn both grammatical form itself and the actual functions of the form, it is required to construct the native speaker baseline data as well as Korean grammatical knowledge. Therefore in this study, the actual usage of 83 korean native speakers and application methods of that for Korean language education are researched. Also, with the data collecting method of DCT(Discourse Completion Test), 14 conditions are set up, and the expressional forms of each condition whose the expressional purpose is fixed are researched. For Korean language education, it is needed to develope the material on learning the language form according to expressional intention. This survey of the expressional forms of Korean native speakers can be an meaningful attempt to attain this goal. (Central University of Nationalities, China)

        한국어 어휘의 의미와 그 사용실태

        본 연구는 한국의 신문, 방송, 텔레비전, 광고 등 주로 일상에서 쓰이는 언어를 논의의 대상으로 그의 어휘적 의미를 살펴보고 현재의 어휘 사용 실태를 몇 가지 유형으로 나누어 고찰 분석하였다. 이를 통해 아름다운 한국어를 보호하고, 한국어 사용을 진작해야 할 중요성을 호소하고, 그 책임감을 고양하고자 하였다. 이런 목적 아래, 언론매체 뿐만이 아니라 한국어 보호와 사용 및 홍보에 앞장서야 할 한국 정부의 기관이 오히려 외국어를 과도하게 사용하고 있다는 점을 지적하였다. 그리고 친족어의 확대 사용과 경어의 사용 실태를 고찰하면서 사회적으로 비록 널리 사용되고 있지만 우려할 만큼 비문법적인 경향으로 흘러가고 있으며, 빠른 속도로 일상용 언어로 자리 잡고 있는 문제점에 대해 심각한 우려를 표했다. 따라서 신조어의 남용과 어휘의 과다 생략 사용실태를 통해 외국어를 그대로 받아들이거나 차용하여 만들어낸 언어 그리고 한국어 어휘의 생성 규칙에 위배되거나 한국어 어휘의 특성을 지니지 않은 언어들을 무절제하게 만들어 내어 사용하는 현상에 대해서는 마땅한 주시와 관찰이 요구된다는 점을 지적하였다. 한국 정부가 한국어의 순화작업을 꾸준히 진행해 왔는데도 불구하고 최근에 와서 영어를 위주로 하는 외국어들이 한국 고유어의 자리를 빼앗고 있는 점에 대해 우려를 표했으며 한국의 것보다는 해외의 것들을 더 격식 높고 수준 높은 것으로 보고 한국어를 사용하는 것보다 외국어를 사용하는 것을 더 박식한 것으로 느끼는 사회 풍토를 비판하였다. 따라서 바야흐로 세계화의 길에 들어서는 현시대에 한국정부와 산하의 많은 관련기구들 그리고 한국국민 모두가 한국어를 사랑하고 지키고 홍보하는 선두에서 최선을 다해야 한다고 주장하였다. Korea has a long history as a nation have a beautiful language and culture. Korean in the world`s population are within the top 10 with a language and is the world`s most beautiful language. These excellent Korean and Korea`s own culture and tradition of the beautiful features need to be held well. In the agricultural and food, Koreans very much emphasize "local purchasing (身土不二)". But otherwise nothing seems to be in use Korean. The language of their country than the other countries prefer the language of everyday conversation, and more Korean Foreign Origin in the more fun to use. They use easy and clear Korean incorrectly sometimes. Accordingly, the original appearance of Korean is increasingly becoming. In this paper, we will analyze the Korean language which the meaning is becoming gradually or is misused. The purpose is to protect the Korean language, promote the importance of using Korean, and enhance the accountability.

      • Three-Port Laparoscopic Exploration is not Sufficient for Patients with T4 Gastric Cancer

        Gastric cancer continues to be a leading cause of cancer death. The majority of patients with gastric adenocarcinoma in China present with advanced disease. Ruling out unresectable cancers from an unnecessary "open" exploration is very important. The aim of this study was to assess the value of five-port anatomical laparoscopic exploration in T4 gastric cancer in comparison with three-port laparoscopic exploration and laparotomy exploration. We conducted a retrospective study on 126 patients with T4 stage scheduled for D2 curative gastrectomy based on computed tomography (CT) staging at Department of Gastric Cancer and Soft Tissue Sarcoma, Fudan University Shanghai Cancer Center, from Apr. 2011 to Apr. 2013. Laparotomy exploration (Group I), three-port laparoscopic exploration (Group II) or five-port anatomical laparoscopic exploration (Group III) were performed prior to radical gastrectomy. Accuracy rate for feasibility of D2 curative gastrectomy in laparotomy exploration and five-port anatomical laparoscopic exploration groups was higher than that in the three-port laparoscopic exploration group. Five-port anatomical laparoscopic exploration group had the highest accuracy resection rate (Group I vs Group II vs Group III,92.6% vs78.6% vs 97.7%; p<0.05) and shorter length of hospitalization (Group I vs Group II vs Group III, $9.58{\pm}4.17$ vs $6.13{\pm}2.85$ vs $5.00{\pm}1.81$; p<0.001). Three-port laparoscopic exploration has low accuracy rate for assessing feasibility of D2 curative gastrectomy and five-port anatomical laparoscopic exploration should be performed on patients with T4 gastric cancer.

        Sliding mode control based on neural network for the vibration reduction of flexible structures

        A discrete sliding mode control (SMC) method based on hybrid model of neural network and nominal model is proposed to reduce the vibration of flexible structures, which is a robust active controller developed by using a sliding manifold approach. Since the thick boundary layer will reduce the virtue of SMC, the multilayer feed-forward neural network is adopted to model the uncertainty part. The neural network is trained by Levenberg-Marquardt backpropagation. The design objective of the sliding mode surface is based on the quadratic optimal cost function. In course of running, the input signal of SMC come from the hybrid model of the nominal model and the neural network. The simulation shows that the proposed control scheme is very effective for large uncertainty systems.

        Fabrication and Properties of Mo2FeB2 Cermet Coating on Stainless Steel Surface via One-Step Forming Technology of Powder Metallurgy

        Mo2 FeB2 cermet coating and stainless steel substrate were integrally fabricated by one-step forming technology of powder metallurgy. The phase and microstructure of the coating and interface were analysed through X-ray diffraction and scanning electron microscopy. Microhardness, bending strength, and wear resistance were also measured by Vickers tester, the electronic universal testing machine and wear tester. The microstructure of Mo2 FeB2 cermet coating showed Mo2 FeB2 hard phase and Fe binder phase, which was uniform and dense. A compacted interface without pores or defects was formed between the cermet coating and stainless steel substrate. The formation of eutectic liquid phase and applied pressure in sintering process promoted densification of the coating. Mo2 FeB2 cermet coating presented a microhardness of 1275 HV200 , which was obviously higher than that of 316L stainless steel substrate. The results of three-point bending test and wear test showed that Mo2 FeB2 cermet coating on steel substrate maintained good metallurgical interface bonding with the substrate, and the coating exhibited high bending strength and excellent wear resistance.

        A unique genetic lineage at the southern coast of China in the agar-producing Gracilaria vermiculophylla(Gracilariales, Florideophyceae)

        Ocean warming can have significant negative impacts on population genetic diversity, local endemism and geographical distribution of a wide range of marine organisms. Thus, the identification of conservation units with high risk of extinction becomes an imperative task to assess, monitor, and manage marine biodiversity for policy-makers. Here, we surveyed population structure and genetic variation of the red seaweed Gracilaria vermiculophylla along the coast of China using genome-based amplified fragment length polymorphism (AFLP) scanning. Regardless of analysis methods used, AFLP consistently revealed a south to north genetic isolation. Populations at the southern coast of China showed unique genetic variation and much greater allelic richness, heterozygosity, and average genetic diversity than the northern. In particular, we identified a geographical barrier that may hinder genetic exchange between the two lineages. Consequently, the characterized genetic lineage at the southern coast of China likely resulted from the interplay of post-glacial persistence of ancestral diversity, geographical isolation and local adaptation. In particular, the southern populations are indispensable components to explore evolutionary genetics and historical biogeography of G. vermiculophylla in the northwestern Pacific, and the unique diversity also has important conservation value in terms of projected climate warming.

        Prediction and analysis of structural noise of a box girder using hybrid FE-SEA method

        With the rapid development of rail transit, rail transit noise needs to be paid more and more attention. In order to accurately and effectively analyze the characteristics of low-frequency noise, a prediction model of vibration of box girder was established based on the hybrid FE-SEA method. When the train speed is 140 km/h, 200 km/h and 250 km/h, the vibration and noise of the box girder induced by the vertical wheel-rail interaction in the frequency range of 20-500 Hz are analyzed. Detailed analysis of the energy level, sound pressure contribution, modal analysis and vibration loss power of each slab at the operating speed of 140 km /h. The results show that: (1) When the train runs at a speed of 140km/h, the roof contributes more to the sound pressure at the far sound field point. Analyzing the frequency range from 20 to 500 Hz: The top plate plays a very important role in controlling sound pressure, contributing up to 70% of the sound pressure at peak frequencies. (2) When the train is traveling at various speeds, the maximum amplitude of structural vibration and noise generated by the viaduct occurs at 50 Hz. The vibration acceleration of the box beam at the far field point and near field point is mainly concentrated in the frequency range of 31.5-100 Hz, which is consistent with the dominant frequency band of wheel-rail force. Therefore, the main frequency of reducing the vibration and noise of the box beam is 31.5-100 Hz. (3) The vibration energy level and sound pressure level of the box bridge at different speeds are basically the same. The laws of vibration energy and sound pressure follow the rules below: web <wing plate <top plate. (4) When the train is running at a higher speed, the noise and vibration of the bridge structure are larger. (5) The hybrid FE-SEA method is used to predict the structural noise of the box beam, which not only improves the efficiency, but also improves the calculation accuracy, thereby expanding the frequency range of the structural noise and improving the prediction accuracy.
