      • Growth, Clonability, and Radiation Resistance of Esophageal Carcinoma-derived Stem-like Cells

        Objective: To separate/enrich tumor stem-like cells from the human esophageal carcinoma cell line OE-19 by using serum-free suspension culture and to identify their biological characteristics and radiation resistance. Methods: OE-19 cells were cultivated using adherent and suspension culture methods. The tumor stem-like phenotype of CD44 expression was detected using flow cytometry. We examined growth characteristics, cloning capacity in soft agar, and radiation resistance of 2 groups of cells. Results: Suspended cells in serum-free medium formed spheres that were enriched for CD44 expression. CD44 was expressed in 62.5% of suspended cells, but only in 11.7% of adherent cells. The suspended cells had greater capacity for proliferation and colony formation in soft agar than the adherent cells. When the suspended and adherent cells were irradiated at 5 Gy, 10 Gy, or 15 Gy, the proportion of CD44+ suspended cells strongly and weakly positive for CD44 was 77.8%, 66.5%, 57.5%; and 21.7%, 31.6%, 41.4%, respectively. In contrast, the proportion of CD44+ adherent cells strongly positive for CD44 was 18.9%, 14.%, and 9.95%, respectively. When the irradiation dose was increased to 30 Gy, the survival of the suspended and adherent cells was significantly reduced, and viable CD44+ cells were not detected. Conclusion: Suspended cell spheres generated from OE-19 esophageal carcinoma cells in serum-free stem medium are enriched in tumor stem-like cells. CD44 may be a marker for these cells.

      • Saliva Supernatant miR-21: a Novel Potential Biomarker for Esophageal Cancer Detection

        Objective: To identify whether saliva supernatant miR-21 can serve as a novel potential biomarker in patients with esophageal cancer (EC). Methods: 32 patients with EC and 16 healthy controls were recruited in this study. Total RNA was extracted from saliva supernatant samples for measurement of miR-21 levels using RT-qPCR and relationships between miR-21 levels and clinical characteristics of EC patients were analyzed. Results: miR-21 was significantly higher in the EC than control groups. The sensitivity and specificity were 84.4% and 62.5% respectively. Supernatant miR-21 levels showed no significant correlation with cancer stage, differentiation and nodal metastasis. Conclusions: Saliva supernatant miR-21 may be a novel biomarker for EC.

        한ㆍ중 선택 관계 접속문 대조 연구

        이 연구는 한국어에서 선택 관계를 나타내는 '-거나', '-든지', '-나' 구문과 이에 대응하는 중국어 '不是…就是…', '或者…或者…', '无论/不管…还是…' 구문의 통사ㆍ의미적 특징과 제약 현상을 비교한다. 한국어와 중국어의 선택 관계 접속문에 대한 비교ㆍ대조는 한국어 교육과정 개발뿐만 아니라 중국인 한국어 학습자가 양국 언어의 차이점을 인지하여 한국어 문장 확대 능력 및 한국어 쓰기 능력을 향상하는 데에 도움이 될 것이다. 한국어 '-거나'와 중국어의 '不是…就是…'의 의미는 선ㆍ후행절의 두 명제 가운데 한 명제는 실현될 수 있음을 선택적으로 나타내는 '지정 선택'이다. 한국어 '-든지'와 중국어의 '或者…或者…'의 의미는 문장 안의 명제 중 하나를 선택하거나 이 외의 것을 선택하는 것을 나타내는 '개방 선택'이다. 한국어 '-나'와 중국어의 '无论/不管…还是…'의 의미는 임의적이고 소극적인 개방 선택이다. Conjunctive sentences in Korean and Chinese have a lot of similar and different features in grammatical and semantic aspects. The understanding about these aspects is important to better Korean grammar and writing skills to Chinese learners of Korean language. This study is a contrastive study on conjunctive sentences in Korean and Chinese, which is focusing on Selection relation conjunctive sentences. This study compares with syntactic and semantic features and constraints of '-거나'/'-든지'/'-나' conjunctive sentences in Korean and '不是…就是…'/'或者…或者…'/'无论/不管…还是…' sentences in Chinese. First, '-거나' sentences and '不是…就是…' sentences express definiteness selection. Second, '-든지' sentences and '或者…或者…' sentences express open selection. The third, '-나' sentences and '无论/不管…还是…' sentences express passive open selection.

        종업원의 매력도 및 관계특성이 고객의 동반이탈의도에 미치는 영향

        본 연구는 종업원의 매력도(호감성, 전문성, 유사성), 대체 종업원의 매력도, 그리고 기존 종업원과의 관계특성(관계투자, 관계혜택, 관계단절비용)이 직접적으로 또는 관계의 질(만족, 신뢰, 애호도)을 통해 간접적으로 고객의 동반이탈의도에 미치는 영향을 검증하였다. 미용실을 이용하는 고객 228명으로부터 자료를 수집하여 분석한 결과, 종업원의 매력도가 관계의 질에는 영향을 미치나 고객의 동반이탈의도에는 직접적인 영향을 미치지 않는 것으로 나타났다. 즉, 종업원의 매력도는 관계의 질을 향상시키는 중요한 요인이지만 고객의 동반이탈의도에는 직접적으로 영향을 미치지 않고 관계의 질을 통하여 간접적으로 영향을 미치는 것으로 나타났다. 또한 대체종업원의 매력도는 고객의 동반이탈의도에 직접적으로 부정적인 영향을 미치는 것으로 나타났다. 관계특성은 종업원과의 관계의 질 및 고객의 동반이탈의도에 유의한 영향을 미치고, 고객애호도는 고객의 동반이탈의도에 유의한 영향을 미치는 것으로 나타났다.

        Selection and validation of reference genes for RT-qPCR analysis in Sf9 cell line infected by Heliothis virescens ascovirus 3h (HvAV-3h)

        The real-time quantitative reverse transcription PCR (qRT-PCR) is an efficient and pervasive technique for analysis gene expression. Despite the fact that this technique has been extensively used to explore the gene functions, the stability of the reference genes still requires being validated at different experimental conditions. This research aims to confirm that the expression stability of seven candidates reference genes under different concentrations sterile hemolymph containing virus (2, 4, 6, 108-fold) infects the Spodoptera frugiperda cells line (Sf9). The appropriate reference genes as endogenous controls were determined through four computational algorithms (ΔCt, NormFinder, BestKeeper, and geNorm) and an integrated online software RefFinder. According to results of this study, TUB and ACT1 were proved to be the most stable reference genes in E2 fold and E4 fold respectively. PPI was the best reference gene in E6 and E8 fold, 28S and TUB were the most stable pair reference genes through all samples. These results can facilitate basic researches of the molecular mechanism of HvAV- 3h and also prove to be helpful for the standard RT-PCR method.

        Lactation performance and rumen fermentation in dairy cows fed a diet with alfalfa hay replaced by corn stover and supplemented with molasses

        Objective: The objective of current study was to investigate the lactation performance and rumen fermentation characteristics of dairy cows fed a diet with alfalfa hay replaced by corn stover but supplemented with molasses. Methods: Sixteen Holstein cows in mid-lactation were randomly assigned to 1 of 2 dietary treatments: i) alfalfa based diet (AH), and ii) corn stover based diet supplemented with molasses (CSM). The experiment was conducted according to a 2×2 crossover design with 22-d each period, consisting of 17 d for adaptation and 5 d for data and samples collection. Results: Dry matter intake and milk yield were higher for cows fed AH than CSM (p<0.01). Milk protein content and nitrogen conversion were higher (p<0.05), while milk urea nitrogen was lower (p<0.01) for cows fed AH than CSM-fed cows. Contents of milk total solids, fat and lactose were not different between two groups (p>0.10). Total rumen volatile fatty acid concentration tended to be higher (p = 0.06) for cows fed AH than CSM-fed cows. Molar proportion of acetate was lower (p = 0.04), but valerate was higher (p = 0.02) in cows fed AH than CSM-fed cows. Rumen concentration of propionate, and isobutyrate, and ratio of acetate to propionate tended to be different (p<0.10) between two groups. The feed cost per kilogram of milk was lower in CSM than AH (p<0.01). No differences were found in feed efficiency and most plasma parameters tested (p>0.10). Conclusion: In comparison with AH diet, CSM diet could be fed to dairy cows without negative effect on feed efficiency, ruminal fermentation, but economically beneficial, indicating that CSM could be an alternative choice for dairy farms instead of AH to feed mid-lactation dairy cows.

        Construction and Characterization of Vitreoscilla Hemoglobin (VHb) with Enhanced Peroxidase Activity for Efficient Degradation of Textile Dye

        Pollution resulting from the discharge of textile dyes into water systems has become a major global concern. Because peroxidases are known for their ability to decolorize and detoxify textile dyes, the peroxidase activity of Vitreoscilla hemoglobin (VHb) has recently been studied. It is found that VHb and variants of this enzyme show great promise for enzymatic decolorization of dyes and may play a role in achieving their successful removal from industrial wastewater. The level of VHb peroxidase activity correlates with two amino acid residues present within the conserved distal pocket, at positions 53 and 54. In this work, sitedirected mutagenesis of these residues was performed and resulted in improved VHb peroxidase activity. The double mutant, Q53H/P54C, shows the highest dye decolorization and removal efficiency, with 70% removal efficiency within 5 min. UV spectral studies of Q53H/P54C reveals a more compact structure and an altered porphyrin environment (λSoret = 413 nm) relative to that of wild-type VHb (λSoret = 406), and differential scanning calorimetry data indicate that the VHb variant protein structure is more stable. In addition, circular dichroism spectroscopic studies indicate that this variant’s increased protein structural stability is due to an increase in helical structure, as deduced from the melting temperature, which is higher than 90°C. Therefore, the VHb variant Q53H/P54C shows promise as an excellent peroxidase, with excellent dye decolorization activity and a more stable structure than wild-type VHb under high-temperature conditions.

        Expression, Purification, and Biological Characterization of The Amino-Terminal Fragment of Urokinase in Pichia pastoris

        Urokinase (uPA) and its receptor (uPAR) play an important role in tumor growth and metastasis. Targeting the excessive activation of this system as well as the proliferation of the tumor vascular endothelial cell would be expected to prevent tumor neovasculature and halt the tumor development. In this regard, the amino-terminal fragment (ATF) of urokinase has been confirmed as effective to inhibit the proliferation, migration, and invasiveness of cancer cells via interrupting the interaction of uPA and uPAR. Previous studies indicated that ATF expressed in Escherichia coli was mainly contained in inclusion bodies and also lacked posttranslational modifications. In this study, the biologically active and soluble ATF was cloned and expressed in Pichia pastoris. The recombinant protein was purified to be homogenous and confirmed to be biologically active. The yield of the active ATF was about 30 mg/l of the P. pastoris culture medium. The recombinant ATF (rATF) could efficiently inhibit angiogenesis, endothelial cell migration, and tumor cell invasion in vitro. Furthermore, it could inhibit in vivo xenograft tumor growth and prolong the survival of tumor-bearing mice significantly by competing with uPA for binding to cell surfaces. Therefore, P. pastoris is a highly efficient and cost-effective expression system for large-scale production of biologically active rATFs for potential therapeutic application.

        A New Triterpenoid from Brucea javanica

        A new triterpenoid, bruceajavanin C (1), together with bruceosides A and B (2 and 3), bruceines D and E (4 and 5), yadanziosides A and G (6 and 7), (20R)-O-(3)-α-L-arabinopyranosylpregn- 5-ene-3β,20-diol (8), and α-D-glucopyranoside, (3β, 20R)-3-hydroxypregn-5-en-20-yl (9) were isolated from the aerial parts of Brucea javanica. The structure of 1 was elucidated on the basis of 2D-NMR spectroscopic analysis. In addition, compounds 1, 3, 4, 5, and 6 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.

        Establishment and Analysis of Erosion Depth Model for Impeller Material FV520B

        The multifactorial erosion was conducted in this paper to test the compressor impeller material FV520B using high-speed gas-solid two phase flow erosion tester and surface morphology analysis method. Based on the particle motion and collision energy equation as well as regression analysis of multi-factor orthogonal experiment, a phenomenological erosion depth model which captures the effects of impact velocity, angle and particle size, has been developed. The model includes removal of material due to both deformation damage and micro-cutting. Results show that the peak of experiment depth and the maximum calculated depth all appeared at near 45o, rather than near 24o where the maximum erosion rate appeared. Comparing the calculated values and the results of each single factor experiment, the errors are within 15%. The predictions of the simplified version of the model were in good agreement with the results of single factor experiments. Also, the reliability of the assessment formula was verified to assess the impeller erosion life, which indicated that this calculation model could be used to estimate the erosion depth of compressor impeller material FV520B.
