        Centromere protein N promotes lung adenocarcinoma progression by activating PI3K/AKT signaling pathway

        Background: As an important member of centromere family, centromere associated protein N (CENPN) was abnormally expressed in varied malignant tumors. Objective: This paper aimed to analyze the expression and related mechanism of CENPN in lung adenocarcinoma (LUAD). Methods: The expression of CENPN in LUAD was analyzed by Gene Expression Profiling Interactive Analysis (GEPIA) database. The mRNA expression, protein expression, cell viability, cell invasion, cell apoptosis, cell stem like characteristics were detected by RT-PCR, western blot, CCK8 assay, transwell assay, flow cytometry and spheroidization assay, respectively. Finally, the pathological changes of xenograft were estimated by H&E staining, and the expression of proteins was detected by immunohistochemistry. Results: GEPIA analysis showed that the CENPN expression in LUAD was significantly higher than that in normal lung tissue, which was negatively correlated with the prognosis. These results were consistent with our clinical data. Besides, CENPN was highly expressed in LUAD cell lines. In addition, the upregulation of CENPN amplified the cell viability, stemness and invasive ability in PC9 cells. However, the knockdown of CENPN inhibited the cell activity, stemness, invasive ability with increased cell apoptosis in A549. Furthermore, CENPN could positively regulate the phosphorylation of PI3K and AKT. The PI3K inhibitor, 740Y-P, could reverse the effect of CENPN silencing on the expression of Ki-67, cleaved caspase 3, OCT4, and snail 1. Finally, the downregulation of CENPN restrained the growth of xenograft and inactivated the PI3K/AKT pathway. Conclusion: CENPN was abnormally overexpressed in LUAD, and promoted tumor progression of LUAD by affecting PI3K/AKT pathway.

      • Gene Expression Profiles of HeLa Cells Impacted by Hepatitis C Virus Non-structural Protein NS4B

        By a cDNA array representing 2308 signal transduction related genes, we studied the expression profiles of HeLa cells stably transfected by Hepatitis C virus nonstructural protein 4B (HCV-NS4B). The alterations of the expression of four genes were confirmed by real-time quantitative RT-PCR; and the aldo-keto reductase family 1, member C1 (AKR1C1) enzyme activity was detected in HCV-NS4B transiently transfected HeLa cells and Huh-7, a human hepatoma cell line. Of the 2,308 genes we examined, 34 were up-regulated and 56 were down-regulated. These 90 genes involved oncogenes, tumor suppressors, cell receptors, complements, adhesions, transcription and translation, cytoskeletion and cellular stress. The expression profiling suggested that multiple regulatory pathways were affected by HCV-NS4B directly or indirectly. And since these genes are related to carcinogenesis, host defense system and cell homeostatic mechanism, we can conclude that HCV-NS4B could play some important roles in the pathogenesis mechanism of HCV.

        일본유교와 "황국(皇國) 국체" 이데올로기

        서구와 동양을 접목시켜 새롭게 이론을 창출하는 ‘탈구축’ 이론 속에는 ‘서구와 동양의 내용 양자’를 통합하여 외부 위치로 새롭게 재구성해 내는 것이라고 보는 시각이 존재한다. 그렇지만 이 탈구축 이론은 실은 오카와 슈메이(大川周明)의 논리에서 보면 ‘반쪽 탈구축’이었다. 오카와 슈메이의 탈구축 이론 속에는 ‘제3의 입장에서’ 심급을 통해 양쪽을 무효화 시키면서 그 구조 자체를 논하기 보다는 전혀 다른 구조를 찾아내는 것으로서 나머지 반쪽 탈구축을 통해 ‘완전한 내적 공동체 논리로서 천황제’를 탈구축해 냈다. 그것은 일본의 ‘우익적사고’가 전통적 탈구축 이론과 다른 구조를 찾아내는 의미에서의 탈구축이 접목되는 특성을 가졌다는 것을 설명해 주는 것이다. 이에 대표적인 사상가가 바로 오카와 슈메이였다. 오카와 슈메이는 『대학』과 『중용』을 ‘유교사상’적 측면에서 관심을 가지면서도 종교철학적 시대적 흐름 속에서 이를 재해석해 냈다. 물론 오카와 슈메이가 사상가인가 철학자인가라는 문제라기보다는, 대학과 중용을 해독하는 방법론에서, 유교가 ‘종교, 도덕, 정치’가 공통적으로 갖고 있는 것을 ‘도(道)’라고 규정하면서 출발하는데, 그것은 종교학이나 종교사 연구에서 ‘모든 종교의 공통적 특성’을 극대화시킨 작업과 연결된다는 점이었다. 이를 통해 새롭게 학문의 지평을 여는 방식을 ‘원리’를 체현해 냈다. 오카와 슈메이는 대학에서는 ‘친민(親民)’을 ‘신민(新民)’이라고 해석하면서 수도(修道)의 ‘수(修)’를 ‘수양한다’가 아니라 ‘수양해야 하는것’으로 다르게 해석한다. 그런데 중요한 것은 개인 개인에게 모두 선험적으로 주어진 도는 수양이 중요하기 때문이었다. 그것은 바로 개인의 수양과 신민을 통한 새로운 사회 구현이 필요했기 때문이다. 이것에는 도의 수양과 성이 핵심으로 작용한다고 보았다. 이를 통해 오카와 슈메이는 유교가 서구 학자들이 주장하는 개인과 사회의 관계를 통합해주고, 유교 내부에서의 논쟁을 극복해 냈다. 그 과정에서 중(中)은, 시대적 흐름에 변화하지 않는 보편성을 가진 것으로 동시에 자아와 타자를 초월하여 존재 것을 자각하는 ‘경지’로 설명된다. 그것은 중국에서는 없어진 ‘전통과 황통이 일본의 ‘신도’ 속에 그것이 모두 포함된다고 주장했다. 그것은 바로 ‘자연적인 것=천사상과 도덕적인 것=성과 덕 그리고 평등한 것=천지인’을 통해, 황국 국체가 『대학』과 『중용』에서 찾을 수 있다는 ‘중용이데올로기’를 창출해 낸 것이었다. In the theory of “deconstruction” that creates a new theory by grafting the west and the orient, there is the point of view that ‘both sides of the west and the orient’is combined and newly restructured to “outer” location. However, this deconstruction theory was ‘half deconstruction’ in the logic of Okawa Shumei. The theory of deconstruction of Okawa Shumei did not discuss the structure itself by nullifying both sides through the instance ‘from the stance of the third party’, but found a whole new structure, deconstructing ‘the Emperor system as a ‘logic of complete internal community’ through the half deconstruction. One of the representative theorists was Okawa Shumei. He not only took an interest into “Daehak” and “Jungyong” from the aspect of ‘Confucian ideas’ but also reinterpreted the theories in a philosophy of religion and stream of times. Of course, rather than the issue that Shumei is a theorist or philosopher, the issue starts from describing the common values of ‘religion, ethics and politics’ of Confucianism as ‘morality’. This is linked to the works maximized the ‘common characteristics of all religions’ in researches on science of religion or religious history. By doing so, he materialized the ‘principle’ of how to open a new prospect in the field of the study. Okawa Shumei interpreted ‘citizens(親民)’ as ‘subjects(新民)’in Daehak and ‘ascetic(修)’ of ascetic practice(修道) as ‘have to cultivate’ rather than ‘cultivate’. Because what’s important was self-discipline of morality intuitively given to individuals. This indicated that self-discipline of morality and sincerity played a key role. By doing so, Okawa Shumei showed that Confucianism integrated the relationship between individuals and nation argued by the western scholars and overcome the disputes within Confucianism. Middle during the process has universality that does not change by the stream of times and at the same time is explained as a ‘level’ to recognize presence by transcending ego and others. It was argued that the lost ‘tradition and imperial line’ in China were all included in Japanese Shinto. In other words, based on the idea of ‘something natural = heavens thought and something moral = sincerity and virtue and something equal = heaven, land, benevolence(仁)’, the forms of the empire created ‘Jungyong ideology’ that can be found in “Daehak” and “Jungyong”.

        Hepatitis C Virus Non-structural Protein NS4B Can Modulate an Unfolded Protein Response

        Viral infection causes stress to the endoplasmic reticulum (ER). The response to endoplasmic reticulum stress, known as the unfolded protein response (UPR), is designed to eliminate misfolded proteins and allow the cell to recover. The role of hepatitis C virus (HCV) non-structural protein NS4B, a component of the HCV replicons that induce UPR, is incompletely understood. We demonstrate that HCV NS4B could induce activating transcription factor (ATF6) and inositol-requiring enzyme 1 (IRE1), to favor the HCV subreplicon and HCV viral replication. HCV NS4B activated the IRE1 pathway, as indicated by splicing of X box-binding protein (Xbp-1) mRNA. However, transcriptional activation of the XBP-1 target gene, EDEM (ER degradation-enhancing $\alpha-mannosidase-like$ protein, a protein degradation factor), was inhibited. These results imply that NS4B might induce UPR through ATF6 and IRE1-XBP1 pathways, but might also modify the outcome to benefit HCV or HCV subreplicon replication.

      • Study on the Optimization Path of Community Sports Operation and Service in China under the Background of Continuous Population Aging

        Purpose - The purpose of this paper is to serve the national strategy of significantly increasing per capita healthy life expectancy, reducing chronic disease morbidity and mortality, and improving health literacy by constructing an integrated community-based middle-aged and old-aged physical education and sport service system in the context of China’s aging population. Design/Methodology/Approach - Based on the theory of structural functionalism and through literature references, expert interviews, questionnaire surveys, mathematical statistics and other methods, a comprehensive service system for middle-aged and old-aged sports is fully demonstrated and constructed with the in-depth integration of the government, community, sports, and recreation and health care to cope with the status quo of a continuously aging population. Findings - This paper concludes that the construction of community sports comprehensive service system for the middle-aged and the elderly fits the development concepts and practical needs of “treatment of the future disease” and “integration of sports and recreation”, and realizes the value co-creation of community sports and recreation in four functional needs. On this basis, the optimization path of community sports operation and service is proposed. Research Implications - In the process of building and operating a comprehensive community sports service system for the middle-aged and the elderly, it is necessary to give full play to the positive roles of government-led, community-based and social participation, and to absorb and integrate favorable resources from all walks of life, so as to effectively build a community of integrated development of comprehensive sports services.

        Resource Allocation with Proportional Rate In Cognitive Wireless Network: An Immune Clonal Optimization Scheme

        In this paper, the resource allocation problem with proportional fairness rate in cognitive OFDM-based wireless network is studied. It aims to maximize the total system throughput subject to constraints that include total transmit power for secondary users, maximum tolerable interferences of primary users, bit error rate, and proportional fairness rate among secondary users. It is a nonlinear optimization problem, for which obtaining the optimal solution is known to be NP-hard. An efficient bio-inspired suboptimal algorithm called immune clonal optimization is proposed to solve the resource allocation problem in two steps. That is, subcarriers are firstly allocated to secondary users assuming equal power assignment and then the power allocation is performed with an improved immune clonal algorithm. Suitable immune operators such as matrix encoding and adaptive mutation are designed for resource allocation problem. Simulation results show that the proposed algorithm achieves near-optimal throughput and more satisfying proportional fairness rate among secondary users with lower computational complexity.

      • A Multiple Moving Object Segmentation Algorithm Based on Background Modeling and Adaptive Clustering

        A multiple moving object segmentation algorithm based on Background Modeling and Adaptive Clustering (named as BMAC) algorithm is proposed in this paper. For moving object segmentation, the algorithm uses Chebyshev inequality and the kernel density estimation method to do background modeling firstly. Then in order to classify image pixels as background points, foreground points and suspicious points, an adaptive threshold algorithm is proposed accordingly. After using background modeling, adaptive clustering is used for multi-object segmentation. It defines pixel space connectivity rate and designs a perpendicular split method, initial cluster adaptive splitting and merging self-organizing the iterative clustering segmentation algorithm, without pre-set number of clustering, completes multi-object segmentation for the foreground image. The segmentation results are consistent with the human visual judgment, the use of space connectivity information improve the accuracy of clustering segmentation, comparison and analysis the experimental results show that the proposed algorithm is feasible, rapid and effective.

        Protective strategy for the caudate lobe bile duct during left hemihepatectomy based on imaging data analysis

        Purpose: This study was performed to analyze the rule of confluence of the caudate lobe bile duct (CLD) into the left hepatic duct (LHD) and to discuss the protective strategy during left hemihepatectomy. Methods: MRI of 400 patients and T-tube angiography images of 100 patients were collected, and the imaging rules of the confluence of the CLD into the LHD were summarized. The clinical data of 33 patients who underwent left hemihepatectomy using the protective strategy were analyzed. Results: MRI and T-tube angiography images showed that the length from the confluence point of the CLD into the LHD to the confluence of the left and right hepatic ducts was 1.19 ± 0.40 cm and 1.26 ± 0.39 cm, respectively. The average angle between the longitudinal axis of the 2 bile ducts was 68.27° ± 22.59° and 66.58 ± 22.88°, respectively. Coronal and cross-sectional images showed that inflow from the foot side to the cranial side was noted in 79.8% and 82.0% of patients, respectively, and inflow from the dorsal to the ventral side was observed in 84.5% and 88.0%, respectively. Based on these imaging rules, the safe transection length and plane were summarized, and the CLD was effectively protected in 33 cases of left hemihepatectomy. Conclusion: In left hemihepatectomy, the LHD should be transected at least 1.5 cm away from the confluence of the left and right hepatic ducts, and the plane of transection should be oblique to the dorsal side at an angle of 45° with the LHD, these parameters represent an effective strategy to protect the CLD.
