        試論≪莊子≫之“眞”的評判標準

        The concept of “Zhen” in Zhuangzi means nature and true of all things in the world. Two both fables of “Qinshi Diao Laodan”and “Zhuangzi Gu Pen er Ge” in Zhuangzi illustrate the concept of “Zhen”. They are also two good examples exemplifying his ideas, in addition, they are the practice in his philosophy. These two fables perfectly clarify philosophy of Zhuangzi. That is the concept of “Zhen”. In life, we should not impose on others do what you would not want to do and what you desire to do, because people can only know themselves and can't know others, that is why people only know the “Zhen” of themselves, not judge other's false. Therefore, there is no fixed standard of “Zhen”. Person who follows the original intention and agrees with his own heart is called “Zhen”. This is the core connotation of Zhuangzi's “Zhen”. The author makes this connotation down to “Follow your mind”, “Follow your mind” that is do what you want to in your mind, so the only judgment standard of “Zhen” is whether do things follow your mind.

        ORIGINAL ARTICLE : Synergistic Effect of Bone Marrow-Derived Mesenchymal Stem Cells and Platelet-Rich Plasma in Streptozotocin-Induced Diabetic Rats

        Background: Diabetic wounds are a major clinical challenge, because minor skin wounds can lead to chronic, unhealed ulcers and ultimately result in infection, gangrene, or even amputation. Studies on bone marrow derived mesenchymal stem cells (BMSCs) and a series of growth factors have revealed their many benefits for wound healing and regeneration. Platelet-rich plasma (PRP) may improve the environment for BMSC development and differentiation. However, whether combined use of BMSCs and PRP may be more effective for accelerating diabetic ulcer healing remains unclear. Objective: We investigated the efficacy of BMSCs and PRP for the repair of refractory wound healing in a diabetic rat model. Methods: Forty-eight rats with diabetes mellitus induced by streptozotocin were divided into four groups: treatment with BMSCs plus PRP, BMSCs alone, PRP alone, phosphate buffered saline. The rate of wound closure was quantified. A histopathological study was conducted regarding wound depth and the skin edge at 7, 14, and 28 days after surgery. Results: Wound healing rates were significantly higher in the BMSC plus PRP group than in the other groups. The immunohistochemistry results showed that the expression of platelet/endothelial cell adhesion molecule 1, proliferating cell nuclear antigen, and transforming growth factor-β1 increased significantly in the BMSC plus PRP group compared to the other treatment groups. On day 7, CD68 expression increased significantly in the wounds of the BMSC plus PRP group, but decreased markedly at day 14 compared to the controls. Conclusion: The combination of BMSCs and PRP aids diabetic wound repair and regeneration. (Ann Dermatol 26(1) 1∼10, 2014)

        The Dynamic Mechanical Properties for Recycled Aggregate Concrete under Tensile-Compressive States

        The experiments under biaxial tensile-compressive (T-C) complex stress were carried out for two kinds of substitution percentages of recycled coarse aggregate (RCA) on biaxial proportional loading (α = σ1:σ3 =1:0, 0: -1, 0.05: -1, 0.1: -1, 0.25: -1, 0.5: -1, 1: -1 and-1: -1) and four loading deformation rates of 10−5/s, 10−4/s , 10−3/s, and 10−2/s. The research was completed in the static-dynamic true triaxial machine. The experimental phenomena indicate the tensile failure appears in these specimens of recycled aggregate concrete (RAC) under biaxial T-C stress states. The dynamic mechanical behavior for RAC under biaxial T-C loadings was affected by the strain rates, stress ratios and its substitution percentages of RCA. Accompanied by the increase of strain rates, its strength of RAC is improved. As the tensile stress increases, its compressive strength decreases. The T-C strengths for RAC under biaxial T-C states are less than its corresponding uniaxial T-C strengths at a certain identical strain rates, respectively. Considering the effect of loading rates, a new tensile-compressive failure criterion is established under complex stress states.

        A Study of the Content Validity of Reading Comprehension Tests in NMET II

Based on Bachman and Palmer's (1996) framework of task characteristics, this paper aims to compare the reading comprehension tests of the 2013-2017 National Matriculation English Test II (NMET II) with the requirements of the New Curriculum Standards and Testing Syllabus, in particular, concerning the content validity of the tests in terms of the characteristics of input and expected responses. The implications of this study are discussed for test designers, teachers, and students. The results show that reading comprehension tests from 2013 to 2017 basically meet the requirements of the New Curriculum Standard and Testing Syllabus and have a high content validity. In terms of the characteristics of input, the length, reading speed, and readability of comprehension passages are appropriate for students; however, the distribution of topics and genres in each year is uneven. In terms of the characteristics of expected responses, the wordings of questions and the design of question items are reasonable. However, the response type is mainly restricted to selected response questions (multiple-choice), and the measure of reading skills coverage is unbalanced.

        Influence of Initial Pearlite Morphology on the Microstructure Evolution During Heat Treatment of 1.0C–1.5Cr Steel

        Effect of initial pearlite morphology on the microstructure evolution during spheroidization annealing and subsequent hardeningtreatment was studied in hot-rolled 1.0C–1.5Cr bearing steel. The ferrite-to-austenite transformation in spheroidizationannealing can be accelerated by refining the pearlite interlamellar spacing and pearlite colony size. Decreasing interlamellarspacing is also beneficial to increasing the number density of undissolved cementite, leading to the refining of final spheroidizedcementite. During the re-austenitizing process of hardening treatment, the smaller initial spheroidized cementiteleads to faster cementite dissolution and finer undissolved cementite particles. The prior austenite grain size after hardeningtreatment can also be decreased by refining the initial pearlite microstructure.

        Targeted Suppression of Connexin 43 in Ovine Preimplantation Embryos by RNA Interference Using Long Double-stranded RNA

        RNA interference (RNAi) is an acknowledged useful and effective tool to study gene function in various cells. Here, we suppressed the Connexin 43 (Cx 43) gene expression during in vitro development of ovine pre-implantation embryos using the RNAi method. The 353 bp Cx 43 double-stranded RNA was microinjected into in vitro fertilized ovine zygotes, and the levels of target mRNA and protein were investigated. Control groups included uninjected zygotes or those injected with RNase-free water. The dsRNA injection resulted in the specific reduction of Cx 43 transcripts as analyzed by quantitative real-time RT-PCR and decreased protein levels as shown by Western blot analysis at the blastocyst stage. Microinjection of Cx 43 dsRNA led to 20.3%, 21.7% and 34.5% blastocyst rates and 19.2%, 37.5% and 41.3% hatched blastocyst rates in Cx 43 dsRNA-injected, water-injected and uninjected groups, respectively. Then the RNAi could not significantly affect cell number and cell death rates of blastocysts. Therefore, suppression of Cx 43 dsRNA and proteins did not apparently affect the development potential of ovine pre-implantation embryos but may play a role in embryo quality. RNAi technology is a promising approach to study gene function in early ovine embryogenesis.

      • Sensitivity of Gastric Cancer Cells to Chemotherapy Drugs in Elderly Patients and Its Correlation with Cyclooxygenase-2 Expression

        Objective: To explore the sensitivity of gastric cancer cells to chemotherapy drugs in elderly patients and its correlation with cyclooxygenase-2 (COX-2) expression in cancer tissue. Materials and Methods: Forty-three elderly patients with gastric cancer (observation group) and 31 young patients with gastrointestinal tumors (control group) who were all diagnosed by pathology and underwent surgery in the 89th Hospital of Chinese People's Liberation Army were selected. Drug sensitivity testing of tumor cells in primary culture was carried out in both groups using a methyl thiazolyl tetrazolium (MTT) method, and the expression of COX-2 and the factors related to multi-drug resistance (MDR) in cancer tissue were assessed by immunohistochemistry. Results: The inhibition rates (IR) of vincristine (VCR), 5-fluorouracil (5-FU), oxaliplatin (L-OHP), mitomycin (MMC) and epirubicin (eADM) on tumor cells in the observation group were dramatically lower than in the control group, with statistical significance (P<0.05 or P<0.01). The positive rates of COX-2, glutathione s-transferase-${\pi}$ (GST-${\pi}$) and P glycoprotein (P-gp) expression in cancer tissue in the observation group were all higher than in control group (P<0.05), while that of DNA topoisomerase $II{\alpha}$ ($TopoII{\alpha}$) expression lower than in the control group (P<0.01). In the observation group, COX-2 expression in cancer tissue had a significantly-positive correlation with GST-${\pi}$ and P-gp (r=0.855, P=0.000; r=0.240, P=0.026), but a negative correlation with $TopoII{\alpha}$ (r=-0.328, P=0.002). In the control group, COX-2 expression in cancer tissue was only correlated with P-gp positively (r=0.320, P=0.011). Bivariate correlation analysis displayed that COX-2 expression in cancer tissue in the observation group had a significantly-negative correlation with the IRs of 5-FU, L-OHP, paclitaxel (PTX) and eADM in tumor cells (r=-0.723, P=0.000; r=-0.570, P=0.000; r=-0.919, P=0.000; r=-0.781, P=0.000), but with hydroxycamptothecine (HCPT), VCR and 5-FU in the control group (r=-0.915, P=0.000; r=-0.890, P=0.000; r=-0.949, P=0.000). Conclusions: Gastric cancer cells in elderly patients feature stronger MDR, which may be related to high COX-2 expression.
