Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)

Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax,namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax

Mesenchymal stem cells (MSCs) are widely used in cartilage tissue engineering to repair articular cartilage defects. However, hypertrophy of chondrocytes derived from MSCs might hinder the stabilization of hyaline cartilage. Thus, it is very important to find a suitable way to maintain the chondrogenic phenotype of chondrocytes. It has been reported that cordycepin has anti-inflammatory and anti-tumor functions. However, the role of cordycepin in chondrocyte hypertrophy remains unclear. Therefore, the objective of this study was to determine the effect of cordycepin on chondrogenesis and chondrocyte hypertrophy in MSCs and ATDC5 cells. Cordycepin upregulated chondrogenic markers including Sox9 and collagen type II while down-regulated hypertrophic markers including Runx2 and collagen type X. Further exploration showed that cordycepin promoted chondrogenesis through inhibiting Nrf2 while activating BMP signaling. Besides, cordycepin suppressed chondrocyte hypertrophy through PI3K/Bapx1 pathway and Notch signaling. Our results indicated cordycepin had the potential to maintain chondrocyte phenotype and reconstruct engineered cartilage. [BMB Reports 2016; 49(10): 548-553]

The present study aimed to investigate the function of translationally controlled tumor protein (TCTP) in the regulation of Oct4 in mouse embryonic carcinoma P19 cells and mouse J1 embryonic stem (ES) cells. The mRNA level of endogenous TCTP in somatic cells was 2-4 folds higher than that in pluripotent P19 and J1 ES cells. Overexpression of TCTP in mouse pluripotent cells not only reduced the level of Oct4 transcription, but also decreased the pluripotency of stem cells. The N-terminal end of TCTP (amino acids 1-60) played an important role in suppressing the Oct4 promoter. Moreover, overexpression of TCTP in P19 cells suppressed the Oct4 promoter activity in a dose- and a time-dependent manner. In addition, knockdown of TCTP by small interfering RNA increased the expression of Oct4. Our study indicates that TCTP downregulates the Oct4 expression by binding the Sf1 site of Oct4 promoter in mouse pluripotent cells. [BMB reports 2012; 45(1): 20-25]

The differentially virulent race T1 of common bunt (Tilletia tritici) was used to inoculate the wheat lines Neepawa (compatible) and its sib BW553 (incompatible) that are nearly isogenic for the Bt-10 resistance gene. Inoculated crown tissues were used to construct a suppression subtractive hybridization (SSH) cDNA library. Of the 1920 clones arrayed from the SSH cDNA library, approximately 10% were differentially regulated. A total of 168 differentially up-regulated and 25 down-regulated genes were identified and sequenced; 71% sequences had significant homology to genes of known function, of which 59% appeared to have roles in cellular metabolism and development, 24% in abiotic/biotic stress responses, 3% involved in transcription and signal transduction responses. Two putative resistance genes and a transcription factor were identified among the up regulated sequences. The expression of several candidate genes including a lipase, two non-specific lipid transfer proteins (ns-LTPs), and several wheat pathogenesis-related (PR)-proteins, was evaluated following 4 to 32 days post-inoculation in compatible and incompatible interactions. Results confirmed the higher overall expression of these genes in resistant BW553 compared to susceptible Neepawa, and the differential up-regulation of wheat lipase, chitinase and PR-1 proteins in the expression of the incompatible interaction.

<P>Microcellular polypropylene (PP)/WGRT blends, a new outlet for the recycling of waste tire rubber, were prepared in an injection-molding process by using a chemical blowing agent. The effects of WGRT content and chemical blowing agent content on the density, cell morphology, and physicomechanical properties of the foamed PP/WGRT blends were investigated. The foam morphologies were characterized in terms of void fraction, average cell size, and cell density. The results indicated that both the WGRT and the blowing agent content had huge effects on the cell morphology and tensile properties of the PP/WGRT foams. J. VINYL ADDIT. <P><B>Abstract</B></P><P>Nowadays the economic recycling of waste tires has become a global challenge. The use of waste tire powder as a dispersed elastomeric phase in a polypropylene (PP) matrix offers an interesting opportunity for recycling of waste tire rubber. Compatibilized PP/(waste tire powder) composites are microcellularly processed to create a new class of materials with unique properties. Recent studies have demonstrated the feasibility of developing microcellular structures in PP/waste ground rubber tire (WGRT) composites. Microcellular PP/WGRT composites are prepared by an injection‐molding process using a chemical blowing agent. In this study, cell sizes, cell density, void fraction, and mechanical properties of the composite foams were measured, as well as the shear viscosity of the unfoamed composites. The influence of various compatibilizers and processing temperatures on cell morphology and the mechanical properties of injection‐molded PP/WGRT composites were investigated. It was seen that the addition of maleic anhydride‐grafted styrene‐ethylene‐butylene‐styrene (SEBS‐<I>g</I>‐MA) increased the shear viscosity of the composites. The void fraction and cell density of the PP/WGRT composites increased with addition of compatibilizers, whereas the average cell sizes decreased. A processing temperature range of 180–195°C gave finer microcellular structure and regular cell distribution. <P>A new approach towards the recycling of waste ground rubber tire (WGRT) powder was demonstrated in this study by introducing the polypropylene/ waste ground rubber tire (PP/WGRT) foaming method by using CO<SUB>2</SUB> as the foaming agent in an extrusion foaming process. The regression models were constructed to study the relationships between the foam structure (i.e., void fraction, average cell size, and cell density) of foamed PP/WGRT blends, the processing parameters (extruder’s die temperature and CO<SUB>2</SUB> concentration), and WGRT content by applying a three-factor central composite design (CCD) statistical approach. The response surface plots generated using the regression models allow the rapid selection of the proper process parameters to obtain microcellular PP/WGRT blends with the desired density and morphology.</P>
웹소설 이용만족도의 결정요인에 관한 연구 -중국 사례를 중심으로-

본 연구는 중국 웹소설 이용자의 동기구조를 발견하고 만족도에 영향을 미치는 요인을 규명하였다. 분석결과, 첫째, 웹소설 이용의 동기는 ‘사회적 상호작용’, ‘유용성’, ‘호기심충족’, ‘오락지향’, ‘자아표현’, ‘편리성’ 등 6개 요인으로 구성되었다. 둘째, 웹소설 이용동기 요인은 모두 만족도에 영향력을 행사하는 것으로 나타났다. 이용만족도 요인 중 ‘사회적 상호작용’ 요인이 가장 강력하였으며, 이어 ‘호기심 충족’, ‘자아표현’, ‘오락지향’ ‘편리성’, ‘유용성’ 등의 순이었다. 반면, 인구사회학적 요인(연령, 학력, 수입 등)은 웹소설 이용만족도에 영향력을 행사하지 못한 것으로 드러났다. 즉, 웹소설 이용만족도는 연령, 학력, 수입 등과는 무관하고, 단지 어떤 동기를 보유하였느냐에 따라서 결정된다고 할 수 있다. 이용만족도가 윕소설 이용동기에 따라서 달리 나타난다는 점을 실증적으로 규명하였다는 점에서 연구의 의미가 있다. This study examined the motivational factors of web novel users and analyzed the determinants of satisfaction. The analysis showed followings. First, Chinese web novels users’ motivational structure was made of six factors: ‘social interaction’, ‘utility’, ‘satisfaction of curiosity’, ‘entertainment orientation’, ‘self-expression’, and ‘convenience’. Second, these six factors influenced users’ satisfaction level. The ‘social interaction’ exerted the strongest influence. It was followed by ‘satisfaction of curiosity’, ‘self-expression’, ‘entertainment orientation’, ‘convenience’ and ‘utility’ in order. On the other hand, demographic factors(age, education, income, etc.) was irrelevant with the satisfaction of using web novels. In other words, users’ satisfaction was not affected by age, education, income, etc. The importance of this study would be that it showed on the basis of empirical data the level of satisfaction would vary according to web novel users’ motivational factors.
