        Experimence Study of Trace Water and Oxygen Impact on SF6 Decomposition Characteristics Under Partial Discharge

        It is common practice to identify the insulation faults of GIS through monitor the contents of SF6 decomposed components. Partial discharges (PD) could lead to the decomposition of SF6 dielectric, so new reactions usually occur in the mixture of the newly decomposed components including traces of H2O and O2. The new reactions also cause the decomposed components to differ due to the different amounts of H2O and O2 even under the same strength of PD. Thus, the accuracy of assessing the insulation faults is definitely influenced when using the concentration and corresponding change of decomposed components. In the present research, a needle-plate electrode was employed to simulate the PD event of a metal protrusion insulation fault for two main characteristic components SO2F2 and SOF2, and to carry out influence analysis of trace H2O and O2 on the characteristic components. The research shows that trace H2O has the capability of catching an F atom, which inhibits low-sulfide SFx from recombining into high-sulfide SF6. Thus, the amount of SOF2 strongly correlates to the amount of trace H2O, whereas the amount of SO2F2 is weakly related to trace H2O. Furthermore, the dilution effect of trace O2 on SOF2 obviously exceeds that of SO2F2.

        An efficient protocol for Agrobacterium-mediated transformation of the biofuel plant Jatropha curcas by optimizing kanamycin concentration and duration of delayed selection

        Jatropha curcas is considered a potential biodiesel feedstock crop. Currently, the value of J. curcas is limited because its seed yield is generally low. Transgenic modification is a promising approach to improve the seed yield of J. curcas. Although Agrobacterium-mediated genetic transformation of J. curcas has been pursued for several years, the transformation efficiency remains unsatisfying. Therefore, a highly efficient and simple Agrobacterium-mediated genetic transformation method for J. curcas should be developed. We examined and optimized several key factors that affect genetic transformation of J. curcas in this study. The results showed that the EHA105 strain was superior to the other three Agrobacterium tumefaciens strains for infecting J. curcas cotyledons, and the supplementation of 100 mM acetosyringone slightly increased the transient transformation frequency. Use of the appropriate inoculation method, optimal kanamycin concentration and appropriate duration of delayed selection also improved the efficiency of stable genetic transformation of J. curcas. The percentage of b-glucuronidase positive J. curcas shoots reached as high as 56.0 %, and 1.70 transformants per explant were obtained with this protocol. Furthermore, we optimized the root-inducing medium to achieve a rooting rate of 84.9 %. Stable integration of the T-DNA into the genomes of putative transgenic lines was confirmed by PCR and Southern blot analysis. Using this improved protocol, a large number of transgenic J. curcas plantlets can be routinely obtained within approximately 4 months. The detailed information provided here for each step of J. curcas transformation should enable successful implementation of this transgenic technology in other laboratories.

        Studies on PEGylated Gold Nanoparticles Loaded with 5-Hydroxydecanoate for Chemo-Photothermal Therapy of Human Lung Adenocarcinomas In Vitro

        Gold nanoparticles (Au NPs) have promising applications in the fields of drug delivery and photothermal therapy. 5-hydroxydecanoate (5-HD) is a kind of highly selective mitochondrial ATP sensitive potassium (mitoKATP) channel blocker. In this research, firstly, 5-HD was studied whether it could induce human lung adenocarcinomas (A549) cells apoptosis; secondly, PEGylated gold nanoparticles loaded with 5-HD (Au NPs-PEG-5-HD) were prepared to develop a new chemotherapy and photothermal therapy in one system under the irradiation of green light-emitting diode (LED). Subsequently, in vitro cytotoxicity test was analyzed by cell counting kit-8 (CCK-8), the change of mitochondrial membrane potential (Δψm) was determined by immunofluorescence microscopy with R-123 fluorescence, and cell apoptosis and necrosis rate were detected by flow cytometry. The results of CCK8 revealed that the inhibition rates of A549 cells were all greatly increased when cells were treated with free 5-HD, free 5-HD +LED irradiation, Au NPs-PEG-5-HD and Au NPs-PEG-5-HD+LED irradiation, and Au NPs-PEG-5-HD had enhanced cell-killing effect compared with 5-HD, furthermore, the Au NPs-PEG-5-HD and LED irradiation played a very great synergy effect. The immunofluorescence microscopy data also exhibited a reduction of Δψm correspondingly. Flow cytometric analysis showed that the apoptosis rate of cells that were incubated with free 5-HD, 5-HD+LED irradiation, Au NPsPEG-5-HD or Au NPs–PEG–5-HD+LED irradiation significantly increased to about 9.2%, 10.7%, 18.3% or 12.4% and the percentage of necrosis cells increased to around 8.8%, 9.7%, 48.0% or 69.8%, respectively. In a word, all the results indicated that the 5-HD had shown the ability to induce A549 cells apoptosis as a chemotherapy agent, and its ability would be improved when 5-HD is loaded on PEGylated Au NPs, as well as Au NPs-PEG-5-HD exhibited significantly enhanced photothermal effects for treatment of lung adenocarcinomas.

        Aldehyde Volatiles Emitted in Succession from Mechanically Damaged Leaves of Poplar Cuttings

        Plant aldehydes are volatiles necessary to defenses against environmental stress. To explore their emissions in response to wounding, we performed gas chromatography-mass spectrometry (GC-MS) on cuttings from poplar (Populus simonii×P. pyramidalis ``Opera 8277``) that were mechanically damaged to mimic herbivore attack. We detected 16 aldehydes, including 11 linear saturated aldehydes, 3 linear unsaturated aldehydes, and 2 non-linear aldehydes. Emissions of these aldehydes were clearly enhanced by such treatment, and exhibited a similar pattern of change, i.e., increasing in the first 2 h, then sharply decreasing before rising again at about 12 h. Two release peaks for these aldehydes were observed. Therefore, we propose two pathways for the mediation of aldehyde emissions following damage. The first peak may represent emissions from plant storage pools, whereas the second release peak might result from greater formation de novo through an activated synthesis pathway.

        Myeloid-Derived Suppressor Cells Are Associated with Viral Persistence and Downregulation of TCR ζ Chain Expression on CD8<sup>+</sup> T Cells in Chronic Hepatitis C Patients

        Myeloid-derived suppressor cells (MDSCs) play an important role in impairing the function of T cells. We characterized MDSCs in two chronic hepatitis C (CHC) cohorts: a cross-sectional group that included 61 treatment-naive patients with CHC, 14 rapid virologic response (RVR) cases and 22 early virologic response (EVR) cases; and a longitudinal group of 13 cases of RVR and 10 cases of EVR after pegylated-interferon-${\alpha}$/ribavirin treatment for genotype 1b HCV infection. Liver samples from 32 CHC patients and six healthy controls were subjected to immunohistochemical analysis. MDSCs frequency in treatment-naive CHC was significantly higher than in RVR, EVR, or healthy subjects and was positively correlated with HCV RNA. Patients infected with HCV genotype 2a had a significantly higher frequency of MDSCs than those infected with genotype 1b. Decreased T cell receptor (TCR) ${\zeta}$ expression on $CD8^+$ T cells was significantly associated with an increased frequency of MDSCs in treatment-naive CHC patients and was restored by L-arginine treatment in vitro. Increased numbers of liver arginase-$1^+$ cells were closely associated with the histological activity index in CHC. The TCR ${\zeta}$ chain was significantly downregulated on hepatic $CD8^+$ T cells in CHC. During antiviral follow up, MDSCs frequency in peripheral blood mononuclear cells was directly correlated with the HCV RNA load in the plasma and inversely correlated with TCR ${\zeta}$ chain expression in $CD8^+$ T cells in both RVR and EVR cases. Notably, the RVR group had a higher frequency of MDSCs at baseline than the EVR group. Collectively, this study provides evidence that MDSCs might be associated with HCV persistence and downregulation of CD8 ${\zeta}$ chain expression.

        Molecular features and functional studies of transcription factor, Cap ‘n’ collar C, in the brown planthopper, Nilaparvata lugens Stål (Hemiptera Delphacidae)

        Cap ‘n’ collar C (CncC), a transcription factor, plays a vital role in the development of insect resistance by regulating the expression of multiple detoxifying genes. Clarifying the molecular characteristics of CncC and its pathway regulating insecticide resistance will aid the development of integrated pest control strategies. Here we cloned and identified the molecular feature of CncC genes from the brown planthopper (BPH), Nilaparvata lugens (Stål), (Hemiptera Delphacidae). The full-length open reading frame of NlCncC was 3015 nucleotides with 1005 amino acids. The deduced amino acid sequence has a high similarity with other insect homologs and contains the characteristic Cnc/bZip domain architecture. Functional studies showed that silencing NlCncC by RNA interference (RNAi) could downregulate the transcript expression of NlCYP6ER1, NlCYP6CW1, and NlCYP6AY1, which increase the susceptibility to imidacloprid in N. lugens. The identification of molecular characteristics of NlCncC and func tional studies in this paper will contribute to designing effective control strategies for N. lugens.

        Fast Extraction of Pedestrian Candidate Windows Based on BING Algorithm

        In the field of industrial applications, the real-time performance of the target detection problem is very important. The most serious time consumption in the pedestrian detection process is the extraction phase of the candidate window. To accelerate the speed, in this paper, a fast extraction of pedestrian candidate window based on the BING (Binarized Normed Gradients) algorithm replaces the traditional sliding window scanning. The BING features are extracted with the positive and negative samples and input into the two-stage SVM (Support Vector Machine) classifier for training. The obtained BING template may include a pedestrian candidate window. The trained template is loaded during detection, and the extracted candidate windows are input into the classifier. The experimental results show that the proposed method can extract fewer candidate window and has a higher recall rate with more rapid speed than the traditional sliding window detection method, so the method improves the detection speed while maintaining the detection accuracy. In addition, the real-time requirement is satisfied.

        The Effect of Herb-Partition Moxibustion on Toll-like Receptor 4 in Rabbit Aorta during Atherosclerosis

        Objective: To explore the mechanism of Toll-like receptor (TLR4) inhibition in the delay of formation of atherosclerosis by herb-partition moxibustion. Method: Seventy-five rabbits were randomly assigned to one of five groups: blank, atherosclerosis (AS) model, direct moxibustion, herb-partition moxibustion, and drug treatment. With the exception of the blank group, all rabbits were given a high-fat diet in addition to immunologic injury to create the AS model. The experiments were carried out for 16 weeks, at which time the aorta was removed from each rabbit. Immunohistochemical methods were used to detect the gray level of the aortic TLR4 to observe the immunologic competence of its antigens. Fluorescent quantitative polymerase chain reaction was used to detect the expression of TLR4 messenger RNA (mRNA) in the aorta. Results: The gray-scale value of TLR4 and the TLR4 mRNA expression significantly decreased (p < 0.05) in the direct moxibustion, herb-partition moxibustion, and drug treatment groups. Furthermore, the effects of the herb-partition moxibustion and drug treatment were superior to those of the direct moxibustion. Conclusion: Herb-partition moxibustion inhibits aortic TLR4 activity and mRNA expression,showing that herb-partition moxibustion delays the formation of atherosclerosis through the inhibition of TLR4 expression.
