A novel M2e-multiple antigenic peptide providing heterologous protection in mice

Swine influenza viruses (SwIVs) cause considerable morbidity and mortality in domestic pigs, resulting in a significant economic burden. Moreover, pigs have been considered to be a possible mixing vessel in which novel strains loom. Here, we developed and evaluated a novel M2e-multiple antigenic peptide (M2e-MAP) as a supplemental antigen for inactivated H3N2 vaccine to provide cross-protection against two main subtypes of SwIVs, H1N1 and H3N2. The novel tetra-branched MAP was constructed by fusing four copies of M2e to one copy of foreign T helper cell epitopes. A high-yield reassortant H3N2 virus was generated by plasmid based reverse genetics. The efficacy of the novel H3N2 inactivated vaccines with or without M2e-MAP supplementation was evaluated in a mouse model. M2e-MAP conjugated vaccine induced strong antibody responses in mice. Complete protection against the heterologous swine H1N1 virus was observed in mice vaccinated with M2e-MAP combined vaccine. Moreover, this novel peptide confers protection against lethal challenge of A/Puerto Rico/8/34 (H1N1). Taken together, our results suggest the combined immunization of reassortant inactivated H3N2 vaccine and the novel M2e-MAP provided cross-protection against swine and human viruses and may serve as a promising approach for influenza vaccine development.

In the presence of extremely high precision turning the complicated curved surface with geometry features of large slope gradient and multi convex-concave characteristics, the micron-scale deviation of contour resulting from the tracking deviation of the feed axis becomes non-negligible. To diminish the micron-scale contouring deviation, this study presents a valid contouring error estimation and pre-compensation method for slow tool servo (STS) precision turning. Aiming at estimating the tracking deviation of the axis of the turning machine tool in the machining process, the steady-state tracking deviation model of the axis is first constructed. The proposed multiply tangential estimation method is derived from the Taylor series of the foot point. Since the rotation feed movement of the C axis is the main movement in cutting forming movement for STS turning machining, the monotonicity of the displacement for the C axis is ensured. The compensation value for the axis position is derived through the Jacobi matrix of the STS turning machine. With the presented approach, the tool tip location deviation induced by the tracking deviation of the feed axis reduces from 1.087 μm to 0.025 μm. The measurement results of the comparison experiments indicate that with the presented method in this study, the contouring error of the machined part reduces from 5.72 μm to 4.36 μm. The micron-scale contouring error resulting from the tracking deviation of the feed axis is effectually compensated.

Novel reassortant H3N2 swine influenza viruses (SwIV)with the matrix gene from the 2009 H1N1 pandemic virushave been isolated in many countries as well as duringoutbreaks in multiple states in the United States, indicatingthat H3N2 SwIV might be a potential threat to public health. Since southern China is the world’s largest producer of pigs,efficient vaccines should be developed to prevent pigs fromacquiring H3N2 subtype SwIV infections, and thus limit thepossibility of SwIV infection at agricultural fairs. In thisstudy, a high-growth reassortant virus (GD/PR8) wasgenerated by plasmid-based reverse genetics and tested as acandidate inactivated vaccine. The protective efficacy of thisvaccine was evaluated in mice by challenging them withanother H3N2 SwIV isolate [A/Swine/Heilongjiang/1/05(H3N2) (HLJ/05)]. Prime and booster inoculation withGD/PR8 vaccine yielded high-titer serum hemagglutinationinhibiting antibodies and IgG antibodies. Completeprotection of mice against H3N2 SwIV was observed, withsignificantly reduced lung lesion and viral loads invaccine-inoculated mice relative to mock-vaccinatedcontrols. These results suggest that the GD/PR8 vaccine mayserve as a promising candidate for rapid intervention ofH3N2 SwIV outbreaks in China.

Microbial metabolites have emerged as critical components that mediate the metabolic effects of the gut microbiota. Here, we show that indole-3-propionic acid (IPA), a tryptophan metabolite produced by gut bacteria, is a potent anti-non-alcoholic steatohepatitis (NASH) microbial metabolite. Here, we demonstrate that administration of IPA modulates the microbiota composition in the gut and inhibits microbial dysbiosis in rats fed a high-fat diet. IPA induces the expression of tight junction proteins, such as ZO-1 and Occludin, and maintains intestinal epithelium homeostasis, leading to a reduction in plasma endotoxin levels. Interestingly, IPA inhibits NF-κB signaling and reduces the levels of proinflammatory cytokines, such as TNFα, IL-1β, and IL-6, in response to endotoxin in macrophages to repress hepatic inflammation and liver injury. Moreover, IPA is sufficient to inhibit the expression of fibrogenic and collagen genes and attenuate diet-induced NASH phenotypes. The beneficial effects of IPA on the liver are likely mediated through inhibiting the production of endotoxin in the gut. These findings suggest a protective role of IPA in the control of metabolism and uncover the gut microbiome and liver cross-talk in regulating the intestinal microenvironment and liver pathology via a novel dietary nutrient metabolite. IPA may provide a new therapeutic strategy for treating NASH.
Key challenges, recent advances and future perspectives of rechargeable lithium-sulfur batteries

Lithium-sulfur (Li-S) battery, which releases energy by coupling high abundant sulfur with lithium metal,is considered as a potential substitute for the current lithium-ion battery. Thanks to the lightweight andmulti-electron reaction of sulfur cathode, the Li-S battery can achieve a high theoretical specific capacityof 1675 mAh g1 and specific energy of 2600 Wh kg1. However, some key scientific issues limit its practicalapplication, such as the ‘‘shuttle effect” of lithium polysulfides, large volume change, poor conductivityof sulfur and its solid-state products, and self-discharge phenomenon. Of these, the ‘‘shuttle effect”is recognized as the most critical challenge affecting on electrochemical performance. Hence, this reviewfirst systematically introduces the development of Li-S batteries and the corresponding ‘‘shuttle effect”. Then, the latest work on anode, cathode, separator and electrolyte are summarized. Finally, some promisingviews on the future research direction of this battery system are put forward.

A series of 4′-N-substituted (aminomethyl)benzoate derivatives of scutellarein were designed and synthesized. Evaluation of their physiochemical properties showed that the designed target compounds 5a–e exhibit higher chemical stability and aqueous solubility than scutellarin and scutellarein. The permeability (Papp AP to BL ) of 5c–e in Caco-2 cells were 2.8, 8.1, and 12.6 times higher than that of scutellarin and 1.3, 4.1, and 6.0 times higher than that of scutellarein; especially, 5e had the highest P app AP to BL value (7.19 ± 0.31 × 10−6 cm/s) and the lowest ER (P app BL to AP /P app AP to BL ) value of 0.17. In vitro antioxidative evaluation results revealed that 5e could protect against H2O2 -induced PC12 cells’ oxidative damage by attenuating mitochondrial membrane potential loss and decreasing H2O2 -induced reactive oxygen species (ROS) production.
Stress-strain relationship for recycled aggregate concrete after exposure to elevated temperatures

In this paper, the effects of elevated temperatures on the strength and compressive stress-strain curve (SSC) of recycled coarse aggregate concrete with different replacement percentages are presented. 90 recycled coarse aggregate concrete prisms are heated up to 20, 200, 400, 600, 800°C. The results show that the compressive strength, split tensile strength, elastic modulus of recycled aggregate concrete specimens decline significantly as the temperature rise. While the peak strain increase of recycled aggregate concrete specimens as the temperature rise. Compared to the experimental curves, the proposed stress-strain relations for recycled aggregate concrete after exposure elevated temperatures can be used in practical engineering applications.
Novel approaches to pin cluster synchronization on complex dynamical networks in Lur’e forms

<P><B>Abstract</B></P> <P>This paper investigates the cluster synchronization of complex dynamical networks consisted of identical or nonidentical Lur’e systems. Due to the special topology structure of the complex networks and the existence of stochastic perturbations, a kind of randomly occurring pinning controller is designed which not only synchronizes all Lur’e systems in the same cluster but also decreases the negative influence among different clusters. Firstly, based on an extended integral inequality, the convex combination theorem and S-procedure, the conditions for cluster synchronization of identical Lur’e networks are derived in a convex domain. Secondly, randomly occurring adaptive pinning controllers with two independent Bernoulli stochastic variables are designed and then sufficient conditions are obtained for the cluster synchronization on complex networks consisted of nonidentical Lur’e systems. In addition, suitable control gains for successful cluster synchronization of nonidentical Lur’e networks are acquired by designing some adaptive updating laws.
