A glassy carbon electrode (GCE) modified with poly(p-aminobenzene sulfonic acid) [Poly(p-ABSA)] film is fabricated by voltammetric technique in phosphate buffer solution (pH 8.0) containing 5.010-3molL-¹ p-ABSA. Electrochemical behaviors of tryptophan at the Poly(p-ABSA) film electrode are investigated with voltammetry. The results indicate that the electrochemical response of tryptophan is improved significantly in the presence of poly(p-ABSA) film. Compared with the bare glassy carbon electrode, the Poly(p-ABSA) film electrode remarkably enhances the irreversible oxidation peak current of tryptophan. Some parameters such as voltammetric sweeping segments for the electrochemical polymerization, pH, accumulation potential and accumulation time are optimized. Under the optimal conditions, the oxidation peak current is proportional to tryptophan concentration in the range of 1.0 10-7 to 1.0 10-6 mol L-¹, and 2.0 10-6 to 1.0 10-5 mol L-¹ with a detection limit of 7.0 10-8 mol L-¹. The proposed procedure is successfully applied to the determination of tryptophan in a commercial amino acid oral solution.
Asymptomatic carriage of Clostridioides difficile in pet shop puppies

Crowded pet shops can become infected with various pathogens. Although dog carriers of Clostridioides difficile tend to be asymptomatic, the toxigenic strains sometimes cause gastrointestinal symptoms in humans. The present study was conducted to determine the infection rate of C. difficile in pet shops. The feces of 137 dogs younger than one year of age, collected from four pet shops, were assessed by culture and PCR. Carriage rates of C. difficile in dogs aged ≤90, 91-180, and ≥181 day-old were 12.2% (10/82, 95% confidence interval: 6.0-21.3%), 0.0% (0/42, 95% confidence interval: 0.0-8.4%), and 7.7% (1/13, 95% confidence interval: 0.2-36.0%), respectively. Five C. difficile isolates carried genes encoding toxins A and B, while the remaining isolates contained only toxin B. However, none of the C. difficile carriers had diarrhea. These results highlight the necessity of promoting adequate hygiene precautions in pet shops to prevent infection by C. difficile.

Tungsten particles have long been used as microcarriers in biolistic bombardment because of their cost-effectiveness compared to alternative gold particles— even if the former have several drawbacks, including their DNA-degrading activity. We characterized tungsteninduced DNA degradation to assess the value of this metal particle and to improve tungsten-based biolistic bombardment. Alkaline pH, low temperature, and high salt concentration were found to diminish tungsten-induced DNA breakdown. The pH was the most influential factor in this phenomenon, both in aqueous solutions and on the particles. Furthermore, alkaline pH greater than 9.4 of an adsorption mixture was found to be essential for DNA binding to metal particles. Based on these findings, we propose a new formula of DNA/tungsten adsorption by using TE buffers that keep alkaline pH (>9.4) of the mixture, in which tungsten-bound plasmid DNA cleavage was suppressed to half the level of that in the conventional DNA-binding condition.

This study examines Japanese students’ awareness regarding critical thinking (CT) attitudes and their attention to building their speaking skills in an EFL context to determine effective CT approaches. The study began with a literature review of CiNii articles and studies from the Web of Science database regarding CT theories and educational approaches. It examined critical thinking approaches in EFL contexts as well as foreign-language approaches of International Baccalaureate® (IB) programmes. The review indicated that critical thinking can be fostered when students are asked questions that require higher-order thinking skills (HOTS) in EFL courses. Then, EFL lessons were organised based on the theories and approaches examined in the review. Participants included 22 Japanese senior high school students. Various higher cognitive level questions were posed to the participants. Additionally, students were required to prepare and present academic presentations and answer discussion questions after the presentation. To ascertain students’ awareness of attitudes toward CT, the Critical Thinking Attitudes Scale was used, and students’ reflective essays were analysed using the co-occurrence network of words in the KH Coder. To measure students’ speaking skills, their performance on speaking tasks was analysed by two International Baccalaureate’s examiners. The results indicate that the improvement of CT attitudes and speaking skills is significant regardless of English proficiency level. These findings suggest that if EFL teachers ask HOTS questions, students would become aware of critical thinking attitudes and acquire English speaking skills by answering the questions in a Japanese EFL context.
A Generation Method of Maximal-Period Sequences Obtained by Discretized r-adic Maps

An algorithm of generation of maximal-period chaotic sequences obtained from the one-to-one quantized (discretized) r-adic maps on finite phase space is proposed. This algorithm is drived based on the relationship between such discretized maps and graph theory. The long period sequences can be achieved from shorter period sequences by using the proposed algorithm recursively.

Yield and grade are the key factors that affect production value of peanut. The objective of this study was to identify QTLs for pod yield, hundred-seed weight, and total sound mature kernel (TSMK). A total of 90 recombinant inbred lines, derived from Tamrun OL07 and a breeding line Tx964117, were used as a mapping population and planted in Brownfield and Stephenville, Texas. A genetic map was developed using 1,211 SNP markers based on double digest restriction-site associated DNA sequencing (ddRAD-seq). A total of 10 QTLs were identified above the permutation threshold, three for yield, three for hundred-seed weight and four for TSMK, with LOD score values of 3.7 - 6.9 and phenotypic variance explained of 12.2% - 35.9%. Among those, there were several QTLs that were detected in more than one field experiment. The commonly detected QTLs in this study may be used as potential targets for future breeding program to incorporate yield and grade related traits through molecular breeding.
Promoting Critical Thinking Skills in an Online EFL Environment

This study examined the effects of an approach that promotes EFL learners’ critical thinking in an online EFL setting. Although recent studies have examined approaches to foster EFL learners’ critical thinking, studies conducted in online settings are under-development. The participants were 31 Japanese undergraduate students enrolled in an online EFL course; their degree of critical thinking and resistance towards critical thinking were examined. First, the students’ sense of classroom community was measured using the Rovai’s Classroom Community Scale. Second, the frequency of using critical thinking in their English writing was observed adopting with Stapleton’s rubric, and the resistance towards learning critical thinking was measured through a questionnaire. The results indicated that students could feel a sense of classroom community in online lessons as well as face-to-face settings, and improvements in critical thinking were found in students’ writing regardless of the differences in their English proficiency level. A resistance towards tackling questions that required higher-order thinking was observed among low English proficiency level students compared to a face-to-face classroom setting. This study suggests that an online EFL course aiming to promote critical thinking reduce students’ resistance offering both informal and formal interactive opportunities to answer questions involving higher-order thinking skills.

The objective of this randomized, double-blind,placebo-controlled, crossover study was to examine theeffects of onion extract containing concentrated cysteinesulfoxides on improving sleep quality. In total, 30 healthymen and women who were dissatisfied with their sleepconsumed the test food sample (onion extract tablets) for5 days. The delta power during non-rapid eye movementsleep and the delta power per minute in the first sleep cycleincreased significantly with the intake of onion extractcontaining concentrated cysteine sulfoxides compared withthe intake of placebo. A significant decline in sleep latencywas also observed. Salivary a-amylase level, a stressindicator, decreased significantly with the intake of onionextract containing concentrated cysteine sulfoxides comparedwith the intake of placebo. These findings indicatethat onion extract containing concentrated cysteine sulfoxidesalleviates stress, improves the quality of sleep andpromotes smooth transition into sleep.
