In vitro Development of Interspecies Somatic Cell Nuclear Transfer Embryos Derived from Murine Embryonic Fibroblasts and Bovine Oocytes

        Yun, J.I.,Koo, B.S.,Yun, S.W.,Lee, Chang-Kyu Asian Australasian Association of Animal Productio 2008 Animal Bioscience Vol.21 No.11

Interspecies somatic cell nuclear transfer (iSCNT) is a useful method to preserve endangered species and to study the reprogramming event of a nuclear donor cell by the oocyte. Although several studies of iSCNT using murine cells and bovine oocytes have been reported, the development of murine-bovine iSCNT embryos beyond the 8-cell stage has not been successful. In this paper, we examined the developmental potential of embryos reconstructed with a murine embryonic fibroblast as the nuclear donor and a bovine oocyte as the cytoplasm recipient. The reconstructed embryos were cultured in CZB (murine medium) or CR1aa (bovine medium). In addition, for the development of a murine-bovine iSCNT blastocyst, the antioxidant ${\beta}$-mercaptoethanol (${\beta}ME$) was supplemented to CR1aa medium. Furthermore, to verify the mouse genome activation in murine-bovine iSCNT embryos, RT-PCR analysis of murine Xist was performed. The development of the murine-bovine iSCNT embryos cultured in CR1aa was significantly higher than that in CZB (p<0.05). With respect to the effect of BME on the development of the murine-bovine iSCNT blastocyst, addition of BME produced a significant increase in blastocyst development (p<0.05). Karyotype analysis confirmed that the reconstructed embryos were derived from murine cells (40XX). The Xist gene was gradually increased from the 8-cell stage to the blastocyst stage. This is the first report of blastocyst development of iSCNT embryos derived from murine somatic cells and bovine oocytes. These results demonstrate that bovine cytoplasm can support the development of later stages of a preimplantation embryo from murine-bovine iSCNT.

Management of Pulmonary Hypertension Due to Brachycephalic Obstructive Airway Syndrome in a Dog

        Yunji Song,Yeji Kim,Ji-Hyun Kim,Kwon-Neung Kim,Songju Oh,김하정 한국임상수의학회 2022 한국임상수의학회지 Vol.39 No.5

A 15-year-old, neutered male, Shih-Tzu, was presented at the Chon- nam National Veterinary Medical Teaching Hospital for evaluation of acute onset of persistent coughing, exercise intolerance, and abnormal heart sound. On tho- racic auscultation, a split-second heart sound and a wheezing sound were de- tected on both sides of the chest walls. On physical examination, the dog’s body condition score (BCS) was 7/9, and had stenotic nares. Thoracic radiographs revealed right-sided enlargement of the cardiac silhouette (vertebral heart score (VHS) 11.2; reference interval = 8.9-10.1), mild main pulmonary artery (MPA) bulging, mild interstitial infiltration, and hepatomegaly. The electrocardiogram showed right axis deviation, suggesting right ventricular hypertrophy. The echo- cardiographic study showed moderate pulmonary hypertension and moderate tricuspid regurgitation. There were no findings of a tracheobronchial disease, pulmonary thromboembolism, congenital shunt, left heart disease, or parasitic disease. Based on clinical signs and diagnostic findings, the dog was diagnosed with pulmonary hypertension secondary to brachycephalic syndrome. To rectify respiratory exacerbating factors, the dog was recommended weight control by restricting dietary intake and managing concurrent Cushing’s syndrome. Treat- ments included sildenafil, pimobendan, furosemide, and ramipril. After five months of taking medications and weight control, the severity of pulmonary hypertension improved from moderate to mild. The clinical signs of the patient, including coughing and exercise intolerance, improved a lot. For 5 months of fol- low-up, the patient has not reported further recurrence of respiratory distress.

Wind pressure characteristics of a low-rise building with various openings on a roof corner

Yunjie Wang,Q.S Li 한국풍공학회 2015 Wind and Structures, An International Journal (WAS Vol.21 No.1

Wind tunnel testing of a low-rise building with openings (holes) of different sizes and shapes on a roof corner is conducted to measure the internal and external pressures from the building model. Detailed analysis of the testing data is carried out to investigate the characteristics of the internal and external pressures of the building with different openings\' configurations. Superimposition of the internal and external pressures makes the emergence of positive net pressures on the roof. The internal pressures demonstrate an overall uniform distribution. The probability density function (PDF) of the internal pressures is close to the Gaussian distribution. Compared with the PDF of the external pressures, the non-Gaussian characteristics of the net pressures weakened. The internal pressures exhibit strong correlation in frequency domain. There appear two humps in the spectra of the internal pressures, which correspond to the Helmholtz frequency and vortex shedding frequency, respectively. But, the peak for the vortex shedding frequency is offset for the net pressures. Furthermore, the internal pressure characteristics indirectly reflect that the length of the front edge enhances the development of the conical vortices. The objective of this study aims to further understanding of the characteristics of internal, external and net pressures for low-rise buildings in an effort to reduce wind damages to residential buildings

      • Effects of intervention for postural balance and mobility in age-related macular degeneration: A systematic review

        Yunji Lee,Sun-Kyung Hwang,Min-Eun Cho,Bokyoung Kim,Yea-In Yoon 한국간호과학회 2021 한국간호과학회 학술대회 Vol.2021 No.10

Aim(s): Age-related macular degeneration (AMD) leads to difficulty in mobility and risk of falls due to decreased balance and distance sensitivity through impaired central vision. This review aimed to examine the effectiveness of interventions to improve the direct acuity and adaptability of postural balance and mobility in AMD. Method(s): We searched the literature published until May 20, 2021, in Medline, PubMed, EMBASE, CINAHL, and Cochrane databases for systematic reviews conducted using the Cochrane Handbook’s guidelines (PROSPERO registration: CRD42021200271). Eligible articles included studies in adults with vision loss due to AMD and were designed for randomization or non-control trials to improve postural balance and mobility. Studies including children or evaluating the effects of pharmacological and surgical treatments were excluded. Two authors assessed the risk of bias and quality using the Risk Of Bias In Non-randomized Studies-of Intervention (ROBINS-I) tool and the revised Cochrane Risk –of -Bias (RoB2) tool for randomized trials. Result(s): The extracted seven studies, published in 2007 or later, included 761 participants. The interventions applied in seven studies were as follows: multidisciplinary rehabilitation program (3), device application (2), training using the platform (1), and dancing program (1). Most studies (6/7) measured the outcomes as a subcategory of vision-related quality of life, and two studies directly used the balance data from the platform and Dynamic Gait Index. Although insufficient reporting of intervention content was observed, all studies showed significant improvements in outcomes. Conclusion(s): Rehabilitation programs, including mobility training, improved postural balance and vision-related quality of life in AMD. Nurses should monitor the changes in balance and mobility due to vision loss and consider the methods for improving them to increase the quality of life and safety in older adults.

Development of Tissue Equivalent Materials for a Multi-modality (CT&MRI) Phantom in MRI-guided Radiation Treatment

        Yunji Seol,Jina Kim,Aeran Kim,Jinho Hwang,Taegeon Oh,Jin-sol Shin,Hong Seok Jang,Byung Ock Choi,Young-nam Kang 한국물리학회 2018 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.73 No.7

This study proposed to develop a phantom material that can represent the various contrasts of both MRI and CT images and is available to use in MRI-guided radiation treatment. Materials used for making a phantom that can be used for both MRI and CT image were agarose (T2 modifier), gadolinium-based contrast agent (T1 modifier), sodium uoride (CT number modifier), and distilled water. They were mixed at various composition ratios and stirred until transparent. For the relationship between the ingredients and values, 48 samples were manufactured at various composition ratios. The relationship was expressed as equations, to be able to get the composition ratios of organs that we wanted to make. MR relaxation times were measured using 1.5 T MRI equipment. CT scans were performed at 120 kVp and extracted CT numbers from images. Based on the fitted equations derived from the relationship between ingredients and values, materials were manufactured using the composition ratio of human organs; brain (white and gray matter), liver, spleen, kidney, and prostate. The all values were within the reference range, but some exceeded the range due to the image noise. A phantom composed of substitutes made from the derived equations added other substances of different density like bone or lung can be used as an inhomogeneity dose calculation phantom for both CT and MRI. Furthermore, it can be applied to MRI-only based RTP systems and MRI-guided radiation treatment QA in the future.

An image thresholding method based on the target extraction

        Yunjie Zhang,Yi Li,Zhijun Gao,Weina Wang 한국전산응용수학회 2008 Journal of applied mathematics & informatics Vol.26 No.3-4

In this paper an algorithm, based on extracting a certain target of an image, is proposed that is capable of performing bilevel threshold- ing of image with multimodal distribution. Each pixel in the image has a membership value which is used to denote the characteristic relationship between the pixel and its belonging region (i.e. the object or background). Using the membership values of image set, a new measurement, which simultaneously measures the measure of fuzziness and the conditional en- tropy of the image, is calculated. Then, thresholds are found by optimally minimizing calculated measurement. In addition, a fuzzy range is defined to improve the threshold values. The experimental results demonstrate that the proposed approach can select the thresholds automatically and ef- fectively extract the meaningful target from the input image. The resulting image can preserve the object region we target very well.
