A novel DNN tracking algorithm for structural system identification

In the field of structural health monitoring (SHM), cameras record videos and tracking methods can be applied to calculate the structural displacement. Commercial and unmanned aerial vehicle (UAV) cameras are promising non-contact sensors owning to their high availability and easy installation. However, effective tracking methods need to be developed. In this study, we firstly propose an end-to-end vision measuring framework with a novel deep neural network (DNN) tracker, named Siamese Single Decoder Network (SiamSDN). The system requires no target installation and uses cellphone cameras. For SiamSDN, the position and scale of bounding box are formulated through statistical parameter estimation. Unlike generative trackers, SiamSDN does not require manually extracted features or pre-defined motion areas. The tracking object is solely identified in the first frame. A shaking table test of a five-storey structure is carried out to demonstrate the efficiency. Besides, a UAV is used to simulate the field test. To minimize the error caused by the vibrations of UAV, digital video stabilization (DVS) is proposed to eliminate the drifts. Videos taken by both the commercial and UAV cameras are analyzed to calculate the displacements. Comparing our DNN tracker with feature point matching approach, SiamSDN improves the displacement measuring accuracy by 66.16% and 57.54%, respectively, and the frequency characteristics are obtained precisely.

The limitations in current technology for generating transgenic animals, such as the time and the expense, hampered its extensive use in recombinant protein production for therapeutic purpose. In this report, we present a simple and less expensive alternative by directly infusing a recombinant adenovirus vector carrying human lactoferrin cDNA into rabbit mammary glands. The milk serum was collected from the infected mammary gland 48 h post-infection and subjected to a 10% SDS-PAGE and Western blotting. An 80-kDa protein was visualized after viral vector infection. With this method, we obtained a high level of expressed human lactoferrin of up to 2.3 mg/ml in the milk. Taken together, the method is useful for the transient high-level expression recombinant proteins, and the approach established here is probably one of the most economical and efficient ways for large-scale production of recombinant proteins of biopharmaceutical interest.

A combination of electrospray ionization and tandem mass spectrometry was used to characterize the scavenging reactions of acrylamide (AA) in the presence of glutathione (GSH) in vitro. In the presence of GSH, AA was deactivated effectively and scavenged by reactions consuming small amount of GSH. Reaction products and structural information were identified using collision-induced dissociation (CID) in an ion trap mass spectrometer. In the mixture of GSH and AA, significant increase in abundance of fragment ion peak was observed at m/z 233, which was identified as $[Cys-Glu]^+$, formed by the elimination of glycine moiety of GSH. GSH also contributes to the AA scavenging reaction by conjugating with AA through the sulfhydryl group in cysteine moiety. The probable scavenging reaction pathway of AA in the presence of GSH has been proposed based on the CID experimental data.

In this paper an automatic optical inspection system for the advanced fiber coupler assembly manufacturing process is presented. Coupling efficiency variations were compared with axial displacement. The main standard components of light peak fiber couplerswere used in the experiment to determine the corresponding offset and position of the optimal optical coupling efficiency in order toachieve a positioning effect. This study used the parameters of lens curvature, focal length and light source for validation using opticaldesign software. An infrared CCD camera was used to extract the coupled light spot images. The curve fitting method and couplingsimulation database were used for image processing and to calculate optimal coupling efficiency. Under optimal coupling efficiency,the minimum amount of movement of the translation stages was used to calculate the offset of each axle or the coupling efficiencywhen tilted. Moreover, a coupling efficiency database was established. The optical axle center offset in the alignment was calculatedby comparing the data in the database. Finally, the optical axle center offset or tilt direction was identified through the program toachieve the position of optimal coupling efficiency.

Purpose: Human papillomavirus (HPV) infection plays an important role in the development of cervical cancer, but the prevalence of HPV infection in women of Shenzhen city remains unclear. The present study was performed to describe the change of cervical HPV infection in females who participated in voluntary cervical cancer screening from 2006 to 2010 in Shenzhen city, China. Methods: A total of 4, 413 women were recruited. HPV infections were genotyped by polymerase chain reaction (PCR) and reversed dot blot hybridization in Shenzhen Maternity and Child Health Hospital. Results: The prevalence of HPV infection was 13.8%. The five most commonly found HPV types were HPV16 (3.47%), HPV58 (1.68%), HPV33 (1.38%), HPV43 (1.36%) and HPV18 (1.27%). The secular trends of major HPV type-specific were diverse. Among of them, the prevalence of HPV18 increased sharply while others increased slowly or even decreased in the period. The change of total HPV, single HPV and multiple HPV infection were similar during the five years. Conclusions: Our findings suggested that HPV infection is common with HPV16 and HPV 58 as the primary subtypes in women in Shenzhen city.The prevalence of HPV 18 infection is increasing faster than any others, which will lead it to be one of the main subtypes in this city in the future.

Objective: Patients with opioid use disorder (OUD) have impaired attention, inhibition control, and memory function. The aldehyde dehydrogenase 2 (ALDH2 ) gene has been associated with OUD and ALDH2 gene polymorphisms may affect aldehyde metabolism and cognitive function in other substance use disorder. Therefore, we aimed to investigate whether ALDH2 genotypes have significant effects on neuropsychological functions in OUD patients undergoing methadone maintenance therapy (MMT). Methods: OUD patients undergoing MMT were investigated and followed-up for 12 weeks. ALDH2 gene polymorphisms were genotyped. Connors’ Continuous Performance Test (CPT) and the Wechsler Memory Scale-Revised (WMS-R) were administered at baseline and after 12 weeks of MMT. Multivariate linear regressions and generalized estimating equations (GEEs) were used to examine the correlation between the ALDH2 genotypes and performance on the CPTs and WMS-R. Results: We enrolled 86 patients at baseline; 61 patients completed the end-of-study assessments. The GEE analysis showed that, after the 12 weeks of MMT, OUD patients with the ALDH2 *1/*2+*2/*2 (ALDH2 inactive) genotypes had significantly higher commission error T-scores (p = 0.03), significantly lower hit reaction time T-scores (p = 0.04), and significantly lower WMS-R visual memory index scores (p = 0.03) than did patients with the ALDH2 1 */*1 (ALDH2 active) genotype. Conclusion: OUD patients with the ALDH2 inactive genotypes performed worse in cognitive domains of attention, impulse control, and memory than did those with the ALDH2 active genotype. We conclude that the ALDH2 gene is important in OUD and is associated with neuropsychological performance after MMT.

Melanosis is major problem of crustaceans duringtheir rigor mortis storage. This study for the firsttime was designed to optimize the formula of preservativesto maintain the color feature of Pacific white shrimp usingresponse surface methodology. A three-factors-three-levelsBox-Behnken design was applied to evaluate the effect ofchitosan, citric acid and L-cysteine on color features (L*,a*, b* and DE) of Pacific white shrimp. It was found thatthe increasing rate of DE was retarded by the higher concentrationsof chitosan, citric acid and L-cysteine in acertain range. The optimal formula for inhibiting theincrease of DE was 1.36% chitosan, 0.47% citric acid and0.31% L-cysteine. Under the optimal pretreated conditions,the predicted DE of shrimp after 8 days of storage was14.59, close to the measured values (14.49). These resultsindicated that the optimal combined preservatives couldretard the decrease of lightness and the aggregation of DEand melanosis effectively, and might be a potentialapplication for retarding melanosis and extending shelf lifeof Pacific white shrimp.

A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc: 153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Nicharged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PACPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5α harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5α and control.
Seismic Performance of Kiewitt-Sunflower Single Layer Spherical Reticulated Shells

In this paper, seismic behavior of Kiewitt-Sunflower single layer spherical reticulated shells is examined within the time history method, based on our proposed damage constitutive model for thin-walled circular steel tubes. According to a large number of numerical examples, the failure mode of this kind of single layer spherical reticulated shell in the case of earthquake is figured out. In addition, the effect of the damage accumulation, rise-span ratio, initial geometric imperfection, types of seismic wave and amplitude modulation ratio on the dynamic response of the shell structure is explored. Finally, a structural damage factor is proposed and it has a good match with the numerical results, which is productive to assess the damage and failure behavior of such structures suffering the earthquake and post-earthquake.

This is the first report on using Catathelasma ventricosum for production of fruiting body and lignocellulosic enzymes. To improve the laccase activity and productivity of mushroom, the substrate was added with different supplements (eight aromatic compounds, Mn2+, and Cu2+). Based on the results, all these supplements can improve the laccase activity and productivity of C. ventricosum, and it seems that there is a critical value of laccase activity that affects the productivity of C. ventricosum. In addition, when Penicillium decumbens was inoculated into the substrate that had been cultivated with C. ventricosum for 20 days, the highest values of laccase activity, FPA activity, and productivity of C. ventricosum were obtained. Moreover, the laccase activity showed a positive correlation with the productivity of C. ventricosum. Finally, the effect of Mn2+, Cu2+, and P. decumbens on laccase activity was investigated by response surface methodology (RSM).
