        The complete mitochondrial genome of a threatened loach (Sinibotia reevesae) and its phylogeny

        In present study, the complete mitochondrial genome of Sinibotia reevesae was first sequenced using the next-generation sequencing technology and annotated using bioinformatic tools. The circular mitochondrial genome was 16,572 bp in length, and contained 13 proteincoding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and 1 displacement loop locus. It presents a typical gene organization and order for completely sequenced cypriniformes mitogenomes. The control region could be divided into three parts included the extended termination associated sequence domain, the central conserved domain and the conserved sequence block. Interestingly, two stemloop domains were found in control region and OL region, respectively. Furthermore, phylogenetic analyses using concatenated amino acid and nucleotide sequences of the 13 protein-coding genes with two different methods (Maximum likelihood and Bayesian analysis) both highly supported the close relationship of S. reevesae and Sinibotia superciliaris, which was in line with the previous classifications based on morphological and molecular studies. These data provide useful information for a better understanding of the mitogenomic diversities and evolution in fish as well as novel genetic markers for studying population genetics and species identification.

        Association Study of Polymorphisms in Neuronal Nicotinic Acetylcholine Receptor Subunit Genes With Schizophrenia in the Han Chinese Population

        Objective To investigate the relation between nicotinic acetylcholine receptor subunit (nAChR) genes and schizophrenia, and the relation between tag single nucleotide polymorphism (rs1317286, rs1044396, rs6494212, rs16969968, and rs684513) and schizophrenia in Han Chinese people. Methods The protein-protein interaction (PPI) network among nAChR protein and 350 proteins encoded by schizophrenia-related susceptibility genes was constructed through the String database to explore whether nAChR genes were associated with schizophrenia in these known databases. Then, five single nucleotide polymorphisms (SNPs) of CHRNA3 (rs1317286), CHRNA4 (rs1044396), CHRNA7 (rs6494212), and CHRNA5 (rs16969968, rs684513) were analyzed in a sample of 1,035 schizophrenic patients and 816 healthy controls. The interaction between the markers was analyzed using multifactor dimensionality reduction (MDR) software. Power analysis was performed using the Quanto program. Results There are no significant differences in genotype or allele distribution were identified between the patients and controls (p>0.05). The haplotypes constructed by four markers rs1317286, rs6494212, rs16969968, and rs684513 were not associated with schizophrenia either. However, a significant association between models made of rs1317286, rs1044396, rs6494212, and rs684513 and schizophrenia was revealed in interaction analysis (p<0.05). Conclusion The nAChR protein may have effects on the development of schizophrenia through the interaction with proteins encoded by schizophrenia-related susceptibility genes, but no relation was found between selected polymorphisms and schizophrenia in the collected Han Chinese people. However, interaction analysis suggested four-SNP model has an important effect on schizophrenia.

      • Association Study of Single-Nucleotide Polymorphisms of STAT2/STAT3/IFN-γ Genes in Cervical Cancer in Southern Chinese Han Women

        Objective: Interferon-${\gamma}$ (IFN-${\gamma}$) and signal transducers and activators of transcription (STATs) each play an important role in carcinogenesis associated with viral infection. Cervical cancer is almost invariably associated with infection by human papillomavirus (HPV), and previous studies suggested that dysregulation of the signal pathway involved in IFN-${\gamma}$ and STATs is associated. Our objective was to evaluate the association of SNPs in STAT2, STAT3, and IFN-${\gamma}$ with cervical cancer susceptibility in Chinese Han women in Hunan province. Materials and Methods: Genomic DNA was extracted from peripheral blood samples of 234 cervical cancer patients and 216 healthy female controls. STAT2 and STAT3 genotyping was performed using polymerase chain reaction-restriction enzyme (PCR-RE) analysis. IFN-${\gamma}$ genotyping was detected by PCR-amplification of specific allele (PASA). Results: For STAT2 rs2066807 polymorphisms, there was no significant difference of genotype distribution (P=0.827) and allele frequencies (P=0.830, OR=1.09, 95% CI: 0.51-2.31) between cases and controls. For STAT3 rs957970 polymorphisms, there was no significant difference of genotype distribution (P=0.455) and allele frequencies (P=0.560, OR=0.92, 95% CI: 0.71-1.20) between cases and controls. For IFN-${\gamma}$ +874A/T polymorphisms, there was no significant difference of genotype distribution (P=0.652) and allele frequencies (P=0.527, OR=1.12, 95% CI: 0.79-1.59) between cases and controls. Conclusion: These results suggest that polymorphisms in STAT2, STAT3 and IFN-${\gamma}$ genes are not likely to be strong predictors of cervical cancer in Han women in southern China.

        Optimized Planning Target Volume Margin in Helical Tomotherapy for Prostate Cancer: Is There a Preferred Method?

        We compare the dosimetrical differences between plans generated for helical tomotherapy by using the 2D or 3D the margining technique for the treatment of prostate cancer. Ten prostate cancer patients were included in this study. For 2D plans, the planning target volume (PTV) was created by adding 5 mm (lateral/anterior-posterior) to the clinical target volume (CTV). For 3D plans, a 5-mm margin was added not only lateral/anterior-posterior, but also superior-inferior, to the CTV. Various dosimetrical indices, including the prescription isodose to target volume (PITV) ratio, conformity index (CI), homogeneity index (HI), target coverage index (TCI), modified dose homogeneity index (MHI), conformation number (CN), critical organ scoring index (COSI), and quality factor (QF) were determined to compare the different treatment plans. Differences between the 2D and the 3D PTV indices were not significant except for the CI (p = 0.023). 3D margin plans (11195 MUs) resulted in higher (13.0%) monitor units than 2D margin plans (9728 MUs). There were no significant differences in any organs at risk (OARs) between the 2D and the 3D plans. Overall, the average dose for the 2D plan was slightly lower than that for the 3D plan dose. Compared to the 2D plan, the 3D plan increased the average treatment time by 1.5 minutes; however, this difference was not statistically significant (p = 0.082). We confirmed that the 2D and the 3D margin plans were not significantly different with regard to various dosimetric indices such as the PITV, CI, and HI for PTV and the OARs with tomotherapy

        Fabrication of an antibacterial zinc oxide film on biodegradable magnesium alloy and its biocompatibility

        To avoid the rapid degradation of magnesium alloy AZ31B after implantation in the body, the addition of a zinc oxide (ZnO) film was examined. A rough surface was fabricated by femtosecond laser on AZ31B at frequencies of 100, 200, 300, 500, and 800 kHz, before adding a ZnO film by wet metallurgy. Scanning electron microscopy (SEM) indicated that 500 kHz femtosecond laser frequency produced a coral-like surface morphology on AZ31B and the ZnO film had a needle-like structure. X-ray diffraction (XRD) revealed that the film mainly consisted of Zn and ZnO. Potentiodynamic polarization curves revealed that the ZnO film exhibited a better corrosion resistance than bare AZ31B. It also possessed superior cell proliferation ability and higher suppression of a representative device-related infection of Escherichia coli strain than bare AZ31B. This study provides a novel film strategy for manufacturing biodegradable magnesium alloys with good corrosion resistance, biocompatibility, and antibacterial properties.

        An Efficient Privacy Preserving Scheme for Distributed Data Aggregation in Smart Grid

        Privacy-preserving data aggregation in smart grids has been intensively studied to balance utility and privacy requirements. Existing contributions usually rely on a trusted aggregator to process electric power data and then apply privacy preservation strategies to protect the aggregate data. However, the privacy of individuals will be revealed once the aggregator is hacked or cannot be trusted. By injecting independent Gaussian noise into the response to the query before performing aggregation, a distributed Gaussian mechanism is designed to eliminate the negative effect of the untrusted aggregator. Equipping the distributed Gaussian mechanism with a post-processing step based on the moving average filtering technique, an efficient privacy-preserving scheme is proposed to improve individual electric power data utility. The experiments conducted on the real-world datasets show that the proposed scheme can improve the utility of electric power data while protecting the privacy of individual data.

        In‑situ growth of ZIF‑8/CP with ultra‑high adsorption capacity for removing Malachite green from water

        An environmentally friendly and low-cost chitosan-containing polysaccharide (CP) composite ZIF-8/CP was designed and prepared based on the difficulty of separating the traditional adsorbent from the water phase. ZIF-8/CP was synthesized through in-situ growth approach. The physical, chemical and structure properties of ZIF-8/CP were determined through a series of characterization methods, including SEM, FT-IR and PXRD. The effects of touch time, pH, temperature, and coexisting ions on adsorption were assessed. In addition, kinetics, isotherms of adsorption and thermodynamics were examined. The data of isotherms for adsorption indicated that the adsorption of ZIF-8/CP on MG was similar to the Langmuir model, with a maximum adsorption capacity of 1428.57 mg/g. Moreover, the kinetic parameters were consistent with the pseudo- 2nd-order equation. Thermodynamic studies (ΔG < 0, ΔH > 0) demonstrated a heat-absorbing and spontaneous adsorption process. Our study reveals that ZIF-8/CP has good adsorption properties and environmental properties.

        Coexpression of Kex2 Endoproteinase and Hac1 Transcription Factor to Improve the Secretory Expression of Bovine Lactoferrin in Pichia pastoris

        The large-scale production of functional recombinant lactoferrin has become a major goal because of its medicinal value and global demand. Secreting recombinant proteins into a culture medium offers a way to simplify protein purification and avoid toxicity from intracellularly accumulated materials. In this study, after 84 h of induction with methanol in a shaking flask, the recombinant bovine lactoferrin (rbLf) titer in the culture supernatant of the strain that integrated two copies of the rbLf gene was only 121.6 μg/L. A bottleneck might have existed in the folding and secretion pathways of rbLf. We then attempted to further improve the rbLf titer by overexpressing the transcription factor Hac1p and α-signal peptide-cutting protease Kex2p with different promoters. Results showed that the inducible coexpression of Hac1p and Kex2p linked with the 2A sequence improved the rbLf titer 5.0-fold (735.8 μg/L) after 84 h of induction with methanol. The maximal titer in a shaking flask was 1,150.5 μg/L after 120 h of induction. The rbLf titer achieved 35.6 mg/L in a 5 L fed-batch fermenter. Thus, Kex2 and Hac1 overexpression driven by methanol-induced promoter alleviated the bottleneck in the folding and secretion pathways and greatly improved the secretory expression of rbLf in Pichia pastoris.

        Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

        The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under 27 μmol·m-2·s-1 of light intensity and 25oC on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.
