        Effects of α-Si₃N₄ seeds and sintering additives on properties of porous silicon nitride ceramics fabricated by carbothermal reduction

        In this paper, porous Si3N4 ceramics were fabricated by carbothermal reduction between silicon dioxide and carbon [1]. The influences of α-Si3N4 seeds and sintering additives on the microstructure and mechanical properties of porous Si3N4 ceremics were investigated. XRD analysis proved the complete formation of a single-phase β-Si3N4. SEM analysis showed that the resultant porous Si3N4 ceramics had a fine microstructure and a uniform pore structure. The sintered sample with Lu2O3 as sintering additive showed finer, higher aspect ratio β-Si3N4 grains. The addition of Eu2O3 accelerated the densification of porous Si3N4 ceramics, decreased the porosity and increased the flexural strength. The addition of α-Si3N4 seeds accelerated the formation of the α-Si3N4 phase at a low temperature and the α-β phase transformation process at a high temperature. With an increase in the α-Si3N4 seeds content, the porosity decreased, and the flexural strength increased accordingly.         Maintaining Drosha expression with Cdk5 inhibitors as a potential therapeutic strategy for early intervention after TBI

        Traumatic brain injury (TBI) is a major cause of death and disability in adults. The pathological process of TBI involves a multifactorial cascade in which kinases have been proven contribute to interactions between relevant factors and amplification of signaling cascades. Cyclin-dependent kinase 5 (Cdk5) is a promising kinase that has been implicated in various brain disorders, including TBI. However, the mechanism by which Cdk5 induces neuronal damage remains unclear. Here, we show for the first time that Drosha, a key enzyme in microRNA biogenesis, is a pivotal substrate of abnormally activated Cdk5. Cdk5-mediated phosphorylation decreases Drosha expression and exacerbates nerve injury in TBI. We proved that maintaining Drosha expression via the administration of repurposed Cdk5 inhibitors that were previously studied in clinical trials is a promising approach for the early treatment of TBI. Together, our work identifies Drosha as a novel target for neuroprotective strategies after TBI and suggests Cdk5-mediated regulation of Drosha expression as a potential therapeutic strategy for early TBI intervention.

      • EphB1 and Ephrin-B, New Potential Biomarkers for Squamous Cell/adenosquamous Carcinomas and Adenocarcinomas of the Gallbladder

        Squamous cell/adenosquamous carcinoma (SC/ASC) of the gallbladder are rare tumors and there are few clinical reports in the literature. Herein we report our clinical experience with 46 patients with SC/ASC and 80 with adenocarcinoma (AC). Expression of EphB1 and Ephrin-B in each tumor was determined using immunohistochemical methods for determination of correlations with prognosis. There was no difference in EphB1 and Ephrin-B expression between SC/ASC and AC tumors (P>0.05), but greater expression in those less than 3 cm in diameter, stage I or II (TNM stage), with no lymph node metastases, with no local invasion and treated with radical resection was apparent. Expression of EphB1 (P<0.05) and Ephrin-B (P<0.01) was higher in well differentiated than in poorly differentiated AC tumors. Kaplan-Meier survival analysis indicated that degree of differentiation, tumor diameter, lymph node metastases, local invasion, surgical approach and expression rate of EphB1 and Ephrin-B were closely related to the survival of SC/ASC (P<0.05) and AC patients (P<0.01). Patients with tumors that positive expressed EphB1 and Ephrin-B, whether it is SC/ASC ($P_{SC/ASC}$ =0.000) or AC ($P_{AC}$ =0.000 or $P_{AC}$ =0.002) had longer survival than those negative expression. Cox multivariate analysis indicated a negative correlation between expression of EphB1 or Ephrin-B and overall survival. Hence, EphB1 and Ephrin-B could be regarded as independent good prognostic factorsand important biological markers for SC/ASC and AC of gallbladder.

        Regenerated coenzyme-based preparation of bienzyme-polymer nanoconjugates and their applications for the synthesis of ethyl (R)-2-hydroxy-4-phenylbutyrate

        A modular approach was applied for the synthesis of bienzyme-polymer nanoconjugates (nano-BECs) (50- 70 nm) consisting of two enzymes (carbonyl reductase and glucose dehydrogenase) conjugated within a single universal polymer scaffold. The amount of the product ethyl (R)-2-hydroxy-4-phenylbutyrate (R-HPBE) with nano-BECs as the catalyst was 533mM in a dibutyl phthalate-phosphate buffer (dibutyl phthalate-PB) biphasic system, while the amount of R-HPBE was 349mM using carbonyl reductase-poly(acrylic acid) as the catalyst, indicating that the nano- BECs have an advantage for coenzyme regeneration. Compared with a single aqueous phase, the substrate treatment capacity was improved at the interface of the dibutyl phthalate-PB biphasic system. Under the optimal reaction conditions (35 oC, 40 h, dibutyl phthalate-PB 1 : 1), nano-BECs can completely convert substrate into optically pure R-HPBE (enantiomeric excess (e.e.) >99.9%) in the organic-aqueous system.

        Long Non-coding RNA CASC15 Promotes Intrahepatic Cholangiocarcinoma Possibly through Inducing PRDX2/PI3K/AKT Axis

        Purpose Intrahepatic cholangiocarcinoma (ICC) is one of the most common liver primary tumors but its treatments are limited. Bioinformatics showed that the expression level of long non-coding RNA cancer-associated susceptibility 15 gene (CASC15) is correlated with ICC progression, but its functional mechanism remains unclear. Materials and Methods Tissues from ICC patients, tumor and adjacent tissue, were used for detection of the expression of CASC15. Clinical data were also collected for clinicopathologic and survival analysis. Short interfering RNA and lentiviral short hairpin RNA were used to knock down CASC15 and PRDX2 expression in ICC cell lines, for the analysis of changes of cell function and xenografts. RNA-pulldown and RNA immunoprecipitation assays were used to detect RNA-binding protein, PRDX2. Male nude mice were used for ICC xenografts, and livers were collected after 4 weeks for immunohistochemistry. Results CASC15 is highly expressed in ICC tissues and is related to higher TNM stage. Knockdown of CASC15 in ICC cells reduced cell proliferation, migration, invasiveness and increased apoptosis, and G1/S block. PRDX2 bound to CASC15. Knockdown of CASC15 decreased PRDX2 expression which was rescued by the inhibition of proteasome formation. Downregulation of PRDX2 resulted in G1/S block, reduced ICC cell invasion. Downregulation of CASC15 inhibited phosphoinositide 3-kinase (PI3K)/AKT/c-Myc pathway through downregulating of PRDX2 and overexpressed PRDX2 rescued the block. CASC15 knockout in ICC xenografts suppressed tumor development in vivo, decreased the expression of PRDX2 and Ki67 and inhibited PI3K/AKT pathway. Conclusion CASC15 promotes ICC possibly by targeting PRDX2 via the PI3K/AKT pathway, indicating poor prognosis and high degree of malignancy of ICC.

        Identification and characterization of CONSTANS-like genes from Curcuma alismatifolia

        The CONSTANS ( CO ) and CONSTANS - like ( COL ) genes have an important role in the regulation of fl owering in photoperiod-sensitive plants. However, information on the molecular characterization of COL genes in Curcuma alismatifoliais very limited. The objective of this study was to identify and evaluate 8 COL homologs ( CaCOLs ) in the C. alismatifoliatranscriptome. Their structures, phylogenetic relationships and expressing patterns under short-day (SD) and long-day (LD)contexts were investigated. Our results showed that CaCOLs were classifi ed into three groups: CaCOL3 and CaCOL6 in groupI were the most CO - like genes; CaCOL7 and CaCOL8 in group II contains only one B-box; CaCOL1 to 2 and CaCOL4 to 5formed the group III. Phylogenetic analysis of the CaCOLs in Arabidopsis, Rice, Barley, Malaccensis and C. alismatifoliademonstrated that both the B-box and CCT domains were conserved in dicot and monocot plants, but diff erent groups ofCOL genes evolved independently. Diff erent patterns of mRNA accumulation in C. alismatifolia leaves in response to SDand LD treatments were observed. CaCOL1 , CaCOL2 , CaCOL4 and CaCOL7 showed signifi cantly higher expressions inLD treatment than that of SD, indicating that they were potential fl owering inducers, and are candidate genes for research infl owering regulation and circadian controlling in C. alismatifolia plants. Altogether, our study facilitates successful regulationof fl owering in Curcuma species and provides insights for future molecular breeding programs.

        An Iterative Calculation Method for Internal Forces and Deformation of Curved Tunnel Lining

        The accurate prediction of lining forces and deformations is crucial for the design of arched tunnel lining. However, in traditional tunnel structural mechanics methods, the distribution shape of the surrounding rock elastic resistance (SRER), as well as the zero point and maximal point locations of SRER, are not clearly defined. In this paper, an iterative calculation method is proposed based on the gradual convergence of the unitized values of SRER and total displacement (TD) of the lining, and the one-to-one functional relationship between the two in the stability process of the surrounding rock and lining. In this method involves using the initial displacement caused by the pressure of the surrounding rock pressure is used to initiate the iterative process, and iterative calculation is carried out until the errors between the unitized value of SRER and the unitized value of TD of the lining structure meet the error requirements. This enables precise determination of the SRER and TD. In order to verify the effectiveness of the proposed method, a case study is conducted on the tunnel of Lianghe Expressway tunnel in Yunnan province, China . The results showed that when the semi-lining structure is divided into unit blocks and the average error of consistency is required to be less than 1e-3, it takes about 20 iterations are needed to meet the error requirement. Furthermore, the accuracy of the calculated results is verified through site-measuring experiments. This method provides an effective tool for failure analysis of arched tunnel lining structures.
