        Administration of vitamin D3 by injection or drinking water alters serum 25-hydroxycholecalciferol concentrations of nursery pigs

        Objective: Two experiments were conducted to evaluate vitamin D3 administration to nursery pigs by injection or in drinking water on serum 25-hydroxycholecalciferol (25-OHD3) concentrations. Methods: At weaning, 51 pigs (27 and 24 pigs in experiments 1 and 2, respectively) were allotted to vitamin D3 treatments. Treatments in experiment 1 were: i) control (CON), no vitamin administration beyond that in the diet, ii) intramuscular (IM) injection of 40,000 IU of vitamin D3 at weaning, and iii) water administration, 5,493 IU of vitamin D3/L drinking water for 14 d postweaning. Treatments in experiment 2 were: i) control (CON), no vitamin administration, and ii) water administration, 92 IU of d-α-tocopherol and 5,493 IU of vitamin D3/L drinking water for 28 d postweaning. The lightest 2 pigs within each pen were IM injected with an additional 1,000 IU of d-α-tocopherol, 100,000 IU of retinyl palmitate, and 100,000 IU of vitamin D3. Results: In both experiments, serum 25-OHD3 was changed after vitamin D3 administration (p<0.05). In experiment 1, injection and water groups had greater values than CON group through d 35 and 21 post-administration, respectively (p<0.05). In experiment 2, serum values peaked at d 3 post-administration in the injection groups regardless of water treatments (p<0.05) whereas CON and water-only groups had peaks at d 14 and 28 post-administration, respectively (p<0.05). Even though the injection groups had greater serum 25-OHD3 concentrations than the non-injection groups through d 7 post-administration regardless of water treatments (p<0.05), the water-only group had greater values than the injection-only group from d 21 post-administration onward (p<0.05). Conclusion: Serum 25-OHD3 concentrations in pigs increased either by vitamin D3 injection or drinking water administration. Although a single vitamin D3 injection enhanced serum 25-OHD3 concentrations greater than water administration in the initial period post-administration, a continuous supply of vitamin D3 via drinking water could maintain higher serum values than the single injection.

      • A 3D human neural cell culture system for modeling Alzheimer's disease

        Stem cell technologies have facilitated the development of human cellular disease models that can be used to study pathogenesis and test therapeutic candidates. These models hold promise for complex neurological diseases such as Alzheimer's disease (AD), because existing animal models have been unable to fully recapitulate all aspects of pathology. We recently reported the characterization of a novel 3D culture system that exhibits key events in AD pathogenesis, including extracellular aggregation of amyloid-β (Aβ) and accumulation of hyperphosphorylated tau. Here we provide instructions for the generation and analysis of 3D human neural cell cultures, including the production of genetically modified human neural progenitor cells (hNPCs) with familial AD mutations, the differentiation of the hNPCs in a 3D matrix and the analysis of AD pathogenesis. The 3D culture generation takes 1–2 d. The aggregation of Aβ is observed after 6 weeks of differentiation, followed by robust tau pathology after 10–14 weeks.

        성인의 급성 심장사에서 정상 심장례의 분석

        The authors analysed 53 cases of sudden cardiac death(SCD) without prominent structural abnormalies of heart out of witnessed 300 cases. Thirty four cases were associated with sleep and 19 with activity or rest. Twenty one cases were classified as having normal hearts, and the rest as structurally mild abnormal ones. Male was predominant, and almost of the victims were suffered in their twenties or thirties. All cases collapsed instantaneously or in very short time with various and not specific terminal symptoms. Inducing or trigger factors are not clarified in almost of the cases. Mechanisms or causes of death could not be defined clearly. We concluded that careful heart examination with detailed clinical history under the appropriate postmortem investigation system is only effective approach for the true mechanism and cause of death.

        <P><B>Abstract</B></P> <P>Two series of aromatic poly(<I>ortho</I>-hydroxyamide)s (poly(<I>o</I>-hydroxyamide)s, HPAs) were prepared by reaction of two diamines, 2,2-bis(3-amino-4-hydroxyphenyl) propane (APA) and 2,2-bis(3-amino-4-hydroxyphenyl) hexafluoropropane (APAF), with four aromatic diacid chlorides; terephthaloyl dichloride (TPC), isophthaloyl dichloride (IPC), 2,2-bis[4-chlorocarbonylphenyl)hexafluoropropane (6FC) and 4,4′-sulfonyldibenzoyl dichloride (DBSC). Amorphous HPAs with high molecular weights (inherent viscosities higher than 0.5 dL/g) and relatively high glass transition temperatures (220–280 °C) were obtained. Dense membranes of HPAs were able to undergo a thermal rearrangement (TR) process to polybenzoxazoles (β-TR-PBOs) heating at moderate temperatures (between 250 and 375 °C), and their complete conversion was reached at a temperature below 375 °C, depending on the <I>o</I>-hydroxy diamine moiety, APA and APAF. The β-TR-PBOs films derived from APAF showed a higher thermal stability and higher Tg than those from APA. Gas separation properties of TR-PBOs membranes were superior to those of their poly(<I>o</I>-hydroxyamide) precursors, particularly for the following gas pairs: O<SUB>2</SUB>/N<SUB>2</SUB>, CO<SUB>2</SUB>/CH<SUB>4</SUB>, He/CH<SUB>4</SUB> and He/CO<SUB>2</SUB>.</P>

        Optical Properties Related to Two-Dimensional Electron Gas at AlGaN/GaN Heterostructures

        We report photoluminescence (PL) spectra related to a two-dimensional electron gas (2DEG) conned at an AlGaN/GaN heterointerface. The 2DEG related the PL has been identied by measuring PL of the structure as functions of the light intensity and the temperature and by comparing the results with those obtained for GaN exposed via etching. The PL peak related to recombination between the two-dimensional electron gas and photoexcited holes is located at 3.436 eV at 15 K, which is 28 meV below the bound exciton (D0X) emission in GaN. The activation energy is about 10.8 meV.

        Amplicilin biosynthesis by immobilized enzyme

        Ampliciline was synthesized from 6-amino-pencillanic acid (6-APA) and D-.alpha. phenylglycine methyl ester by using amplicilin synthesizing enzyme from Peudomonas melanogenum (IAM 1655). The whole cell enzyme was immobilized by entrapping it in the polyacrylamide gel lattices. The polymer used in the enzyme entrapment was made from 150 mg per ml of acrylamide monomer and 8 mg per ml of N, N'-methylenebisacrylamide. About 200 mg/whole cell enzyme was mixed in the polymer for entrapment. The maximal activity retention after immobilization was 56%. The optimal pH values for the whole cell enzyme and the immobilized whole cell enzyme were 6.0 and 5.9, respectively. The optimal temperature for the enzyme activity were the same for both type of preparations. The enzyme stabilities against pH and heat increased for immobilized whole cell enzyme. Immobilized cell was more stable especially in the acidic condition while both type were found to be very suceptible to thermal inactivation at a temperature above 4.deg.C. The kinetic constants obtained from Lineweaver-Burk plot based on two substate reaction mechanism showed somewhat higher value for immobilized whole cell enzyme as compared to the whole cell enzyme : the Km value for 6-APA were 7.0 mM and 12.5 mM while Km values for phenylglycine methyl ester were 4.5 mM and 8.2 mM, respectively. Using the immobilized whole cell enzyme packed in a column reactor, the productivity of ampiciline was studied by varying the flow rate of substrate solution. At the space velocity, SV, 0.14 hr$^{-1}$ the conversion was 45%. Operational stability found in terms of half life was 30 hr at SV = 0.2 hr.

        Use of the Centroid Method to Estimate Volumes of Japanese Red Cedar Trees in Southern Korea

        Cubic-meter volumes estimated from two proxy taper functions were compared to observed volumes of Japanese red cedar trees (Cryptomeria japonica D. Don) to evaluate accuracy and precision in the centroid method. Centroid volume estimates were also compared to volume estimates from existing whole-tree volume equations developed for another geographic region. This study found that one proxy function produced unbiased volume estimates while the other was biased. Volume estimates from the whole-tree equations were also biased. However, the volume estimates from the whole-tree equations were more precise than those from the centroid method. These results support previous studies that the centroid method can produce reliable volumes of trees when no other reliable volume equations exist.

        Hemophagocytic Lymphohistiocytosis after Lung Transplantation

        Hemophagocytic lymphohistiocytosis (HLH) is a rare but fatal complication after solid organ transplantation. Acquired forms of HLH are described in association with severe sepsis, autoimmune disorders, malignancy, immune-compromised states, infections, and solid organ transplantation. We experienced a case of hemophagocytic lymphohistiocytosis after bilateral lung transplantation. Leukopenia,thrombocytopenia, and hyperbilirubinemia were noted and became aggravated 50 days after transplantation. Diagnosis of HLH was based on clinical and laboratory findings of splenomegaly, cytopenia, elevated ferritin, elevated interleukin-2 receptor, and hemophagocytosis in bone marrow. Other features such as elevated bilirubin, lactate dehydrogenase, and D-dimer which can be present in HLH were also noted. The patient was immediately treated with etoposide and dexamethasone. Despite aggressive therapy, the patient deteriorated and died. Awareness of the diagnostic criteria of HLH after lung transplantation is important for clinicians.

        '금정굴' 유골에 대한 남녀 구분을 위한 형태학적 조사와 유전자 검사

        At "Keumjung-gul" site, bones of more than 153 persons are excavated in October 1995. They were interred in Autumn of 1950 during Koean War. Among them, 70 skulls were examined in morphological method and DNA test for sex determination. With PCR amplification of amelogenin, 60 skulls were proved male, 9; female, 1; not amplified. Result with Y27H39LR, SRY and alphoid repeats were not satisfactory. The morphologic determination for sex were made maily by (1) prominence of the supercilliary arch,(2) prominence of the external occipital protuberance and crest, (3) size and shape of the mastoid process. The concurrence rate between two examiner, and between two morphologic result and DNA result were low (〈70%), largely because of the subjectivity of morphologic trait and the paucity of experiences.
