        TPA로 야기된 HL-60 세포의 기질부착에 대한 Asadisulphide의 억제효과

        항암제로 사용되어 온 아위를 사용하여 극성에 따른 용매추출 분획을 얻고 TPA로 처리된 HL-60 세포를 사용하여 기질부착 억제실험을 수행한 결과, ethyl acetate (EA)층에서 가장 강한 부착억제효과가 있음을 알아 내었기에 ethyl acetate (EA)층을 다시 ethyl acetate (EA), hexane(HEX), ethyl ether (EE)로 추출한 뒤 3회 chromatography하여 아위 순수활성물질인 asadisulphide를 분리해 내었다. 이 순수활성물질을 가지고 HL-60 암세포에 대한 부착 억제실험을 수행한 결과 asadisulphide는 최소농도 2 ㎍/ml에서 HL-60 세포의 부착을 98%억제하였다. 또한 assdisulphide는 HL-60 세포에 대해 ED50값은 2.6 ㎍/ml이었으며, 20 ㎍/ml 농도 이하에서 세포독성이 없고 항암효과가 있다는 사실을 규명하였다. Asadisulphide were purified from Ferrula assafoetida by organic solvent extraction and chromatography. Its inhibitory effects on the TPA-induced adherence of HL-60 cells was analyzed. Since ethyl acetate extracts of F. assafoetida has the strongest inhibitory effect on adherence of HL-60 cells, it was reextracted with ethyl acetate, hexane, and ethyl ether and chromatographed three times to isolate asadisulphide. At the minimum concentration of 2㎍/ml, asadisulphide inhibited adherence of 98% of HL-60 cells that have been treated with TPA. It also showed anti-cancer effect with no cytotoxity in the ED50 value of 20㎍/ml.

      • Guinea Pig 자궁내막 선상피세포의 Acid Phosphatase의 활성

        Histochemical and cytochemical studies were made in order to investigate changes of the acid phosphatase activities in the endometrial glandular epithelial cells of guinea pig during the estrous cycle. The results obtained are as follows: 1. Acid phosphatase activity in the uterine glands was significant throughout the estrous cycle, especially the strongest activity were observed during diestrus. 2. Cytochemical studies indicated that acid phosphatase activities were weak during proestrus and diestrus, while intense activities were observed during metestrus and diestrus in the glandular epithelial cells. 3. Acid phosphatase activities were localized in the rough endoplasmic reticulum, Golgi complex and lysosomes, and were relatively high intercellular spaces of these cells during metestrus. 4. During diestrus this enzyme activities were localized in the lumen of glands, on the lateral cell membrane and intercellular spaces of the glandular epithelial cells. Especially these cells contained many enzyme-rich lysosomes and multilamellated bodies that related autolysis of these cells.

      • 발정주기에 따른 Guinea Pig 수난관 상피세포의 미세구조 변화

        Cyclic changes in the fine structure of the oviductal epithelial cells of guinea pig during the estrous cycle were studied by transmission electron microscopy. The results obtained were as follows: 1. The epithelium of the guinea pig oviduct was consist of the ciliated cells and the secretory cells. The former did not show any cyclic changes but the latter did various cyclic changes during the estrous cycle. 2. The secretory cells characterized by well developed Golgi complexes and dilated rough endoplasmic reticulum were showed highly active during proestrous and estrous stage. 3. During metestrous stage, the secretory cells showed most active secretory activity as a number of secretory granules were most abundant and cytoplasmic buldging into the lumen was most conspicuous. 4. The thinner epithelial layer was observed during diestrous stage. And the secretory cells showed several degenerative changes during this stage as they were characterized by decreased microvilli, swollen mitochondria, cytoplamic vacuolation and the appearance of lysosomes.

      • λ-경쇄형 다발성 골수종 1예

        Multiple myeloma is a disease caused by neoplastic plasma cells that synthesize abnormal amouts of immunoglobulin or immunoglobulin fragments. Light chain myeloma are regarded as a separate category characterized by a more malignant clinical course. Light chain myelomas are said to grow fastest of all and are associated with more osteolytic lesions, more hypercalcemia, and a higher incidence of renal failure and amyloidsis than either the IgG, IgA varienties The authors experienced a case of patients with λ-light chain myeloma. A 43-year-old male patient admitted to our hospital with the chief complaint of both rib and lower back pain. The radiologic findings showed multiple pathologic fracture in ribs. osteolytic lesions in 2nd, 3rd cervical spineimmuture plasma cells. Serum electrophoresis showed normal finding. Urine electrophoresis evealed an M-spike. Urine immunoelectrophoress demonstrated λ-monoclonal protein. With the cycle of melphalan, prednisone and α-interferon chemotherapy improved of pain was observed. So we reported the case with brief review of previous literature.

      • 표준화된 인삼추출물 G115의 중추도파민신경계에 대한 신경화학적 연구(Ⅱ)

        Effects of the standardized ginseng extract (G115) on the central monoaminergic systems were investigated in comparison with that of haloperidol in rats. Immediately after sacrificed by decapitation, the striata and frontzl cortex were removed. Concentrations of the monoamines dopamine and serotonin and their metabolites were determined by HPLC-EC. G115 increased the concnetration of 5-HIAA and DOPAC/DA ratio in striatum. However, dopaminergic neuronal activities were not affected by G115 that decreased the concentrations of 5-HT and 5 HIAA in frontal cortex. G115 in combination with apomorphine significantly increased the concentration of DA and 5-HT but decreased the DOPAC/DA ratio and 5-HIAA/5-HT ratio only in frontal cortex. These results suggest that G115 like HPD inhibits the activity of nigrostriatal dopamine neuron in striatum. However, unlike HPD it activates central monoaminergic neuron activity in frontal cortex.

      • 방사선에 조사된 생쥐의 정소에 미치는 Cysteine의 방어 효과에 관한 현미경적 연구

        This study was made to investigate the radioprotective effects of cysteine on the gamma irra diation injury, by means of electron microscopic examination of the testis of mile treated by cysteine before whole body irradiation. Experimental results obtained are as follows: 1. At 30 minutes after irradiation, the nucleus of spermatogonia began to shrink and round swollen mitochondria of spermatocytes were observed in saline treated group, but cysteine treated group showed slight decrease of cytoplasmic granules. 2. At 2 hours after irradiation, spermatocytes show destruction of nuclear membrane and irregular karyoplasm which was caused by aggregations of chromatin of nucleus in saline treated group. It was also observed aggregation of chromatic in cysteine treated group, but they showed lower appearance than saline treated group. 3. In the group of 9 hours after irradiation, the saline treated group appeared large vacuoles in the cytoplasm and irregular electron dense structures in the nucleus and showed significant aggregation of chromatin. And segregation of nucleolus, aggregations of chromatin were observed in cysteine treated group.

      • Furosemide 및 Hydrochlorothiazide가 家兎 賢皮質의 Na-K-ATPase 活性에 미치는 影響

        The furosemide and hydrochlorothiazide are well known diuretics that inhibit the sodium active reabsorption in the renal tubules. Therefore it was suggested that they had a effect on the Na-K-ATPase activity, but the reports of these drugs on the Na-K-ATPase activity are rare. Author attempted to investigate the action of furosemide and hydrochlorothiazide on the Na-K-ATPase activity in the rabbit renal cortical homogenate. The results were summarized as follows: 1. The control values of Na-K-ATPase activity in the renal cortical homogenate were 1.80±0.27μM pi/10 min/㎎ protein in experimental group of furosemide and 1.58±0.29μM pi/10 min/㎎ protein in that of hydrochlorothiazide. 2. Furosemide tended to increase the Na-K-ATPase activity but was not statistically significant. From the above results, it was suggested that furosemide and hydrochlorothiazide had a diuretic action without the influence of Na-K-ATPase activity.

      • 기억에 미치는 대뇌변연계의 기능적 역할 : 에탄올 급성 투여 흰쥐의 행동 및 신경화학적 변화 Behavioral and Neurochemical Alterations Following Acute Administration of Ethanol in Rats

        Behavioral and neurochemical effects of ethanol were assessed in rats after acute administration. Ethanol, 1-5g/㎏, inhibited the memory and rotarod performance, but significantly increased the spontaneous locomotor activity. Ethanol, 1-5g/㎏, did not affect the striatal dopaminergic neuronal activity and cortical noradrenergic neuronal activity. Ethanol, 1g/㎏, decreased the contents of DA(46.7%), DOPAC(66.7%) and HVA(79.2%) in frontal cortex, but increased the contents of DA(125%), DOPAC(116.7%) and HVA(l40%) in hippocampus. Ethanol, 2-5g/㎏, significantly increased the dopaminergic neuronal activity in frontal cortex, while it remarkably decreased the monoaminergic neuronal activity in hippocampus. These results suggest that the activity of cotical dopaminergic neuron play an important role in modulation of the motot activity induced by low does of ethanol, and that the activity of cotical and hippocampal catecholaminergic neurons could be responsible for the control of memory induced by higher does of ethanol.

      • 백혈병 세포주에 대한(±)-ar-Turmerone, 자근 및 황금추출물에 의한 항암제의 세포독성 증강효과

        Using the colorimetric [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] (MTT) assay, we evaluated the chemosensitivity of 8 anticancer drugs{vincristine(VCR), vinblastine(VBL), adriamycin(ADR), cisplatin(CPDD), etoposide(VP-16), cytosine arabinoside(ara-C), bleomycin(Bleo) and cyclophosphamide(CYC)} and the cytotoxicity-enhancing effects of (±)-ar-turmerone and the extracts of the crude drugs {Lithospermum erythrorhizon(LE) and Scutellaria baicalensis (SB)} on the above mentioned anticancer drugs against HL-60 and KG-1 cells among 8 anticancer drugs, VCR, VBL, ADR, and CPDD inhibited the growth of both cell lines by more than 50%, while VP-16, ara-C, Bleo, and CYC were less effective. (±)-ar-Turmerone had significant inhibitory effects against both cell lines, showing the ID_50 values of 11.730 ㎍/㎖ and 0.292 ㎍/㎖ for HL-60 and KG-1 cells. respectively. But the extracts of LE and SB roots showed no significant cytotoxic effects. According to ID_50 values, the cytotoxicities of VCR, VBL and ADR against HL-60 were enhanced two, eight and three times by mixing (±)-ar-turmerone, five, seven and three times by adding the extract of LE root, and twenty, six and three times by mixing the extract of SB root, respectively. The cytotoxicities of the above mentioned drugs against KG-1 cell were enhanced two, seven and three times by mixing (±)-ar-turmerone, two, three and three times by combining with the extract of LB root, and two, five and two times by adding the extract of SB root, respectively. The cytotoxicity-potentiating effects of (±)-ar-turmerone and the extracts of LE and SB roots againg HL-60 cell were greater than KG-1 cell.
