        Type specimens of Korean vascular plants in the Herbarium of the Komarov Botanical Institute (LE)

        The article provides information on type specimens of 150 taxa of vascular plants from Korea, kept in the Herbarium of the Komarov Botanical Institute of the Russian Academy of Sciences. For all specimens, type category is indicated. Lectotypes for the names of Clematis brachyura Maxim., C. spectabilis Palib., Corydalis wilfordii Regel, Poa viridula Palib., Polygonum marretii H. L$\acute{e}$v., P. sagittatum L. var. hallaisanense H. L$\acute{e}$v., P. taquetii H. L$\acute{e}$v. and P. thunbergii Sieb. et Zucc. var. coreana H. L$\acute{e}$v. were designated. Type specimens examined in this article belong to the taxa described by Russian botanists V.L. Komarov, K.J. Maximovicz and I.V. Palibin, French botanist A.A.H. L$\acute{e}$veill$\acute{e}$ (some with co-author E. Vaniot) and others.

        Kinetic Behavior of Escherichia coli on Various Cheeses under Constant and Dynamic Temperature

        In this study, we developed kinetic models to predict the growth of pathogenic Escherichia coli on cheeses during storage at constant and changing temperatures. A five-strain mixture of pathogenic E. coli was inoculated onto natural cheeses (Brie and Camembert) and processed cheeses (sliced Mozzarella and sliced Cheddar) at 3 to 4 log CFU/g. The inoculated cheeses were stored at 4, 10, 15, 25, and $30^{\circ}C$ for 1 to 320 h, with a different storage time being used for each temperature. Total bacteria and E. coli cells were enumerated on tryptic soy agar and MacConkey sorbitol agar, respectively. E. coli growth data were fitted to the Baranyi model to calculate the maximum specific growth rate (${\mu}_{max}$; log CFU/g/h), lag phase duration (LPD; h), lower asymptote (log CFU/g), and upper asymptote (log CFU/g). The kinetic parameters were then analyzed as a function of storage temperature, using the square root model, polynomial equation, and linear equation. A dynamic model was also developed for varying temperature. The model performance was evaluated against observed data, and the root mean square error (RMSE) was calculated. At $4^{\circ}C$, E. coli cell growth was not observed on any cheese. However, E. coli growth was observed at $10{\circ}C$ to $30^{\circ}C$C with a ${\mu}_{max}$ of 0.01 to 1.03 log CFU/g/h, depending on the cheese. The ${\mu}_{max}$ values increased as temperature increased, while LPD values decreased, and ${\mu}_{max}$ and LPD values were different among the four types of cheese. The developed models showed adequate performance (RMSE = 0.176-0.337), indicating that these models should be useful for describing the growth kinetics of E. coli on various cheeses.

        소 품종별 Melanocortin Receptor 1(MC1R) 유전자의 유전자형 빈도에 관한 연구

        The melanocortin 1 receptor (MC1R) plays a central role in regulation of eumelanin(black/brown) and phaeomelanin (red / yellow) synthesis within the mammalian melanocyte and encoded by the classical Extension (E) coat color locus. The objectives of this study were carried out to observe the genotype frequencies of MC1R in 1,044 samples of imported beef and eight different cattle breeds including Hanwoo, Holstein, Angus, Brown-Swiss, Charolais, Limousin, Simmental and Hereford. The primers for the amplification of bovine MC1R gene were designed based on a bovine MC1R gene sequence (GenBank accession no. Y19103). A size of 350 by was amplified by polymerase chain reaction(PCR), digested with each or together of two different restriction enzyme, BsrFI and MspAII, and electrophoresed in 2.5% or 4% Metaphore agarose gel for determination of genotypes. One thousand and forty four samples including eight different cattle breeds and imported beef were determinated their genotypes by PCR-RFLP. Genotype frequencies of Hanwoo were 0.10 in E e and 0.90 in ee. Allele E^D was shown in all of Holstein and Angus breeds tested which have black coat color phenotypes. However, genotype frequencies of Limousin and Simmental breeds were 0.21 and 0.07 in E^DE^D, 0.00 and 0.03 in E^De^+ , 0.38 and 0.35 in E^De, respectively. These results of this study suggest that alleles of the bovine MC1R gene are associated strongly with coat color except for Simmental and Limousin breeds with dark brown, light yellow or mosaic coat color of red and brown. Therefore, it needs to be studied on relationship between A and E loci and coat color phenotypes. Furthermore, genotypes of the bovine MC1R gene can be used as a DNA marker for distinguishing. beef between Hanwoo and Holstein and Angus having black coat color phenotype.

        Inactivation of Barotolerant Strains of Listeria monocytogenes and Escherichia coli O157:H7 by Ultra High Pressure and tert-Butylhydroquinone Combination

        Antimicrobial efficacy of ultra-high-pressure (UHP) can be enhanced by application of additional hurdles. The objective of this study was to systematically assess the enhancement in pressure lethality by TBHQ treatment, against barotolerant strains of Escherichia coli O157:H7 and Listeria monocytogenes. Two L. monocytogenes Scott A and the barotolerant OSY-328 strain, and two E. coli O157:H7 strains, EDL-933 and its barotolerant mutant, OSY-ASM, were tested. Cell suspensions containing TBHQ (50 ppm, dissolved in dimethyl sulfoxide) were pressurized at 200 to 500 MPa (23±2°C) for 1 min, plated on tryptose agar and enumerated the survivors. The TBHQ-UHP combination resulted in synergistic inactivation of both pathogens, with different degrees of lethality among strains. The pressure lethality threshold, for the combination treatment, was lower for E. coli O157:H7 (≥ 200 MPa) than for L. monocytogenes (> 300 MPa). E. coli O157:H7 strains were extremely sensitive to the TBHQ-UHP treatment, compared to Listeria strains. Interestingly, a control treatment involving DMSO-UHP combination consistently resulted in higher inactivation than that achieved by UHP alone, against all strains tested. However, sensitization of the pathogens to UHP by the additives (TBHQ in DMSO) was prominently greater for UHP than DMSO. Differences in sensitivities to the treatment between these two pathogens may be attributed to discrepancies in cellular structure or physiological functions.

        Evaluation of ARAA steel E-beam welding characteristics for the fabrication of KO HCCR TBM

        Korea has designed a helium cooled ceramic reflector (HCCR) test blanket module (TBM), including a TBM shield, called a TBM set, that will be tested in ITER. Korean RAFM steel was developed as a structural material for the HCCR TBM, and advanced reduced activation alloy (ARAA) was selected as the primary candidate from various program alloys. Fabrication technologies for the HCCR TBM were developed using ARAA to provide the method and procedure for fabricating the TBM for testing in ITER based on RCC-MRx, which was selected as the design and fabrication code and standard for the HCCR TBM. To establish and optimize welding procedures for electron beam welding of an ARAA material, variations in welding current and speed were investigated. A series of performance tests was performed before and after post-weld heat treatment to evaluate the welded specimen under the determined welding conditions.

      • Development of a Saccharomyces cerevisiae strain for the production of 1,2-propanediol by gene manipulation

        The main goal of this research was to achieve a more efficient production of 1,2-propanediol (1,2-PD) using mutated Saccharomyces cerevisiae. 1,2-PD cannot be produced by wild type S. cerevisiae. To develop a S. cerevisiae mutant that could produce 1,2-PD, the mgs gene of E. coli-K12 MG1655 and the dhaD gene of Citrobacter freundii were inserted into yeast expression vectors such as pESC-URA and pESC-TRP and transformed into the wild type of S. cerevisiae. As a result, the batch fermentation of S. cerevisiae YPH500, harboring an mgs gene inserted pJES27 vector, resulted in a yield of 0.17g/L. On the other hand, the methylglyoxal synthase of the recombinant S. cerevisiae YPH500, harboring a dhaD gene inserted pJES29 vector, was inactive and produced no detectable amount of 1,2-PD. Therefore, in order to achieve a maximum yield of 1,2-PD, we selected the pESC-TRP vector that is able to co-express the dhaD gene with the pJES27 vector. By inserting the dhaD gene into the pESC-TRP vector, the pJES30 vector was constructed. The maximal yield of 1,2-PD achieved in a 1% galactose batch fermentation by pJES27 and pJES30 harboring S. cerevisiae was 0.45g/L.

        Raman Scattering Studies of <tex> ${\rm YBa}_{2}{\rm Cu}_{3}{\rm O}_{7-{\rm x}}$</tex> Films Grown by High-Rate e-Beam Co-Evaporation

        <P>We present results of Raman scattering studies of superconducting YBa<SUB>2</SUB>Cu<SUB>3</SUB>O<SUB>7-x</SUB> (YBCO) films grown by a high-rate electron-beam co-evaporation method. It is shown by X-ray diffraction that the as-grown YBCO films have a highly c-axis oriented and in-plane aligned texture. Raman scattering measurements were used to study optical phonon modes, oxygen content and second phases of the YBCO coated conductors. Raman spectra of YBCO films with lower-transport properties exhibit additional phonon modes at ~ 300 cm<SUP>-1</SUP>, ~ 600 cm<SUP>-1</SUP>, and ~ 630 cm<SUP>-1</SUP>, which are related to second-phases such as Ba<SUB>2</SUB>Cu<SUB>3</SUB>O<SUB>5.9</SUB> and BaCuO<SUB>2</SUB>. We also performed micro-Raman scattering measurements, which enable the local characterization of the material over a few micrometer scales. We could clearly determine the chemical homogeneities of the films on which we could not have information using XRD. Our results strongly suggest that micro-Raman scattering can be a very effective tool for characterizing structural and chemical properties, which is essential for optimizing growth conditions.</P>

        Enhanced Homologous Recombination in Fusarium verticillioides by Disruption of FvKU70, a Gene Required for a Non-homologous End Joining Mechanism

        Fusarium verticillioides (teleomorph Gibberella moniliformis) is associated with maize worldwide causing ear rot and stalk rot, and produces fumonisins, a group of mycotoxins detrimental to humans and animals. While research tools are available, our understanding of the molecular mechanisms associated with fungal virulence and fumonisin biosynthesis in F. verticillioides is still limited. One of the restraints that hampers F. verticillioides gene characterization is the fact that homologous recombination (HR) frequency is very low (<2%). Screening for a true gene knock-out mutant is a laborious process due to a high number of ectopic integrations. In this study, we generated a F. verticillioides mutant (SF41) deleted for FvKU70, a gene directly responsible for non-homologous end-joining mechanism, with the aim of improving HR frequency. Here, we demonstrate that FvKU70 deletion does not impact key Fverticillioides phenotypes, e.g., development, secondary metabolism, and virulence, while dramatically improving HR frequency. Significantly, we also confirmed that a high percentage (>85%) of the HR mutant strains harbor a desired mutation with no additional copy of the mutant allele inserted in the genome. We conclude that SF41 is suitable for use as a type strain when performing high-throughput gene function studies in F. verticillioides.

      • Quitline Activity in the Republic of Korea

        To reduce tobacco use and related harm in Korea, telephone based cessation services (Quitlines) began full operation to provide regular behavioral counseling for smoking cessation in 2006. After registration in the cessation program, at least 21 calls per year are given to each client to help quit and encourage maintenance. Tailored programs for males, females, and adolescent smokers have been offered taking into account smokers' characteristics and smoking behavior. Mailing self-help quit packs and e-mail and SMS services are allowable as additional services.A total of 23,201 smokers were registered on the Quitline program from 2006 to 2014. In 2014, an average of 13,343 calls per month have been received by 28 coaches, the 1 year abstinence rate of clients is 26%, and clients' satisfaction rate is 81.6%. After introduction of the call system in 2007, client convenience and effective operations have been achieved with high technology support of a computer-based telephone system. Systematic education and evaluation programs for quit coaches have contributed to quality assurance of the services. Currently, research into development of new programs and evaluation of Quitline performance is being undertaken. A Comprehensive Multi-channel Cessation Center (CMCC) has been suggested and is now planned as a next step in the national program for smoking cessation.
