        Long Intergenic Non-Protein Coding RNA 665 Regulates Viability, Apoptosis, and Autophagy via the MiR-186-5p/MAP4K3 Axis in Hepatocellular Carcinoma

        Purpose: Long intergenic non-protein coding RNA 665 (LINC00665) plays a vital role in the development of cancer. Its functionin hepatocellular carcinoma (HCC), however, remains largely unknown. Materials and Methods: The expressions of LINC00665, miR-186-5p, and MAP4K3 were determined by qRT-PCR. Cell viabilityand apoptosis were evaluated by MTT and flow cytometry, respectively. Autophagic puncta formation was observed by fluorescencemicroscopy. Bioinformatics analysis, luciferase reporter assay, RNA immunoprecipitation, and RNA pulldown were performedto identify associations among LINC00665, miR-186-5p, and MAP4K3. Western blot was utilized to examine the expressionsof MAP4K3, Beclin-1, and LC3. Tumor growth was evaluated in a xenograft model. Results: Elevations in LINC00665 were observed in HCC tissues and cells. The overall survival of HCC patients with high levels ofLINC00665 was shorter than those with low levels. In vitro, LINC00665 depletion inhibited viability and induced apoptosis andautophagy. miR-186-5p interacted with LINC00665 and was downregulated in HCC tissues and cells. Upregulation of miR-186-5p inhibited viability and induced apoptosis and autophagy, which were attenuated by upregulation of LINC00665. MAP4K3 wasfound to possess binding sites with miR-186-5p and was upregulated in HCC tissues and cells. MAP4K3 depletion inhibited viabilityand induced apoptosis and autophagy, which were attenuated by miR-186-5p inhibitor. In vivo, miR-186-5p expression wasnegatively correlated with LINC00665 or MAP4K3 in HCC tissues, while LINC00665 was positively correlated with MAP4K3. LINC00665 knockdown suppressed tumor growth. Conclusion: LINC00665 was involved in cell viability, apoptosis, and autophagy in HCC via miR-186-5p/MAP4K3 axis, which mayprovide a new approach for HCC treatment.

        Synthesis and properties of amino acid functionalized water-soluble perylene diimides

        We prepared amino acid functionalized water-soluble perylene diimides: N,N'-bi(L-glutamic acid)-perylene- 3,4;9,10-dicarboxylic diimide (1), N,N'-bi(L-phenylalanine acid)-perylene-3,4;9,10-dicarboxylic diimide (2), N,N'-bi(Lglutamic amine)-perylene-3,4;9,10-dicarboxylic diimide (3) and N,N'-bi(L-phenylalanine amine)-perylene-3,4;9,10- dicarboxylic diimide (4). The structures of 3 and 4 were confirmed by 1H NMR, FT-IR and MS. The maximal absorption bands of compound 1 and 2 in concentrated sulfuric acid were red-shifted for about 48 and 74 nm, respectively, compared with that of Perylene-3,4,9,10-tetracarboxylic acid dianhydride (PTCDA). Nearly no fluorescence was observed for compounds 1 and 2 in water, while compounds 3 and 4 were significantly water-soluble and had very high fluorescent quantum. The mechanism of the optical properties change was discussed, and the π-π stacking caused by H+ led to the changes of fluorescence spectrum and absorption spectrum. The calculated molecular orbital energies and the frontier molecular orbital maps of compounds 1-2 based on density function theory (DFT) calculations were reported. Owing to the high water-soluble, the perylene derivatives 3 and 4 were successfully applied as high-performance fluorochromes for living hela cells imaging.

      • Two-sided Matching under the Conditions of Strict Order Relations and Threshold Orders

        A matching approach is proposed for solving the two-sided matching problem, where the preferences given by the two-sided agents are in the format of strict order relations and threshold orders. The two-sided matching problem under the conditions of strict order relations and threshold orders is firstly described. The related concepts of the two-sided matching are also introduced. In order to solve the considered two-sided matching problem, the strict order relations are converted into the Borda number matrix, and the threshold orders are transformed into the threshold Borda numbers. According to the Borda number matrix and the threshold Borda numbers of each side, the two Borda number cut matrix can be established, and then the two normalized Borda number cut matrixes can be set up. According to the two normalized Borda number cut matrixes, the synthetical normalized Borda number cut matrix can be established. Based on the synthetical normalized Borda number cut matrix, a matching model considering the two-sided matching constraint conditions can be developed. The matching alternative can be obtained by solving the matching model. Finally, a matching example between positions and staffs is given to illustrate the use of the proposed approach.

        Investigation on damage development of AP1000 nuclear power plant in strong ground motions with numerical simulation

        Seismic safety is considered to be one of the key design objectives of AP1000 nuclear power plant (NPP)in strong earthquakes. Dynamic behavior, damage development and aggravation effect are studied in thisstudy for the three main components of AP1000 NPP, namely reinforced concrete shield building (RCSB),steel vessel containment (SVC) and reinforced concrete auxiliary building (RCAB). Characteristicsincluding nonlinear concrete tension and compressive constitutions with plastic damage are employedto establish the numerical model, which is further validated by existing studies. The author investigatesthree earthquakes and eight input levels with the maximum magnitude of 2.4 g and the results showthat the concrete material of both RCSB and RCAB have suffered serious damage in intense earthquakes. Considering RCAB in the whole NPP, significant damage aggravation effect can be detected, which ismainly concentrated at the upper intersection between RCSB and RCAB. SVC and reinforcing bardemonstrate excellent seismic performance with no obvious damage.

      • Two-sided Matching Decision under Multi-granularity Uncertain Linguistic Environment

        With respect to the two-sided matching problem, where the agents’ preferences are in the format of multi-granularity uncertain linguistic terms, a matching method is proposed. Some basic concepts and definitions on uncertain linguistic term and 2-tuple are firstly given. Based on this, the two-sided matching problem with multi-granularity uncertain linguistic terms is described. Then, multi-granularity uncertain linguistic term matrixes are transformed into multi-granularity 2-tuple matrixes. Furthermore, a multi-objective optimization model is developed by using the extended 2-tuple weighted average. By using the normalization method and the linear weighted method twice, the multi-objective optimization model can be converted into a single-objective optimization model. By solving the model, the matching alternative can be obtained. Example of matching demand and supply in agricultural knowledge illustrates the feasibility and effectiveness of this method.

        Performance Evaluation and Shear Resistance of Modular Prefabricated Two-Side Connected Composite Shear Walls

        This paper investigates the impact of configuration parameters on the seismic performance of the modular prefabricated two-side connected composite shear wall. Firstly, the finite element model of the modular prefabricated two-side connected composite shear wall was established and validated by Xu’s experimental results. Secondly, three aspects of parameter investigationswere discussed in detail based on the validated numerical technology, namely the design parameter of ISP, design parameter of the stud, and design parameter of reinforced concrete faceplate (RCF). Then, the computation formula of shear capacity is deduced on the basis of the finite element model for reference to structural design. The results of parameter analysis displayed that the seismic performance of the modular prefabricated two-side connected composite shear wall has excellent seismic performance with the array studs of the length-diameter ratio of 4 which the center distance of 150 mm, and RCF with the thickness of 75 mm. A satisfactory but unadventurous estimation of the shear capacity of a modular prefabricated two-side connected composite shear wall is supplied by the advised technique.

        Removal of lead, copper, cadmium, zinc, and nickel from aqueous solutions by alkali-modified biochar: Batch and column tests

        Engineered biochar was obtained through slow pyrolysis of hickory wood and then further modifiedwith NaOH. After modification, biochar’s surface area, cation-exchange capacity, and thermal stabilitywere significantly improved. The modified biochar exhibited much larger (2.6–5.8 times) metaladsorption capacities than the pristine biochar. Competitive batch adsorption of mixed metal ions (Pb2+,Cd2+, Cu2+, Zn2+, and Ni2+) showed preferential adsorption of Pb2+ and Cu2+ onto the modified biochar. The alkali-modified biochar in a fixed-bed column also showed strong ability to filter heavy metals fromaqueous solutions. The alkali-modified biochar thus can serve as alternative absorbent for heavy metals

        Parametric Study on Cyclic Behavior of Modular Assembly Composite Walls with Connection to Boundary Beam

        This study focuses on the investigation of seismic performance of modular assembly composite shear wall (MACW) system, which is connected to boundary beams only. The fi nite element method is employed to establish the numerical model of the MACW system and experimental data is adopted to validate the simulation. To clearly show the seismic performance of the system, seven design parameters with 36 computational cases, including height-to-span and height-to-thickness ratios of steel plate, bolt spacing, spacing between the concrete plate and connection steel plate, thicknesses of the connection steel plate and concrete plate, and the number of composite wall modules, are discussed on mechanical property and energy dissipation capacity in details. The results show that ratios of height-to-span and height-to-thickness have a signifi cant infl uence on mechanical bearing and energy dissipation properties in comparison to the other fi ve parameters. Combining systematic seismic behaviors, such as hysteresis property, out-of-plane displacement, and equivalent damping ratio, the optimal values of the above seven parameters are recommended to be 2, 600, 100, 30, 8, 50, and 2 mm respectively. Stiff ness ratios between the post-yield and initial stiff ness of all the 36 models are mainly in the range of (2.6%, 5.5%).

        Entrapment of ball-milled biochar in Ca-alginate beads for the removal of aqueous Cd(II)

        A novel adsorbent was synthesized through the entrapment of ball-milled biochar in Ca-alginate beads for the removal of aqueous Cd(II). Batch adsorption experiments were conducted to compare Cd(II) adsorption characteristics of ball-milled biochar (BMB), Ca-alginate (CA), and Ca-alginate entrapped ball-milled biochar (CA-BMB). All the tested adsorbents showed excellent sorption ability. The maximum Cd(II) adsorption capacity estimated with Langmuir isotherm modeling was 251.8 mg g−1 for CA, 227.1 mg g−1 for CA-BMB, and 40.0 mg g−1 for BMB. The results demonstrate that entrapment of ball-milled biochar in Ca-alginate provides a low-cost and high-effective way for the removal of aqueous Cd(II).

        Seismic response characteristics of base-isolated AP1000 nuclear shield building subjected to beyonddesign basis earthquake shaking

        Because of the design and construction requirements, the nuclear structures need to maintain thestructural integrity under both design state and extreme earthquake shaking. The base-isolation technologycan significantly reduce the damages of structures under extreme earthquake events, andeffectively protect the safeties of structures and internal equipment. This study proposes a base-isolationdesign for the AP1000 nuclear shield building on considering the performance requirements of theseismic isolation systems and devices of shield building. The seismic responses of isolated and nonisolatedshield buildings subjected to design basis earthquake (DBE) shaking and beyond-design basisearthquake (BDBE) shaking are analyzed, and three different strategies for controlling the displacementssubjected to BDBE shaking are performed. By comparing with nonisolated shield buildings, the flooracceleration spectra of isolated shield buildings, relative displacement, and base shear force are significantlyreduced in high-frequency region. The results demonstrate that the base-isolation technology isan effective approach to maintain the structural integrity which subjected to both DBE and BDBE shaking. A displacement control design for isolation layers subjected to BDBE shaking, which adopts fluiddampers for controlling the horizontal displacement of isolation layer is developed. The effectiveness ofthis simple method is verified through numerical analysis
