Aberrant Glycoproteins-Targeted Biomarker Discovery for Liver Cancer

Cancer is often difficult to achieve early diagnosis, which is, however, a decisive requisite for favorable outcome in cancer treatments. Cancer biomarkers have been sought for several purposes preferably in blood and pinpointing cancer cells-derived aberrant glycoproteins would be a well-grounded approach to cancer biomarker discovery. One of the glycosyltransferases responsible for aberrant glycosylation in cancer is N-acetylglucosaminyltransferase V (GnT-V), which catalyzes an addition of b1,6-N-acetylglucosamine (GlcNAc) to the core N-glycan, and many lines of evidence have demonstrated the role of N-acetylglucosaminyltransferase V (GnT-V) in cancer development. Tissue inhibitor of metalloproteinase-1 (TIMP-1) and protein tyrosine phosphatase kappa (PTPk) were suggested to be involved in cancer malignancy upon aberrantly glycosylation by GnT-V. In addition to GnT-V, several glycosyltransferase co-works to render the altered glycan structures in cancer cells including sialyl Lewis antigen and core-fucosylation, which prompted us to mine serological biomarker candidates in cancer sera. Immunodepleted on an immune-LC column, serological proteins were enriched by carbohydrate beads conjugated with lectins like L-PHA, DSA, E-selectin, AAL, Con-A. The fraction refractive to lectin enrichments was resolved on an SDS-PAGE gel, and fractionated by molecular mass. Both the captured glycoproteins and gel-separated proteins were tryptic-digested for sequence determination in an LTQ-FTICR mass spectrometer. Candidate proteins showing high sensitivity and specificity during the discovery phase were selected and the panel of biomarker candidates is currently under in-depth analyses for validation.
Repression of the F-box protein Skp2 is essential for actin damage-induced tetraploid G1 arrest

We previously reported that p53 plays a role as a key regulator in the tetraploid G1 checkpoint, which is activated by actin damage-induced cytokinesis blockade and then prevents uncoupled DNA replication and nuclear division without cytokinesis. In this study, we investigated a role of Skp2, which targets CDK2 inhibitor p27/Kip1, in actin damage-induced tetraploid G1 arrest. Expression of Skp2 was reduced, but p27/Kip1 was increased, after actin damage-induced cytokinesis blockade. The role of Skp2 repression in tetraploid G1 arrest was investigated by analyzing the effects of ectopic expression of Skp2. After actin damage, ectopic expression of Skp2 resulted in DNA synthesis and accumulation of multinucleated cells, and ultimately, induction of apoptosis. These results suggest that Skp2 repression is important for sustaining tetraploid G1 arrest after cytokinesis blockade and is required to prevent uncoupled DNA replication and nuclear division without cytokinesis. A Low-Jitter Phase-Locked Loop Based on a Charge Pump Using a Current-Bypass Technique

A charge-pump circuit using a currentbypass technique, which suppresses charge sharing and reduces the sub-threshold currents, helps to decrease phase-locked loop (PLL) jitter without resorting to a feedback amplifier. The PLL shows no stability issues and no power-up problems, which may occur when a feedback amplifier is used. The PLL is implemented in 0.11-μm CMOS technology to achieve 0.856-ps RMS and 8.75-ps peak-to-peak jitter, which is almost independent of ambient temperature while consuming 4 mW from a 1.2-V supply.

두 개의 동일한 고속의 비교기, RC 필터, 그리고 슈미트 트리거를 장착한 신호 검출기는 6-Gbps 수신단을 위해 설계되었다. 비교기 내에 있는 자체 바이어스된 증폭기는 고속의 큰 출력 전압 폭을 갖기 때문에, 신호 검출기는 모든 PVT 변화에서도 문턱 전압에 대한 규정을 만족할 수 있다. 또한 랜덤 변화에 대해서도, 제안하는 신호 검출기는 상대적으로 작은 값인 ±15 mV (±3σ) 수준의 문턱 전압의 변동을 가진다. 신호 검출기는 0.11-μm CMOS 공정을 이용하여 설계 및 검증되었다. A signal detector which incorporates two identical high-speed comparators, an RC filter, and a Schmitt trigger was designed for a 6-Gbps receiver. Since a self-biased amplifier in the comparator has the high-speed large output swing, the signal detector can meet the threshold-voltage specification over all the PVT variations. The signal detector also has a small random threshold variation of ±15 mV (±3σ). The proposed signal detector is designed and verified using a 0.11-μm CMOS technology.
The Immersed Boundary Method for Two-Dimensional Foam with Topological Changes

<B>Abstract</B><P>We extend the immersed boundary (IB) method to simulate the dynamics of a 2D dry foam by including the topological changes of the bubble network. In the article [Y. Kim, M.-C. Lai, and C. S. Peskin, J. Comput. Phys. 229:5194-5207,2010], we implemented an IB method for the foam problem in the two-dimensional case, and tested it by verifying the von Neumann relation which governs the coarsening of a two-dimensional dry foam. However, the method implemented in that article had an important limitation; we did not allow for the resolution of quadruple or higher order junctions into triple junctions. A total shrinkage of a bubble with more than four edges generates a quadruple or higher order junction. In reality, a higher order junction is unstable and resolves itself into triple junctions. We here extend the methodology previously introduced by allowing topological changes, and we illustrate the significance of such topological changes by comparing the behaviors of foams in which topological changes are allowed to those in which they are not.</P>
다국적기업 판매 자회사의 혁신 활동에 관한 탐색적 연구 : 기술 퓨전 혁신 활동 중심으로

본 연구는 다국적기업 판매 자회사의 적극적인 혁신 활동에 관한 연구이다. 기존 다국적기업 판매 자회사의 적극적인 혁신 활동에 관한 문헌은 혁신 유형을 구분해 논의된 적이 없다. 본 연구는 혁신 유형을 구분하되, 과연 기존과 크게 거리가 있는 혁신에서도 판매 자회사의 적극적인 혁신 활동이 가능한지를 확인하고자 하였다. 본 연구의 주제는 기존 문헌에서 다루지 않았던 새로운 주제이므로 연구 방법을 한국 휴대전화 제조업체를 고객으로 하는 판매자회사 사례를 통한 탐색적 연구로 채택하였다. 기존과 크게 거리가 있는 혁신으로써 기술 퓨전을 선정하였으며, 기술 퓨전 혁신 관점에서 판매 자회사의 적극적인 혁신 활동을 연구하였다. 연구 결과는 예상과 달리 판매 자회사가 현지국에 연구개발 조직이 없음에도 불구하고 기존 점진적 혁신과 크게 거리가 있는 기술 퓨전 혁신에서도 적극적인 혁신 활동이 가능하다는 것을 확인하였다. 이러한 연구 결과는 연구개발 조직이 없는 판매 자회사의 혁신 활동영역이 연구개발 조직이 있는 자회사에서나 기대되는 영역까지 확장되었음을 의미하며, 앞으로 판매 자회사의 적극적인 혁신 활동에 대한 보다 더 심도 있는 연구가 필요하다는 것을 시사한다. This paper is a study on active innovation activities of MNE(Multinational Enterprise) sales subsidiaries. The existing literature on innovation activities of MNE sales subsidiaries has not been discussed about innovation activities by the type of innovation. The purpose of this study was to identify the types of innovation, but to confirm whether innovation activities of sales subsidiaries are possible even in innovations that are far from existing. Since the subject of this paper is a new subject, the research method was adopted as an exploratory study. Technology fusion was selected as an innovation far from the existing ones, and research on innovation activities of sales subsidiaries from the view point of technology fusion innovation was conducted. As a result of the study, it was confirmed that it is possible to innovate in technology fusion innovation. The results of this study indicate that the scope of innovation activities of sales subsidiaries has been expanded to those expected by sales subsidiary with R&D organization, and this suggests that further in-depth research is needed on the innovation activities of MNE sales subsidiaries.
