        Clinical Study on Safety and Efficacy of Microwave Ablation for Primary Hyperparathyroidism

        Objective: To evaluate the safety, feasibility, and efficacy of microwave ablation (MWA) for the treatment of primary hyperparathyroidism (PHPT). Materials and Methods: This study enrolled 67 PHPT patients (22 men, 45 women; mean age, 56.0 ± 16.3 years; range, 18– 83 years) from January 2015 to December 2018. The laboratory data, including the serum intact parathyroid hormone (iPTH), calcium, phosphorus, and alkaline phosphatase (ALP) levels, were evaluated before MWA and again 2 hours, 1 day, 7 days, 1 month, 3 months, 6 months, 12 months, 18 months, and 24 months after. Results: Complete ablation was achieved with all 72 hyperplastic parathyroid glands found on the 67 patients enrolled, 64 of whom were treated in one session and 3 were treated over two sessions. The technical success rate was 100%. The median follow-up time was 13.6 months (range, 10.0–31.1 months). The clinical success rate was 89.4%. The volume reduction rate was 79.4% at 6 months. Compared to pre-MWA, the serum iPTH, calcium, phosphorus, and ALP levels had significantly improved 6 months post-MWA (iPTH, 157.3 pg/mL vs. 39.2 pg/mL; calcium, 2.75 ± 0.25 mmol/L vs. 2.34 ± 0.15 mmol/L; phosphorus, 0.86 ± 0.20 mmol/L vs. 1.12 ± 0.22 mmol/L; ALP, 79 U/L vs. 54 U/L, respectively; all, p < 0.01). Hoarseness was a major complication in 4 patients (6.0%), but it improved spontaneously within 2–3 months. Conclusion: MWA is safe, feasible, and effective for the treatment of PHPT.

        Comparison of methods for library construction and short read annotation of shellfish viral metagenomes

        The emergence and widespread use of high-throughput sequencing technologies have promoted metagenomic studies on environmental or animal samples. Library construction for metagenome sequencing and annotation of the produced sequence reads are important steps in such studies and influence the quality of metagenomic data. In this study, we collected some marine mollusk samples, such as Crassostrea hongkongensis, Chlamys farreri, and Ruditapes philippinarum, from coastal areas in South China. These samples were divided into two batches to compare two library construction methods for shellfish viral metagenome. Our analysis showed that reverse-transcribing RNA into cDNA and then amplifying it simultaneously with DNA by whole genome amplification (WGA) yielded a larger amount of DNA compared to using only WGA or WTA (whole transcriptome amplification). Moreover, higher quality libraries were obtained by agarose gel extraction rather than with AMPure bead size selection. However, the latter can also provide good results if combined with the adjustment of the filter parameters. This, together with its simplicity, makes it a viable alternative. Finally, we compared three annotation tools (BLAST, DIAMOND, and Taxonomer) and two reference databases (NCBI’s NR and Uniprot’s Uniref). Considering the limitations of computing resources and data transfer speed, we propose the use of DIAMOND with Uniref for annotating metagenomic short reads as its running speed can guarantee a good annotation rate. This study may serve as a useful reference for selecting methods for Shellfish viral metagenome library construction and read annotation.

        Fault-Tolerant Control of Nonlinear System

        The design strategies of fault-tolerant control system (FTCS) are developed, which are categorized into two types: redundant design and fault compensation. In the first type, a certain degree of redundancy is introduced to controller. The resulting closed-loop system is stable not only when all actuators are operational but also only when one actuator experiences fault. In the second type, the on-line fault-tolerant control (FTC) scheme is developed based on the fuzzy logic system. The closed-loop stability is established based on Lyapunov’s sense. The resulting closed-loop system is stable by using the corrective control law. Compared with the existing results, the fault is accommodated by the fault compensator. The updating control law is so derived that all the parameters of the closed-loop system are bounded. The advantage of redundant design is that it is unnecessary to know the position and magnitude of the fault and is performed without FDI, which reduce the computing time and avoid the false alarms, non-detection and delay due to FDI. The advantage of fault compensation is the dynamics caused by faults that can be accommodated online. Examples of the proposed design indicate that the approaches are effective.

        The gradient recovery for finite volume element method on quadrilateral meshes

        We consider the finite volume element method for elliptic problems using isoparametric bilinear elements on quadrilateral meshes. A gradient recovery method is presented by using the patch interpolation technique. Based on some superclose estimates, we prove that the recovered gradient $R(\nabla u_h)$ possesses the superconvergence: $\|\nabla u-R(\nabla u_h)\|=O(h^2)\|u\|_3$. Finally, some numerical examples are provided to illustrate our theoretical analysis.

        Emerging 2D material-based nanocarrier for cancer therapy beyond graphene

        <P><B>Abstract</B></P> <P>2D materials (e.g., graphene, graphene analogues), with unique physical and chemical properties, shows intriguing potential for the realization of versatile biomedical applications. Tremendous attention has been attracted on 2D material based nanocarriers, which play a crucial role in therapeutic drug, gene, and biomedical agent delivery for cancer theranostics. This review provides an up-to-date review on the development of nanocarriers based on 2D materials beyond graphene, including basic and emerging synthesis methods, the catalog of 2D material-based nanocarriers, biosafety, surface functional engineering, the realization of stimuli-responsive controllable release and biomedical applications. Among the properties and performance to be summarized, there is particular focus on nanocarriers loading capability, biosafety, and therapy efficiency and so on.         IL-6-miR-210 Suppresses Regulatory T Cell Function and Promotes Atrial Fibrosis by Targeting Foxp3

        The aim of this study was to explore the role of IL-6-miR-210 in the regulation of Tregs function and atrial fibrosis in atrial fibrillation (AF). The levels of interleukin (IL)-6 and IL-10 in AF patients were detected by using ELISA. Proportions of Treg cells were detected by fluorescence activated cell sorting analysis in AF patients. The expression of Foxp3, α-SMA, collagen I and collagen III were determined by western blot. The atrial mechanocytes were authenticated by vimentin immunostaining. The expression of miR-210 was performed by quantitative real-time polymerase chain reaction (qRT-PCR). TargetScan was used to predict potential targets of miR-210. The cardiomyocyte transverse sections in AF model group were observed by H&E staining. The myocardial filaments were observed by masson staining. The level of IL-6 was highly increased while the level of IL-10 (Tregs) was significantly decreased in AF patients as compared to normal control subjects, and IL-6 suppressed Tregs function and promoted the expression of α-SMA, collagen I and collagen III. Furthermore, miR-210 regulated Tregs function by targeting Foxp3, and IL-6 promoted expression of miR-210 via regulating hypoxia inducible factor-1α (HIF-1α). IL-6-miR-210 suppresses regulatory T cell function and promotes atrial fibrosis by targeting Foxp3.

        Semiparametric estimation methods for the accelerated failure time mixture cure model

        This paper provides an overview of two semiparametric estimation methods recently proposed in the literature for the accelerated failure time mixture cure model. We prove that the two estimation methods are asymptotically equivalent. A simulation is conducted to investigate the rate of convergence of the two methods. We apply these methods to fit the accelerated failure time mixture cure model to the survival times of leukemia patients receiving bone marrow transplantation.
