Background: Although sunscreen is used as a primary strategy to prevent sunburn, photoaging and skin cancer, only a few people regularly use sunscreen. Objectives: To investigate the awareness of effects of sunlight, the extent of sun exposure, and the behaviors about sunscreen uses in Korean. Methods: A questionnaire was administered 255 adult patients visited Dermatology Department at Seoul National University Boramae Hospital. It included questions about the awareness of benefit or harm of sunlight, perceived and actual extent of sun exposure, the use of sun-protective methods including sunscreen. Results: Sun exposure was chosen as the major cause of dyschromia (61.2%), skin cancer (62.8%) and wrinkle (28.6%). Respondents were likely to underestimate the extent of sun exposure. On average, quite a few respondents stated that they were exposed to sunlight more than one hour per day (23.1% on weekdays, 53.4% on weekends). But, only 8.7% thought that their sun exposures were problematic and 62.2% of respondents considered moderate sunlight exposure good for health. People`s sun protective behaviors were inadequate: only 30.8% used sunscreen regularly; 18.4% have never used sunscreen. SPF was the most important factor choosing sunscreen and 86.0% used sunscreen labeled SPF over 30. In contrast, only 40.3% used PA+++ sunscreen. Conclusion: Despite a fairly good knowledge about harmful effects of sunlight, people underestimated the risks of their sun exposure and behaviors were suboptimal.

Background: Although sunscreen is used as a primary strategy for the prevention of sunburn, photoaging, and skincancer, few people regularly use sunscreen. Objective: To investigate awareness regarding the effects of sunlight, the extent of sun exposure, and sunscreen usebehaviors in Korean subjects. Methods: A questionnaire was administered to 467 adult patients who visited the Dermatology Department at SeoulNational University Boramae Hospital. The questionnaire covered demographic characteristics, causes of wrinkles,sunspots, and skin cancer, awareness of the harmful effects of sun exposure, perceived and actual extent of sunexposure, and the use of sun-protective methods including sunscreen. Results: Sun exposure was selected as the major cause of age spots (60.6%), skin cancer (60.9%), and wrinkles(25.9%). Respondents were likely to underestimate the extent of sun exposure. On average, quite a few respondentsstated that they were exposed to sunlight for more than one hour per day (22.7% on weekdays, 52.4% onweekends). However, only 9.4% of respondents thought that their sun exposure was problematic and 62.7% ofrespondents considered moderate sunlight exposure healthy. Respondents`` sun-protective behaviors were inadequate:only 29.8% used sunscreen regularly, and 16.5% have never used sunscreen. SPF was the most important factor inchoosing sunscreen, and 83.3% used a sunscreen with a labeled SPF over 30. By contrast, only 34.6% ofrespondents used PA+++ sunscreen. Conclusion: Despite fairly good knowledge regarding the harmful effects of sunlight, subjects underestimated therisks of their sun exposure and sun-protective behaviors were suboptimal. Education on the risk of UV exposure andeffects of sunscreen is still needed. (Korean J Dermatol 2015;53(1):16∼22)

Background: The recognition of prevention and early detection of skin cancer has been increased more than before. Although skin cancer is not rare, most patients are little known about general awareness of skin cancer. Objectives: In this study, we survey the awareness and knowledge of skin cancer and sun protection behavior among South-Gyeongnam province. Methods: We reviewed a written questionnaire and patients’ self-checked records including patients’ basic medical information and knowledge of skin cancer protective modalities. Results: In total 284 subjects (94.7%(284/300), 67%(190/284) females, mean age 57.4 years) completed the survey. Most people (77.5%, 220/284) knew that the sun was most dangerous for their skin but relatively large portion of them (42.7%, 94/220) had no action to protective attitude like sun screen, large hat and gloves. Conclusion: The results show that many participants had known necessity of skin cancer prevention and importance of early detection, but didn’t have had any protective action. Therefore, a more effective skin cancer education and prevention program is needed. We suggest that the measurement of effects of a multimodal educational intervention on knowledge of sun protection practices and engagement in self sun-protection behaviors also need to be check.

Fire characteristics of a typical apartment building in Korea was studied through full scale experiment and zone model simulation. The fire was ignited at the living room and allowed to spread to other parts of a single unit in a five storied apartment building. Various data including temperatures, species concentrations, and images were collected in the experiment. A zone model(CFAST) was used to analyze the same apartment building that represents the average households in Korea. The results were compared with a full scale experiments. While CFAST allows one compartment involved with fire, the experiment allowed the fire to spread to other compartments. Therefore, the comparison between experimental data and Zone-Model data is valid until the living-room fire spread to other parts of the apartment. Flashover occurred at approximately 380 seconds in a fire experiment, and at approximately 420 seconds in Zone-Model. Based on all of data between experimental data and Zone-Model data, it is concluded that the safe escape time is about 250 seconds.

Manual calculation of the cortical thickness index (CI) and canal-to-calcar ratio (CC) using radiographs has been widely used. The purpose of this study was to investigate the diference between manual gold standard and automatic calculation based on deep convolutional neural networks (CNNs) of the proximal femur. We obtained institutional review board approval to utilize previous radiographs for the study and the radiograph images were used to train CNN architecture. The calculation experiment of a dataset of 136 images of the proximal femur to estimate CI and CC was performed using a trained CNN architecture (Automatic). Also, manual segmentation method (Manual) to calculate CI and CC was conducted using the standard protocol by two experts as a reference for the results comparison. The mean values of the Manual and Automatic calculation of CI for the proximal femur were 0.56 and 0.54, respectively, showing a statistically signifcant diference (p=0.035). Signifcant diference (p<0.001) was also seen in the calculation of CC by Manual and Automatic method resulting in 0.42 and 0.47, respectively. In addition, the automatic method showed far better results in terms of calculation speed (less than 30 s per single image). Therefore, we suggest that the manual method be carefully considered while planning a surgery.

복잡한 게놈 분석에 용이하도록 효모의 인공 염색체(YAC) 클론닝 시스템은 발달되어왔다. YAC은 박테리아의 인공 염색체(BAC)보다 더 큰 단편을 클론닝할 수 있고, 또한 클론된 단편을 쉽게 변형시킬 수 있다. 형질전환과 연계된 재조합법(Transformation-Associated Recombination; TAR)은 게놈 라이브러리를 만들지 않고 직접 게놈 DNA로부터 분리하고자 하는 유전자나 특정 염색체 부분을 클론닝 할 수 있는 방법이다. 이 방법은 spheroplast transformation을 수행하는 동안, 목적으로 하는 유전자의 5' 그리고 3' 염기 배열(hooks)을 지닌 TAR 벡터와 게놈 DNA 사이에서 일어나는 상동성 재조합에 의해 이루어진다. 효모 내의 in vivo 재조합을 이용한 TAR 클론닝법은 복잡한 게놈으로부터 목적의 염색체 부분을 원형 YAC의 형태로서 선택적으로 분리할 수 있다. 그러므로 TAR 클론닝 법은 특정 염색체로부터 YACs을 만드는데 매우 유용하여, 전체 게놈으로부터 특이적 유전자나 유전자의 family를 분리하는데 효과적인 방법으로 사료된다. Yeast artificial chromosome (YAC) cloning systems have advanced the analysis of complex genomes considerably. They permit the cloning of larger fragments than do bacterial artificial chromosome systems, and the cloned material is more easily modified. Transformation-associated recombination (TAR) is a cloning technique that allows specific chromosomal regions or genes to be isolated directly from genomic DNA without prior construction of a genomic library. This technique involves homologous recombination during spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences (hooks). Using in vivo recombination in yeast, TAR cloning selectively isolates, as circular YACs, desired chromosome segments or entire genes from complex genomes. We propose that TAR cloning can provide an efficient means for generating YACs from specific chromosomes and that TAR cloning may be useful for isolating families of genes and specific genes from total genome DNA.

Background: Actinic keratosis (AK) is a common sun-induced skin disorder of importance for the progression to invasive squamous cell carcinoma. However, there is still no good way to predict high risk AK. Objectives: To identify markers which reflect the biologic behavior of AK and to understand the pathogenesis of AK Methods: A total of 52 patients with AK and site-matched 17 normal controls were included. We evaluated solar elastosis and immunohistochemical features using the following antibodies: p53, vitamin D receptor (VDR), claudin-1, and Langerin. Results: Solar elastosis increased and Langerhans cell (LC) density decreased with aging in both patients and controls. Solar elastosis and p53 expression were higher and VDR expression was lower in patients than controls; however, they showed no statistical difference in relation to the pathologic grade of AK. Claudin-1 expression gradually decreased from normal control to severe AK and decreased in the areas with epidermal atypia. LC density in severe AK was significantly lower than in normal control and mild AK; no difference in LC density was seen among control, mild AK and moderate AK. Conclusion: Claudin-1 can be a useful marker of pathologic severity of AK. In contrast, p53 increases and VDR decreases in AK not in gradual manner but in the early steps of carcinogenesis. LC density is relatively maintained in AK until it reaches severe dysplasia.
