        임신 및 각종 갑상선질환에서 갑상선 기능 판정에 관한 연구 : 혈청유리 T4의 진단적 의의에 관한 고찰 The diagnostic value of free thyroxine by RIA

        To evaluate the diagnostic accuracy of the measurement of free thyroxine(FT4) by radioimmunoassay, we measured free T4 and T4, T3, T3RU, TSH and TBG serum levels by radioimmunoassay in 18 healthy persons and 52 patients with various thyroid diseases and 11 normal pregnant women. The results are as follows. 1) In 19 cases of overt hyperthyroidism, T3, free T4 and FTI, T4/TBG ratio reflect hyperfunction in all cases. T4 is increased in 94%(18/19) and TBG and TSH are decreased in 79%(15/19). 2) In 8 patients with overt hypothyroidism, TSH is increased in all cases and free T4 and FTI is decreased in all cases. T4 is decreased in 87.5%(7/8), T3 is decreased in 75%(6/8) and T4/TBG ratio is decreased in 62.5%(5/8). 3) In 5 patients who are clinically in euthyroid state after treatment of hyperthyroidism, T4 free T4, FTI and TSH are in the normal range in all cases and T3 is normal in 60%(3/5) and slightly increased in 40%(2/5). 4) In 10 patients who showed clinically borderline hypothyroidism after treatment of hyperthyroidism, TSH is increased in all cases and free T4 and FTI are decreased in all cases, but T4 and T3, T4/TBG ratio are in the normal limit in all cases. So after treatment of hyperthyroidism, TSH, free T4 or FTI are recommended as optimal function test. 5) In normal pregnancy, free T4, FTI and T4/TBG ratio reflect normal function, but the other parameters unreliable due to the influence of increased TBG. Also TBG and TSH level in pregnancy is increased significantly compared with normal healthy control group. 6) The coefficients of correlation between T4 and FTI were 0.862(p〈0.001) and 0.685(p〈0.001) between free T4 and T4/TBG ratio. In most patients, diagnostic value of free T4 was comparable and even superior to FTI, so free T4 measurement can be used routinely with thyrotropin assay in the diagnosis of hypothyrodism or with T3 for the diagnosis of hyperthyroidism.

        Two-stage partial melting and contrasting cooling history within the Higher Himalayan Crystalline Sequence in the far-eastern Nepal Himalaya

        The timing of partial melting and the pressure-temperature (P-T) paths in the High Himalayan Crystalline Sequence (HHCS) in far-eastern Nepal has been investigated using zircon chronology, rare earth element (REE) compositions, and P-T pseudosection analysis. Zircon from migmatites formed during Himalayan thermal events displays inherited magmatic core overgrown by two generations of metamorphic rims. The new rims are distinguished on the basis of their Tertiary ages, low MREE contents, and low Th/U ratios. The inner zircon rims from Sil+Grt+Bt+Kfs+Pl+Qtz and Ky+Sil+Grt+Bt+Ms+Pl+Qtz migmatites at different structural level of the HHCS display ages of c. 33-28Ma (Early Oligocene) and c. 21-18Ma (Early Miocene): these rims are characterized by flat MREE to HREE patterns and were overgrown by partial melt through muscovite dehydration melting under the stability of garnet, which occurred at P=c. 7-10kbar and T=c. 730-780<SUP>o</SUP>C, and at P=c. 8-14kbar and T=c. 720-770<SUP>o</SUP>C, respectively. The outer zircon rims are relatively enriched in HREE with respect to the inner rims and were overgrown at c. 27-23Ma (Late Oligocene) and at c. 18-16Ma (Early Miocene) during melt crystallization accompanying breakdown of garnet at P=c. 4-7kbar and T=c. 650-725<SUP>o</SUP>C. Early Miocene Ms-Bt leucogranites with two successively overgrown zircon rims at c. 18.3+/-0.3Ma and c. 16.3+/-0.2Ma were intruded into Early Oligocene migmatite hosts. Microstructural observations and the corresponding P-T conditions associated with the two generations of zircon rims indicate that the Early Oligocene and Early Miocene migmatites show relatively isobaric and nearly isothermal P-T paths during exhumation, respectively. The inferences are consistent with higher average cooling rates for the Early Miocene (c. 30-40<SUP>o</SUP>C/My) than the Early Oligocene (c. 15-25<SUP>o</SUP>C/My) migmatites, inferred from peak-T conditions and FT (c. 6Ma for both migmatites) and U-Pb zircon ages. The P-T-t paths of the two migmatites indicate that burial of the Early Miocene migmatites has been coeval with exhumation of the Early Oligocene migmatites, implying the formation of large-scale thrust within the HHCS.

        Effect of Dietary Copper Sources (Cupric Sulfate and Cupric Methionate) and Concentrations on Performance and Fecal Characteristics in Growing Pigs

        This study was conducted to assess the effects of organic and inorganic copper on performance in growing pigs. A total of 100 pigs, average age 63 d and initial body weight 21.46${\pm}$1.13 kg, were assigned to five treatment groups. Dietary treatments included i) CON (basal diet, 0 ppm Cu), ii) T1 (basal diet with 67 ppm Cu as cupric sulfate, $CuSO_4$), iii) T2 (basal diet with 134 ppm Cu as $CuSO_4$), iv) T3 (basal diet with 67 ppm Cu as cupric methionate, CuMet) and v) T4 (basal diet with 134 ppm Cu as CuMet). Throughout the entire experimental period, ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain: feed) ratios showed no significant differences. The dry matter digestibility was improved in the T1, T2, T3, and T4 treatments (p<0.05), as compared with CON. Nitrogen digestibility was improved in the T3 treatment group as compared with CON (p<0.05). As compared with the T1 treatment group, fecal pH values were improved in the CON, T3, and T4 treatment groups (p<0.05). Fecal Cu concentrations were significantly lower in the CON, T3, and T4 treatment groups than in T1 and T2 (p<0.05). The incidence of diarrhea was reduced when the pigs were fed on the T2, T3, and T4 diets as compared with CON. In conclusion, diets supplemented with 67 or 134 ppm Cu as CuMet may prove effective in improving nutrient digestibility and fecal pH value in growing pigs, and fecal Cu concentrations may be reduced by CuMet supplementation.

        Survival of tissue-resident memory T cells requires exogenous lipid uptake and metabolism

        Tissue-resident memory T (T<SUB>RM</SUB>) cells persist indefinitely in epithelial barrier tissues and protect the host against pathogens. However, the biological pathways that enable the long-term survival of T<SUB>RM</SUB> cells are obscure. Here we show that mouse CD8<SUP>+</SUP> T<SUB>RM</SUB> cells generated by viral infection of the skin differentially express high levels of several molecules that mediate lipid uptake and intracellular transport, including fatty-acid-binding proteins 4 and 5 (FABP4 and FABP5). We further show that T-cell-specific deficiency of Fabp4 and Fabp5 (Fabp4/Fabp5) impairs exogenous free fatty acid (FFA) uptake by CD8<SUP>+</SUP> T<SUB>RM</SUB> cells and greatly reduces their long-term survival in vivo, while having no effect on the survival of central memory T (T<SUB>CM</SUB>) cells in lymph nodes. In vitro, CD8<SUP>+</SUP> T<SUB>RM</SUB> cells, but not CD8<SUP>+</SUP> T<SUB>CM</SUB> cells, demonstrated increased mitochondrial oxidative metabolism in the presence of exogenous FFAs; this increase was not seen in Fabp4/Fabp5 double-knockout CD8<SUP>+</SUP> T<SUB>RM</SUB> cells. The persistence of CD8<SUP>+</SUP> T<SUB>RM</SUB> cells in the skin was strongly diminished by inhibition of mitochondrial FFA β-oxidation in vivo. Moreover, skin CD8<SUP>+</SUP> T<SUB>RM</SUB> cells that lacked Fabp4/Fabp5 were less effective at protecting mice from cutaneous viral infection, and lung Fabp4/Fabp5 double-knockout CD8<SUP>+</SUP> T<SUB>RM</SUB> cells generated by skin vaccinia virus (VACV) infection were less effective at protecting mice from a lethal pulmonary challenge with VACV. Consistent with the mouse data, increased FABP4 and FABP5 expression and enhanced extracellular FFA uptake were also demonstrated in human CD8<SUP>+</SUP> T<SUB>RM</SUB> cells in normal and psoriatic skin. These results suggest that FABP4 and FABP5 have a critical role in the maintenance, longevity and function of CD8<SUP>+</SUP> T<SUB>RM</SUB> cells, and suggest that CD8<SUP>+</SUP> T<SUB>RM</SUB> cells use exogenous FFAs and their oxidative metabolism to persist in tissue and to mediate protective immunity.

        Relative Genetic Effects of Duroc and Taoyuan Breeds on the Economic Traits of Their Hybrids

        For determining the relative genetic effects of Duroc (D) and Taoyuan (T) breeds on the economic traits of their hybrids, 72 litters of pigs, from four mating types, namely TT (T♂$\times$T♀), DD (D♂$\times$D♀) and D-T hybrids (TD, T♂$\times$D♀ and DT, D♂$\times$T♀) were used in this study. The various crossbreeding parameters were estimated by comparisons among mating types using linear contrasts of least-square analysis. The results of reproductive traits analysis showed that T breed had contributed superior genetic effects on the total number of piglets at birth (TBN) (p<0.10) and number of live piglets at 21 days (LP21) (p<0.05) to the D-T hybrids. Estimates of maternal genetic effects showed that the T females were superior in TBN (p<0.05), but inferior in average birth weight (ABW) and average litter birth weight (LBW) (p<0.01) to the D females. Direct heterosis effects were significant for LBW, LP21 and LWT21 (p<0.01). Least-squares analysis of other economic traits showed that T breed had relative negative effects on all growth traits, withers height (WH), body type index (BTI), average backfat thickness (ABF), carcass length (LENG), loin eye area (longissimus) (LEARA), and lean percentage (LEAN) of D-T hybrids (p<0.05). Estimates of direct genetic effects showed that the D breed was superior to the T breed in all growth and carcass traits except the average backfat (BF). Estimates of maternal genetic effects showed that average body weight at 180 days (WT180) of progenies from T sows were lighter than from D sows. Progenies from D females had larger and leaner carcass than those from T females. Direct heterosis effects were significant for average daily weight gains from 150 to 180 days ($ADG_{150-180}$) (p<0.05) and for average body weights at 150 (WT150), and 180 days (WT180), average daily weight gains from birth to 150 and 180 days ($ADG_{150}$ and $ADG_{180}$, respectively), WH, body length (BL), ABF, BTI, and LENG (p<0.01). The results showed that D-T hybrids tended to have superior TBN and LP21 than D breed, and to be superior in all growth and most conformation and carcass traits to the T breed.

      • Deletion in HSP110 T<sub>17</sub>: correlation with wild-type HSP110 expression and prognostic significance in microsatellite-unstable advanced gastric cancers

        <P>Deletion of the HSP110 T-17 mononucleotide repeat has recently been identified as a prognostic marker that is correlated with wild-type HSP110 (HSP110wt) expression in microsatellite instability-high (MSI-H) colorectal cancers. The aim of this study was to assess the correlation between deletion of the HSP110 T-17 repeat and expression of HSP110wt using DNA testing and immunohistochemistry and to determine the prognostic implications of HSP110 T-17 deletion in MSI-H advanced gastric cancers (GCs). The status of HSP110wt expression was evaluated by immunohistochemistry using an HSP110wt-specific antibody in 142 MSI-H advanced GCs. The size of the HSP110 T-17 repeat deletion was analyzed in 96 MSI-H advanced GCs; deletions were divided into small (0-2 base pairs) and large deletions (3-5 base pairs). Low and high expressions of HSP110wt were detected in 38 (26.8%) and 104 (73.2%) of the 142 cases, respectively. The HSP110 T-17 deletion was observed in 45 (46.9%) of the 96 MSI-H GC samples. Tumors with high expression of HSP110wt showed a tendency to have small or no deletion of HSP110 T-17. In Kaplan-Meier survival analysis, tumors with a large HSP110 T-17 deletion were associated with favorable overall survival and disease-free survival compared with those with small/no deletion of HSP110 T-17. However, HSP110 T-17 deletion size was not an independent prognostic factor in multivariate analysis. In summary, deletion of the HSP110 T-17 repeat was frequently observed in MSI-H GCs, and HSP110 T-17 deletion size was inversely correlated with HSP110wt expression status.         Programmed cell death ligand 1 alleviates psoriatic inflammation by suppressing IL-17A production from programmed cell death 1-high T cells

        <P>Background: Psoriasis is one of the most common chronic inflammatory diseases of the skin. Recently, IL-17-producing T cells have been shown to play a critical role in psoriatic inflammation. Programmed cell death 1 (PD-1) is a coinhibitory receptor expressed on T cells in various chronic inflammatory diseases; however, the expression and function of PD-1 during psoriatic inflammation have not previously been characterized. Objective: We examined PD-1 expression on IL-17A-producing T cells from imiquimod-treated mice and patients with psoriasis. Additionally, we investigated the therapeutic effect of recombinant programmed cell death ligand 1 (PD-L1) protein on imiquimod-induced psoriatic inflammation. Methods: PD-1 expression on IL-17A-producing gamma delta T cells from imiquimod-treated mice was examined by means of multicolor flow cytometric analysis. In the psoriatic skin of patients, PD-1 and IL-17A expression was analyzed by using immunofluorescence. The therapeutic effect of PD-L1-Fc fusion protein (PD-L1-Fc) was assessed in imiquimod-treated mice ex vivo and in vivo. Results: During imiquimod-induced psoriatic inflammation, PD-1 is overexpressed on CD27(-)V gamma 1(-) gamma delta T cells. Furthermore, PD-1 expression on IL-17A(+) T cells was confirmed in psoriatic skin tissues from patients and imiquimod-treated mice. In the CD27(-)V gamma 1(-) gamma delta T-cell population, V gamma 4(-) gamma delta T cells with V gamma 6 mRNA expression showed a high level of PD-1 expression. Furthermore, these PD-1(hi)V gamma 4(-)(V gamma 6(+)) gamma delta Tcells were specialized for anti-CD3-induced IL-17A production, which was inhibited by PD-L1-Fc treatment. In imiquimod-treated mice PD-L1-Fc reduced psoriatic inflammation when given alone and enhanced the therapeutic effect of anti-p40 when given in combination. Conclusion: PD-1 is overexpressed in IL-17A-producing T cells in both imiquimod-treated mice and patients with psoriasis. Moreover, recombinant PD-L1-Fc alleviates psoriatic inflammation in imiquimod-treated mice.</P>

      • Pseudonocardia carboxydivorans sp. nov., a carbon monoxide-oxidizing actinomycete, and an emended description of the genus Pseudonocardia

        <P>A bacterial strain, Y8(T), capable of oxidizing carbon monoxide, was isolated from a soil sample collected from a roadside in Seoul, Korea. On the basis of 16S rRNA gene sequence similarity analyses, strain Y8(T) was shown to belong to the genus Pseudonocardia and was related most closely to the type strain of Pseudonocardia alni (99.6 % similarity). The cells were aerobic and stained Gram-positive, with white aerial mycelium and brown substrate mycelium. The predominant fatty acids were 16 : 0 iso and 16 : 1 iso. The cell-wall peptidoglycan of strain Y8(T) contained meso-diaminopimelic acid. The DNA G+C content was 77 mol%. Strain Y8(T) contained MK-9 as the major menaquinone, which is different from the major menaquinone reported previously in the genus Pseudonocardia, MK-8(H(4)). DNA-DNA relatedness between strain Y8(T) and the type strains of P. alni and Pseudonocardia antarctica was respectively 10 and 63 %. Based on phylogenetic, morphological and chemotaxonomic evidence, it is proposed that strain Y8(T) (=KCCM 42678(T) =JCM 14827(T)) be classified as the type strain of a novel species, Pseudonocardia carboxydivorans sp. nov. An emended description of the genus Pseudonocardia is also presented.</P>
