Long noncoding RNA: multiple players in gene expression

Previously considered as a component of transcriptional noise, long noncoding RNAs (lncRNAs) were neglected as a therapeutic target, however, recently increasing evidence has shown that lncRNAs can participate in numerous biological processes involved in genetic regulation including epigenetic, transcriptional, and post-transcriptional regulation. In this review, we discuss the fundamental functions of lncRNAs at different regulatory levels and their roles in metabolic balance. Typical examples are introduced to illustrate their diverse molecular mechanisms. The comprehensive investigation and identification of key lncRNAs will not only contribute to insights into diseases, such as breast cancer and type II diabetes, but also provide promising therapeutic targets for related diseases. This study was to undertaken to investigate the impacts of AhR, CYP1A1, GSTM1 genetic polymorphisms on the R273G mutation in exon 8 of the tumor suppressor p53 gene (TP53) among polycyclic aromatic hydrocarbons (PAHs) exposed to coke-oven workers. One hundred thirteen workers exposed to PAH and 82 control workers were recruited. We genotyped for polymorphisms in the AhR, CYP1A1, GSTM1, and TP53 R273G mutation in blood by PCR methods, and determined the levels of 1-hydroxypyrene as PAH exposure marker in urine using the high pressure liquid chromatography assay. We found that the distribution of alcohol users and the urinary excretion of 1-OHP in the exposed workers were significantly higher than that of the control workers (p=0.004, p<0.001, respectively). Significant differences were observed in the p53 genotype distributions of smoking subjects (p=0.01, 95%CI: 1.23-6.01) and PAH exposure (p=0.008, 95%CI: 1.24-4.48), respectively. Further, significant differences were observed in the p53 exon 8 mutations for the genetic polymorphisms of Lys/Arg for AhR (p=0.02, 95%CI: 0.70-15.86), Val/Val for CYP1A1 (p=0.04, 95%CI: 0.98-19.09) and null for GSTM1 (p=0.02, 95%CI: 1.19-6.26), respectively. Our findings indicated that polymorphisms of PAH metabolic genes, such as AhR, CYP1A1, GSTM1 polymorphisms may interact with p53 genetic variants and may contribute to PAH related cancers.

The perceived aroma of oolong tea is primarilyand directly affected by its infusion matrix, and the releasepersistence of headspace volatiles can better illustrate thepersistent aroma. Headspace solid phase microextractioncoupled with gas chromatography–mass spectrometry wasperformed to analyze the headspace constituents of oolongtea. The presence of the infusion matrix seemed to preventthe headspace release of certain odorants. The release ofindole, nerolidol, and a-farnesene was also remarkablyenhanced or depressed (2.70, 1.56, and 0.69-fold in teainfusion versus in dry leaves). Moreover, the amount ofvolatile species gradually decreased with increased waterratio. Eight odorants were determined to be stable andpersistent during continuous infusion, whereas six weredetermined to be less persistent (gradually decreased orstopped releasing). The volatile dilution test further confirmedthe persistent release of nerolidol.

Applying chemical elicitors is considered a promising approach for controlling pests in the field. Our previous studies have shown that gibberellin (GA) signaling positively regulates the resistance of rice (Oryza sativa) to the brown planthopper (BPH) Nilaparvata lugens (Stål). Yet, whether the exogenous application of GAs can enhance the resistance of rice to BPH remains unknown. Here, we investigated the influence of exogenous GA3 on both defense responses in rice and BPH performance. We found that pretreating rice with GA3 decreased transcript levels of a mitogen-activated protein kinase (MAPK) and three WRKY transcription factors, as well as levels of BPH-elicited jasmonic acid (JA) and three phenolic acids but increased the release of ethylene. Moreover, GA3 treatment negatively affected the survival and development of BPH nymphs as well as the fecundity of BPH female adults. These findings suggest that GA3 promotes the resistance of rice to BPH and may, thus, be used to improve pest management in rice.

Samarium oxide (Sm2O3) nanoparticles with a narrow sizedistribution were successfully synthesized by microwaveassistedusing urea as precipitant without surfactant ortemplate. The Sm2O3 particles were characterized using X-raydiffraction analysis, field-emission scanning electronmicroscopy, field-emission transmission electron microscopyand ultraviolet-visible-near-infrared spectrophotometer. Theresults showed that the samples prepared with differentconcentration of urea had different particle sizes. When theconcentration of urea was 1.2 mol/L, the sample had thesmallest particle size. A possible mechanism for the formationof the nanoparticles was proposed. Optical properties ofSm2O3 nanoparticles showed that the nanoparticles had astrong absorption property in the deep ultraviolet regionbetween 200 nm and 270 nm.
Investigation on Texture, Magnetic Properties and Inhomogeneities of Hot Deformed Nd–Fe–B Magnet

The anisotropic hot deformed nanocyrstalline Nd.Fe.B magnet was prepared by spark plasma sintering technique using MQU-F powders. The location dependence along radial direction on the magnetic properties, microstructure and crystal alignment of the hot deformed magnet have been investigated. The inhomogeneity of the magnetic performance along radial direction was revealed with a vibrating sample magnetometer (VSM). The crystal structure and microstructure were characterized by X-ray diffractometry (XRD) and scanning electron microscopy (SEM), respectively. In addition, quantitative texture analysis was carried out based on the electron backscattered diffraction technique (EBSD). The experimental results reveal that the coordination of grain size and orientation of Nd.Fe.B grains leads to the inhomogeneities of magnetic performance.
