Rice is the major food for half of the world population. The nutrition component in rice is critical for improvement of people’s health. Vitamin E serves as important antioxidant by quenching the free radical intermediates and thus protects the cell membrane. Because of the high nutritional value and the benefits of vitamin E in human health, increasing the tocochromanol content of major agricultural crops has long been in the focus of breeding programs and genetic engineering approaches. The key genes involved in tocopherol biosynthesis have been elucidated in Arabidopsis and other model organisms. Quantitative trait locus (QTL) study performed in Arabidopsis suggested that some of these key genes and a few additional loci contribute to natural tocopherol variations. Identifying such genetic variations in rice, enrich our understanding of the genetic mechanisms controlling tocopherol variation, which can be directly applied to rice breeding programs. The application of carbon fiber reinforced polymer (CFRP) in steel structures primarily includes two categories, i.e., the bond-critical application and the contact-critical application. Debonding failure and buckling failure are the main failure modes for these two applications. Conventional electrometric techniques may not provide precise results because of the limitations associated with single-point contact measurements. A nondestructive full-field measurement technique is a valuable alternative to conventional methods. In this study, the digital image correlation (DIC) technique was adopted to investigate the bond behavior and buckling behavior of CFRP-steel composite members. The CFRP-to-steel bonded joint and the CFRPstrengthened square hollow section (SHS) steel column were tested to verify the suitability of the DIC technique. The stereo- DIC technique was utilized to measure continuous deformation. The bond-slip relationship of the CFRP-to-steel interface was derived using the DIC data. Additionally, a multi-camera DIC system consisting of four stereo-DIC subsystems was proposed and applied to the compressive test of CFRP-strengthened SHS steel column. The precise buckling location and CFRP delamination of the CFRP-strengthened SHS steel column were identified. The experimental results confirm that the stereo-DIC technique can provide effective measurements for investigating the behaviors of CFRP-steel composite members.
Grain Boundary-Mediated Reduction of Radiation Defects in Different W-Based Alloys

Grain boundaries (GBs) are believed as potent defect sinks that contribute to the radiation damage reduction of materials. The defect-GB interaction has been extensively studied in pure metals, but only a few works shed light on the influence ofmaterial component such as alloying elements. In this study, the GB-mediated reduction of radiation defects in four W-basedalloys (W–Re, W–Ta, W–Mo and W–V) is systematically investigated by atomistic simulations. Quantitative results showthat the defect reduction in W-based alloys is significantly decreased comparing with elementary W. The principal reasonfor this decrease is the presence of various micro configurations where the solute-defect binding interactions are strong. Solutes with high local stress for each atom have a positive effect on the absorption of interstitials by GB. The ability ofGB to absorb interstitials is enhanced with the increasing temperature, and also the accumulation of vacancies is increasedwhen the temperature is as high as 1200 K. The findings in this work provide useful information on the design of radiationtolerance materials.

Di-2-ethylhexyl phthalate (DEHP) which belongs to phthalatic-acid esters, is a kind of harmful, global environmental pollutants. It is a known endocrine disrupting chemical and male reproductive toxicant. However, the mechanism by which DEHP exposure result in male reproductive toxicity is still unclear. To elucidate the productive toxicity mechanism of DEHP, we attemptted to investigate oxidative stress, apoptotic effects, mRNA and protein expression of apoptosis-associated genes including FasL, caspase-3, and caspase-8 in GC-2spd (mouse spermatogenic cells). The results showed that, with the increase of DEHP concentration, cell apoptosis rate increased; the activities of relation index of oxidative stress such as malondialdehyde (MDA), superoxide dismutase (SOD) and glutatione peroxidase (GSH-PX) changed significantly; the mRNA and protein expression levels of FasL, caspase-3, -8, altered obviously. These results suggested that DEHP could induce apoptosis of GC-2spd cells through oxidative stress and FasL-dependent pathway.

The E2F-1 transcription factor is post-translationally modified and stabilized in response to various forms of DNA damage to regulate the expression of cell-cycle and pro-apoptotic genes. The sustained overexpression of E2F-1 is a characteristic feature of gastric cancer. In this study, we investigated the role of short hairpin RNA (shRNA) targeting E2F-1 gene on human gastric cancer MGC-803 cell growth in vivo, and preliminarily revealed the mechanism. Thus, we constructed recombinant pGCSIL-GFP-shRNA-E2F-1 lentiviral vector to knock down E2F-1 expression in human gastric cancer MGC-803 cells in vivo, and studied the effect of E2F-1 shRNA on growth of MGC-803 tumor and evaluated its treatment efficacy. Our data demonstrated that in a mouse model of established gastric cancer, intratumor injection of lentiviral shRNA targeting E2F-1 definitely decreased the endogenous E2F-1mRNA and protein expression in MGC-803 tumor, and inhibited tumor growth and promoted tumor cells apoptosis. Moreover, we found that E2F-1 shRNA increased the expression of phosphatase and tensin homolog (PTEN), activated caspase-3 and caspase-9,and suppressed nuclear factor (NF)-κB expression in tumor tissue as determined by reverse transcription (RT)-PCR and western blotting. In summary, shRNA targeting of E2F-1 can effectively inhibits human gastric cancer MGC-803 cell growth in vivo and may be a potential therapeutic strategy for gastric cancer.

Although the overall structure of the chloroplast genome is generally conserved, a number of sequence variations have been identified, which are valuable for plant population and evolutionary studies. Here, we constructed a chloroplast variation map of 30 landrace rice strains of Korean origin, using the Oryza rufipogon chloroplast genome (Genbank: NC_017835) as a reference. Differential distribution of single nucleotide polymorphisms (SNPs) and indels across the rice chloroplast genome is suggestive of a region-specific variation. Population structure clustering revealed the existence of two clear subgroups (indica and japonica) and an admixture group (aus). Phylogenetic analysis of the 30 landrace rice strains and six rice chloroplast references suggested and supported independent evolution of O. sativa indica and japonica. Interestingly, two “aus” type accessions, which were thought to be indica type, shared a closer relationship with the japonica type. One hypothesis is that “Korean aus” was intentionally introduced and may have obtained japonica chloroplasts during cultivation. We also calculated the nucleotide diversity of 30 accessions and compared to six rice chloroplast references, which shown a higher diversity in the indica and aus groups than in the japonica group in lower level substitution diversity.

Quantitative Assessment of the Effects of MMP-2 Polymorphisms on Lung Carcinoma Risk

Background: Previous studies assessing associations between matrix metalloproteinase 2 (MMP-2) polymorphisms and lung cancer risk reported conflicting results. A meta-analysis was therefore performed to derive a more precise estimation. Method: Case-control studies assessing associations between MMP-2 C735T and C1306T polymorphisms and lung cancer risk were included. Pooled odds ratios (ORs) with 95% confidence intervals (CIs) were estimated. Results: 7 studies with a total of 3,189 lung cancer cases and 3,013 controls were finally included into this meta-analysis. Overall, the MMP-2 C735T polymorphism was associated with lung cancer risk under the homozygote model (CC versus TT: OR =1.44, 95% CI = 1.03-2.02, $I^2$ = 0%), while the MMP-2 C1306T polymorphism also associated demonstrated links with all four models (all P values less than 0.05). Subgroup analyses by race suggested obvious associations between MMP-2 C735T and C1306T polymorphisms and lung cancer risk in Asians but not in Caucasians. There was no evidence for publication bias. Conclusion: Currently available evidence supports teh conclusion that MMP-2 C735T and C1306T polymorphisms influence susceptibility to lung cancer in Asians.
