Enhanced β-carotene production by Rhodotorula glutinis using high hydrostatic pressure

        Sui-Lou Wang,Jun-She Sun,Bei-Zhong Han,Xiao-Zong Wu 한국화학공학회 2008 Korean Journal of Chemical Engineering Vol.25 No.3

High hydrostatic pressure (HHP) technology was used for improving the ability of β-carotene biosynthesis of Rhodotorula glutinis R68. After the treatments of five repeated cycles at 300MPa for 15 min, the barotolerant mutant PR68 was obtained. After 72 h of culture, the biomass of mutant PR68 was 21.6 g/L, decreased by 8.5% compared to the parent strain R68, but its β-carotene production reached 19.4 mg/L, increased by 52.8% compared to the parent strain R68. The result of restriction fragment length polymorphism analysis suggested that mutant strain PR68 was likely to change in nucleic acid level, and thus enhanced β-carotene production in this strain was a result of gene mutation induced by HHP treatment. HHP technology seems a promising approach for improving industrial microbes

A Dynamic Offset and Delay Differential Assembly Method for OBS Network

Sui Zhicheng,Xiao Shilin,Zeng Qingji The Korea Institute of Information and Commucation 2006 Journal of communications and networks Vol.8 No.2

We study the dynamic burst assembly based on traffic prediction and offset and delay differentiation in optical burst switching network. To improve existing burst assembly mechanism and build an adaptive flexible optical burst switching network, an approach called quality of service (QoS) based adaptive dynamic assembly (QADA) is proposed in this paper. QADA method takes into account current arrival traffic in prediction time adequately and performs adaptive dynamic assembly in limited burst assembly time (BAT) range. By the simulation of burst length error, the QADA method is proved better than the existing method and can achieve the small enough predictive error for real scenarios. Then the different dynamic ranges of BAT for four traffic classes are introduced to make delay differentiation. According to the limitation of BAT range, the burst assembly is classified into one-dimension limit and two-dimension limit. We draw a comparison between one-dimension and two-dimension limit with different prediction time under QoS based offset time and find that the one-dimensional approach offers better network performance, while the two-dimensional approach provides strict inter-class differentiation. Furthermore, the final simulation results in our network condition show that QADA can execute adaptive flexible burst assembly with dynamic BAT and achieve a latency reduction, delay fairness, and offset time QoS guarantee for different traffic classes.

Electroacupuncture Promotes Motor Function Recovery in MCAO/R Rats by Activating Astrocyte-Related PI3K/AKT Pathway

        Zhang Xiao-Qing,Wang Yi-He,Sun Li,Dong Bao-Qiang,Sui Yue-Jiao,Dong Jia-Zi,Han Yang 사단법인약침학회 2022 Journal of Acupuncture & Meridian Studies Vol.15 No.5

Background: Electroacupuncture (EA) is a widely used traditional Chinese medicine method to manage various diseases, including cerebral ischemia-reperfusion injury (CIRI). Objectives: We assessed the neuroprotective effects of EA and examined its mechanism in a rat model of the middle cerebral artery occlusion-reperfusion (MCAO/R). The gait analysis was performed to evaluate the neuroprotective effects. Western blot and immunohistochemistry assays were carried out to determine the molecular mechanisms of EA. Methods: Male SD rats were randomly divided into the sham operation group, right MCAO/R group, and EA group. EA was administered every day (4/20 Hz, 10 min/1 d) at the following acupoints: Baihui (DU20), Yintang (EX-HN3), and Zusanli (ST36). Gait and motor function were analyzed from day 8 onward. Results: The plantar support and balance coordination of MCAO/R rats decreased, and the cellular structure of the ischemic penumbra was unclear. EA improved the gait dynamics of the rats, adjusted the cell structure, further activated astrocytes, and increased the expression and phosphorylation of phosphoinositide 3-kinase/protein kinase B (PI3K/PKB or AKT). Conclusion: EA promoted astrocyte-related effects in the rat model. Our findings suggest that the neuroprotective mechanism of EA may be related to the activation of the PI3K/ AKT signaling pathway. The intervention enhanced brain protection and improved motor functions.

In vitro Antioxidant Activities of Proanthocyanidins Extracted from the Lotus Seedpod and Ameliorative Effects on Learning and Memory Impairment in Scopolamine-induced Amnesia Mice

        Juan Xiao,Shuyi Li,Yong Sui,Xiaopeng Li,Qian Wu,Ruifen Zhang,Mingwei Zhang,Bijun Xie,Zhida Sun 한국식품과학회 2015 Food Science and Biotechnology Vol.24 No.4

The antioxidant activity of proanthocyanidins extracted from the lotus seedpods (LSPC) in vitro and ameliorative effects on memory impairment induced using scopolamine in mice were studied. 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging ability, oxygen radical absorbance capacity, and cellular antioxidant activity assays revealed a potent LSPC antioxidant activity. Y-maze and step-down avoidance testing showed that LSPC administration at 30, 60, and 90 mg/kg ameliorated memory impairment. LSPC improved glutathione peroxidase and superoxide dismutase activities, inhibited activities of monoamine oxidase-B, total nitric oxide synthase, neuronal nitric oxide synthase, and acetylcholinesterase, and had no influence on inducible nitric oxide synthase and nitric oxide levels in the brain. LSPC ameliorated scopolamineinduced memory impairment based on improvement of the antioxidant system and cholinergic activity, which may be associated with potent antioxidant ability.

Self-assembly and Mechanism of L-Alanine-based Dihydrazide Derivative as Excellent Gelator of Organic Solvents

        Wang, Chuan-Sheng,Wang, Xiao-Hong,Li, Zhi-Yuan,Wei, Wei,Shi, Zhong-Liang,Sui, Zhi-Tong Korean Chemical Society 2011 Bulletin of the Korean Chemical Society Vol.32 No.4

A new organogelator, L-Alanine dihydrazide derivative can self-assemble in various organic solvents and turned them into thermally reversible physical supramolecular organogels at extremely low concentrations (< 2 wt %). The gel-sol phase transition temperatures ($T_{GS}$) were determined as a function of gelator concentration and the corresponding enthalpies (${\Delta}H_g$) were extracted. Scanning electron microscopy (SEM) measurements revealed that the interspaces of fiber-like network structures were diminished with the increasing of the LMOG concentration. FT-IR spectroscopy studies revealed that hydrogen-bonding and hydrophobic interaction were the driving forces for the formation of the gels. Based on the data of XRD and molecular modeling, the possible packing modes for the formation of organogelator aggregates were proposed.

Agricultural Process and Food Engineering : Original Article ; Drying Ginseng Slices Using a Combination of Microwave and Far-Infrared Drying Techniques

( Yuan Juan Gong ),( Ying Sui ),( Chung Su Han ),( Xiao Feng Ning ) 한국농업기계학회 2016 바이오시스템공학 Vol.41 No.1

Purpose: This study was performed to improve the drying quality and drying rate of ginseng slices by combining microwave and far-infrared drying techniques. Methods: Based on single-factor experiments and analyses, a quadratic regression orthogonal rotation combination design was adopted to study the effects of the moisture content at the conversion point between the microwave and far-infrared techniques, the ginseng slice thickness and the far-infrared drying temperature on the chip drying time, the surface color difference value, the nutritional composition and the surface shrinkage rate index. Results: Compared to the far-infrared drying alone, the combined microwave and far-infrared drying resulted in an increase in the saponin content of the ginseng slices and reductions in the drying time, surface color difference, and shrinkage rate. Conclusions: We established a mathematical model of the relationships between the surface shrinkage rate index and the experimental factors using the multi-objective nonlinear optimization method to determine the optimal parameter combination, which was confirmed to be the following: microwave and far-infrared moisture contents of 65%, a ginseng slice thickness of 1 mm, and a far-infrared drying temperature of 54℃.

      • Overexpression of Platelet-derived Growth Factor-D as a Poor Prognosticator in Endometrial Cancer

Ding, Jie,Li, Xiao-Mao,Liu, Sui-Ling,Zhang, Yu,Li, Tian Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.8

Background: Emerging evidence implicates the platelet-derived growth factor-D (PDGF-D) in many types of human solid tumors. We investigated whether PDGF-D plays an important role in endometrial cancer (EC) in relation to clinicopathologic phenotype, angiogenesis, and patient prognosis. Materials and Methods: We analyzed PDGF-D protein expression by Western blotting in twenty-seven human endometrial cancer tissues, and matched normal endometrial controls collected at the third Affiliated hospital of Sun Yat-sen University during 2012-2013 (n=27). Immunohistochemical staining was performed using a human PDGF-D antibody on the endometrial cancer patients collected in the same facility during January 2001 and October 2013 (n=152). Patients were followed from the time of primary surgery in 2001-2013 until death or last follow-up. We correlated the PDGF-D expression levels with clinicopathologic parameters and prognosis in human endometrial cancer patients. Results: Compared with matched normal endometrial cases, PDGF-D was up-regulated in endometrial cancer. Expression of PDGF-D protein, found in 78% of the cases, was associated with nonendometrioid histologic type (p=0.028), FIGO stage III/IV (p=0.039), >50% solid tumor growth (p=0.048), pelvic LN metastasis (p=0.035) and ER and PR negativity (p=0.04 and 0.002). PDGF-D expression was also significantly associated with expression of VEGF-A (p=0.021). In multivariate analysis, PDGF-D expression proved to be an independent prognostic factor in addition to histologic grade and FIGO stage. Patients with high expression levels of PDGF-D had a significantly poorer overall survival rate compared with patients with no expression. Conclusions: PDGF-D expression is frequently up-regulated in endometrial cancer, and is associated with aggressive features and poor prognosis.
