Cyclin-dependent kinase 2 (CDK2) is a member of serine/threonine protein kinases, which initiates the principal transitions of the eukaryotic cell cycle and is a promising target for cancer therapy. The present study was designed to inhibit cdk2 gene expression to induce cell cycle arrest and cell proliferation suppression. Here, we constructed a series of RNA interference (RNAi) plasmids which can successfully express small interference RNA (siRNA) in the transfected human cells. The results showed that the RNAi plasmids containing the coding sequences for siRNAs down-regulated the cdk2 gene expression in human cancer cells at the mRNA and the protein levels. Furthermore, we found that the cell cycle was arrested at G0G1 phases and the cell proliferation was inhibited by different siRNAs. These results demonstrate that suppression of CDK2 activity by RNAi may be an effective strategy for gene therapy in human cancers. [BMB reports 2010; 43(4): 291-296]

Regulatory Network Analysis of MicroRNAs and Genes in Neuroblastoma

Neuroblastoma (NB), the most common extracranial solid tumor, accounts for 10% of childhood cancer. To date, scientists have gained quite a lot of knowledge about microRNAs (miRNAs) and their genes in NB. Discovering inner regulation networks, however, still presents problems. Our study was focused on determining differentially-expressed miRNAs, their target genes and transcription factors (TFs) which exert profound influence on the pathogenesis of NB. Here we constructed three regulatory networks: differentially-expressed, related and global. We compared and analyzed the differences between the three networks to distinguish key pathways and significant nodes. Certain pathways demonstrated specific features. The differentially-expressed network consists of already identified differentially-expressed genes, miRNAs and their host genes. With this network, we can clearly see how pathways of differentially expressed genes, differentially expressed miRNAs and TFs affect on the progression of NB. MYCN, for example, which is a mutated gene of NB, is targeted by hsa-miR-29a and hsa-miR-34a, and regulates another eight differentially-expressed miRNAs that target genes VEGFA, BCL2, REL2 and so on. Further related genes and miRNAs were obtained to construct the related network and it was observed that a miRNA and its target gene exhibit special features. Hsa-miR-34a, for example, targets gene MYC, which regulates hsa-miR-34a in turn. This forms a self-adaption association. TFs like MYC and PTEN having six types of adjacent nodes and other classes of TFs investigated really can help to demonstrate that TFs affect pathways through expressions of significant miRNAs involved in the pathogenesis of NB. The present study providing comprehensive data partially reveals the mechanism of NB and should facilitate future studies to gain more significant and related data results for NB.
Experimental and numerical studies of stress/strain characteristics in riveted aircraft lap joints

The fatigue property of riveted lap joint is greatly related to the riveting-induced residual stress, especially the stress distribution on the faying surface. However, an accurate study of the residual stress characteristics in the riveted sheet could be very difficult. In this paper, both numerical and experimental investigations were carried out on the stress/strain characteristics in riveted aircraft lap joints. A special specimen was designed for the test of strain variations on the faying surface of the sheet by microstrain gages. For the numerical simulation, the rivet squeezing process was analyzed using the explicit dynamic finite element (FE) method, whilst a general static FE analysis was employed for the elastic springback after the squeeze force was removed. A comparison of the strain variations between the experimental results and FE simulations shows a general good agreement, although there may be some difference for points measured near the hole surface. The FE analysis reveals that both compressive and tensile residual stresses could be introduced in the riveted sheet. Massive compressive residual stress can be created in the near-surface layer of the hole. However, the stress level is not always increased with increasing the squeeze force, and so is the improvement of fatigue life observed. Further study is still necessary to account for the fatigue life decreasing effect caused by a high squeeze force.
Study of the Band-gap Structure of a 1D-photonic Crystal by Using Different Numerical Approaches

Comparative studies between the transfer matrices method (TMM) and plane wave method (PWM) approaches have been performed on 1D photonics crystal under different conditions to show the differences between these two kinds of calculations. TMM is suitable for the design of 1D photonic crystal device with high precision and is in good agreement with experimental results, but is not suitable for the 2D and 3D photonic structures which are limited by the complicated boundary conditions at micro interfaces. The result based on the PWM approach to deal approximately with the photonic structure in approximation has not yet been strictly verified by experiment, not even for 1D photonic crystal structures. More efforts will be required to explore its validation under all physical conditions to enhance its application.
