The aim of this study was to investigate the effects of tectorigenin on chemically induced liver fibrosis in rats. Liver fibrosis was induced in rats with carbon tetrachloride, a diet high in fat, cholesterol and alcohol in the drinking water. Our results indicate that tectorigenin treatment significantly inhibited the increases in the activities of alanine aminotransferase (ALT),aspartate aminotransferase (AST) and the increases in the serum levels of hyaluronate (HA), laminin (LN) and procollagen III N-terminal peptide (PIIIP); tectorigenin treatment also significantly inhibited the increases in the amount of collagen in the livers of the fibrogenic rats. Chemically induced liver fibrosis caused a drop in the serum albumin concentration and a decrease in the ratio of albumin to globulin (A/G). Tectorigenin caused a remarkable increase at a dose of 30 mg/kg, but only a slight increase at the lower doses. Tectorigenin was also able to inhibit the increase in the liver lipid peroxidation (LPO), as well as the decrease in the activities of liver superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), caused by liver fibrosis. In addition, we present a related metabolic profile determined, using a 1H NMR spectroscopy and multivariate pattern recognition techniques. The results were consistent with the pathological examination, liver function analysis and liver fibrosis marker analysis. Furthermore, tectorigenin does not cause acute toxicity.
Effect of Joint Stiffness on the Stability of Cable-braced Grid Shells

Bolted joints used in cable-braced grid shells are typically semi-rigid joints, and the joint stiffness has a significant effect on the stability of cable-braced grid shells. The effect of joint stiffness on the stability of cable-braced grid shells is studied in this paper. Based on the experimental results of improved bolted joints, finite element models of elliptic paraboloid cable-braced grid shells with bolted joints are established, and spring elements are used to simulate the joint stiffness. The effect of the joint stiffness on the nonlinear buckling load is studied by changing the joint stiffness. The main conclusions are as follows. First, the joint rotational stiffness has a significant effect on the failure mode. When the joint rotational stiffness is small to a certain extent, the failure mode of cable-braced grid shells changes from overall buckling to local buckling. Second, the nonlinear failure mode is similar to the first-order eigenvalue buckling mode and the maximal compression stress distribution. The structural integrity is weakened, and the maximal steel tube compression stress decreases with the decrease of the joint rotational stiffness. The smaller the joint rotational stiffness, the lower the utilization rate of steel strength. The results suggest that the joint stiffness of elliptic paraboloid cable-braced grid shells should not be less than 20% of the rigid joint stiffness.

Tyrosinase inhibitors are clinically effective for treating some dermatological disorders related to melanin hyperpigmentation. Accordingly, the discovery and development of tyrosinase inhibitors have great value in the pharmaceutical and cosmetic industry. Here, a novel tyrosinase inhibitor, 6′-O-cinnamoyl-helicid (helicid cinnamylate) was successfully synthesized by a simple and effective biocatalytic approach with Aspergillus oryzae cells. Investigation of the effects of several key variables on helicid cinnamylate synthesis found that the reaction conversion, reaction rate and regioselectivity reached 99%, 9.40 mM/h and > 99%, respectively, at the optimal conditions with anhydrous acetone as the solvent, whole-cell concentration of 40 mg/mL, and the molar ratio of vinyl cinnamate to helicid of 10 at 45°C. The whole-cells retained 68.87% of its initial activity after reusing for seven batches, indicating a potent application potential in non-aqueous biocatalytic systems. It was worth noting that helicid cinnamylate demonstrated a more potent tyrosinase inhibitory activity with an IC50 value of 3.55 mM than helicid (IC50 = 4.48 mM) and arbutin (IC50 = 5.48 mM), which suggest that helicid cinnamylate could be developed as a more potential tyrosinase inhibitor. In conclusion, this study provides a novel whole-cell catalytic approach for the synthesis of helicid cinnamylate and insight into its application as a tyrosinase inhibitor.

The existence of glycine residues in long-chain scorpion toxins has been well documented. However, their role as analgesics has not been evaluated. To address this issue, we investigated the functional role of glycines in the C-terminal end of Chinese-scorpion toxin from Buthus martensii Karsch (BmK AGP-SYPU2) using site-directed mutagenesis and analgesic activity assays. Recombinant BmK AGP-SYPU2 and its mutants were efficiently expressed in E. coli and purified to homogeneity using immobilized metal ion affinity chromatography (IMAC) and cation exchange chromatography. The mouse-twisting test was used to detect the analgesic activity of BmK AGP-SYPU2 and its mutants. As a result, we identified glycines at the C-terminal end that, when altered, significantly affected analgesic activity. Also, Mut6566 was significantly decreased compared to BmK AGP-SYPU2. These data indicate that the glycines at the C-terminal end are important for the analgesic activity of BmK AGP-SYPU2. [BMB reports 2010; 43(12): 801-806]

The nonlinear optical properties have been studied using the Z-scan technique for silicon nanocrystals embedded in a SiO2 matrix formed by high-temperature annealing of SiOx/SiO2 superlattices grown by thermal evaporation. A mode-locked Ti:sapphire laser system producing 140-fs-long pulses at 800 nm was used as the optical source for the Z-scan measurements. The nonlinear refractive index and the nonlinear absorption coefficient of the silicon nanocrystals were found to be 1.2 × 10−13 cm2/W and 1.5 × 10−9 cm/W, respectively, and to be strongly enhanced compared to those of bulk silicon. Such enhancement of the nonlinear optical properties is considered to be due to the quantum confinement effect of silicon nanocrystals.

We present a novel coupled organic Rankine cycle (CORC) system driven by the low-grade waste heat, which couples a transcriticalorganic Rankine cycle with a subcritical organic Rankine cycle. Based on classical thermodynamic theory, a detailed performance analysison the novel CORC system was performed. The results show that the pressure ratio of the expander is decreased in the CORC and theselection of the working fluids becomes more flexible and abundant. With the increase of the pinch point temperature difference of theinternal heat exchanger, the net power output and thermal efficiency of the CORC all decrease. With the increase of the critical temperatureof the working fluid, the system performance of the CORC is improved. The net power output and thermal efficiency of the CORCwith isentropic working fluids are higher than those with dry working fluids.
Modeling of flow-induced crystallization of colored polypropylene in injection molding

We present a study on flow-induced crystallization of an injection-molded isotactic polypropylene (iPP) mixed with colorant additives. Two types of blue colorants were used in the study: one is the ultramarine Blue composed of Sodium Alumino Sulpho Silicate (UB) and the other is the PV Fast Blue composed of Cu-Phthalocyanine (CuPc). The CuPc colorant exhibits increased nucleation of both quiescent and flow-induced crystallization, and results in more oriented microstructures, causing a high degree of anisotropy in material properties and shrinkage of the injection molded parts. In contrast, the nucleating effect of the UB colorant is negligible under the quiescent condition, but becomes notable in the case of short-term shear treatment at high shear rates. The UB colorant does not significantly increase the degree of anisotropy as compared with the virgin iPP. A flow-induced crystallization kinetic model is used in the numerical simulation for the injection molding process, and predicted the colorant effects as observed in experiments.
Characterization of a Baculovirus Newly Isolated from the Tea Slug Moth, Iragoidae fasciata

The tea slug moth Iragoidae fasciata (Lepidoptera, Eucleidae) is one of the main insect pests that attack tea bushes. A new nucleopolyhedrovirus (NPV) called Iragoidae fasciata NPV (IrfaNPV) was recently isolated from diseased larvae. An 11,626 bp fragment of the viral genomic DNA containing the polyhedrin gene and other 12 genes was cloned and sequenced. Gene comparison and phylogenetic analysis showed that IrfaNPV is a member of the Group I NPVs. However, the genomic organization of IrfaNPV is highly distinct. In addition, electron microscopy analysis showed that IrfaNPV is a single nucleocapsid NPV (SNPV). An inoculation assay showed that IrfaNPV is semi-permissive in the Trichoplusia ni cell line Tn-5B1-4. Bioassays on lethal concentration (LC50) and lethal time (LT50) were conducted to test the susceptibility of I. fasciata larvae to the virus.
Isolation and Structural Elucidation of Related Impurities in Canrenone

Ten steroidal compounds as impurities in canrenone were isolated from the enriched mother liquor by using various chromatographic methods. Their structures were elucidated by spectrometric analysis, among which three new compounds were characterized as 3-(3-oxo-7α-(ethoxycarbonyl)methyl-17β-hydroxy-4-androsten-17α-yl) propionic acid γ-lactone (1), 3-(3-oxo-7α-ethoxy-17β-hydroxy-4-androsten-17α-yl) propionic acid γ-lactone (2) and 3-(3-oxo-5β-propionic acid-γ-lactone-6β,17β-hydroxy-4-androstan-17α-yl) propionic acid γ-lactone (3).
