        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

        The objective of present paper is to investigate the engineering properties of concrete using recycling water incorporating saccharic based stabilizing agent in order to improve qualities of concrete in the region of normal strength and high strength. According to test results, fluidity and air content do not exhibit significant difference with an increase in stabilizing agent contents. Bleeding was not observed at 30% of W Ie, regardless of stabilizing agent contents, nonetheless, concrete with stabilizing agent of 0.15% was observed to occur the least bleeding at 40 and 50% of W IC. For setting time, although presence of recycled water accelerates setting time, setting time shows to be retarded gradually with an increase in stabilizing agent content. For compressive strength, use of stabilizing agent enhanced compressive strength of concrete compared with that of concrete without stabilizing agent. In the side of strength enhancement, adequate dosage of stabilizing agent is found to be 0.15%. As dosage of stabilizing agent increased above 0.15%, drying shrinkage exhibited a tendency to decrease.

        This study is to investigate the influence of W/C and fineness modulus of aggregate on the bleeding of concrete. According to the result, the lower W/C is, the less bleeding occurs. As for the properties of bleeding with the variation of fineness modulus, the amount of bleeding is small with the continuous grading in the middle of the standard grading range in the case of coarse aggregate, and it grows smaller with a decrease of fineness modulus in the case of fine aggregate. Therefore, it proves that using coarse aggregate with the continuous grading in the middle of the standard grading range and fine aggregate with smaller fineness modulus in the standard grading range is the effective method for reducing bleeding at the same W/C.

      • 시멘트의 분말도 변화에 따른 시멘트 모르터의 역학적 특성

        This study discusses the influence of fineness of cement on mechanical properties of cement mortar. According to the result, compressive strength increases in order of C>B>OPC>A and C>B>A>OPC at mixture proportion of 1:1 and 1:3 respectively. An increase of compressive strength by increase of fineness of cement is because internal structure of concrete grows dense by an increase of hydration reaction. As for curing condition, compressive strength is higher in standard curing than in air curing at mixture proportion of 1:1 and 1:3. There is not any tendency of strength development with age and fineness of cement by 91days. Length change by drying shrinkage increases in order of C>B>OPC>A at mixture proportion of 1:1 and 1:3 due to an increase of water content by variation of W/C.

        This study is to investigate the influence of the manufacturing company and fineness of cement on bleeding of concrete. According to the results, as fineness of comment increases, slump and air content of fresh concrete decrease, and setting time is shortened. The amount of bleeding abates with an increase of fineness, and it is confirmed that bleeding does not occur especially in the case of B8000. it is confirmed that bleeding does not occur. Bleeding speed is higher in the case of OPC A than in the case OPC B, and grows low with an increase of fineness. It also becomes highest at about 60 minutes, and shows slow tendency in W/C of 45% in comparison with W/C of 50%. And It is confirmed that relativity between fineness of cement and the final amount of bleeding is very favorable. As the properties of hardened concrete, compressive strength do not make much difference between OPC A and B is little, and increases with an increase of fineness.

        In this paper, availability of deteriorated remicon due to delaying delivery time is discussed. According to test results, fluidity and air content of plain concrete exhibited a tendency to decline with a elapse of time. After 90 minutes since mixed, use of superplasticizer and AE admixture resulted in an increase in fluidity and air content. Adequate dosage of superplasticizer and AE adimixture is found to add with 0.4% and 0.002% respectively to restore the level of original slump and air content from loss one. Setting time of concrete exhibited an acceleration with an increase in leaving time of concrete. Compressive strength of plain concrete is enhanced with and increase of leaving time of concrete. Therefore, it is found that the use of superplasticizer with 0.4% and AE admixture with 0.002% has a desirable effect on restoring the deteriorated quality of concrete related to fluidity, air content and setting time without strength loss caused by delivery time delaying.

        This study is to investigate the influence of contents of mineral admixture on bleeding of concrete. According to the results, as W/B increases, bleeding occurs rapidly at early age regardless of replacement of mineral admixture, and when slump and air content are kept equal, the amount of bleeding decreases proportionately with an increase of replacing ratio of fly ash and blast furnace slag powder. Bleeding speed is highest at 60 minutes after placing concrete, and grow slower after 180 minutes. Bleeding speed also grows high with an increase of W/B, and is lower in the case of fly ash than in the case of blast furnace slag powder.

        Roles of Sulfur in the Arsenic Tolerant Plant Adiantum capillus-veneris and the Hyperaccumulator Pteris vittata

        In this study, the role of sulfur (S) and arsenic (As) species in the As hyperaccumulator Pteris vittata (P. vittata) and the As hypertolerant plant Adiantum capillus-veneris (A. capillus-veneris) were investigated. Under As exposure, sulfhydryl groups (-SH) increased in both plants, indicating that arsenate enhanced sulfur assimilation. This S then played different roles in the two plants. In A. capillus-veneris, As was mainly coordinated with S; whereas in P. vittata, it was coordinated with oxygen. Differences in As concentration and in the rate of As reduction were noted in the two plants. In A. capillus-veneris, As was present at lower levels and was reduced and coordinated (combined) with -SH. This was considered to represent a defense strategy to limit As transport to the frond. For P. vittata, we deduced that the SH group was likely used as an electron donor to reduce As (V) to As (III), therefore fewer SH groups were used to coordinate with reduced As (III). This was regarded as an accumulation strategy to facilitate As transport. The results of this study suggested that S played important roles in As detoxification and accumulation in tolerant and hyperaccumulating plants.

      • Genomic Screening for Targets Regulated by Berberine in Breast Cancer Cells

        Berberine, a common isoquinoline alkaloid, has been shown to possess anti-cancer activities. However, the underlying molecular mechanisms are still not completely understood. In the current study, we investigated the effects of berberine on cell growth, colony formation, cell cycle distribution, and whether it improved the anticancer efficiency of cisplatin and doxorubicin in human breast cancer estrogen receptor positive (ER+) MCF-7 cells and estrogen receptor negative (ER-) MDA-MB-231 cells. Notably, berberine treatment significantly inhibited cell growth and colony formation in the two cell lines, berberine in combination with cisplatin exerting synergistic growth inhibitory effects. Accompanied by decreased growth, berberine induced G1 phase arrest in MCF-7 but not MDA-MB-231 cells. To provide a more detailed understanding of the mechanisms of action of berberine, we performed genome-wide expression profiling of berberine-treated cells using cDNA microarrays. This revealed that there were 3,397 and 2,706 genes regulated by berberine in MCF-7 and MDA-MB-231 cells, respectively. Fene oncology (GO) analysis identified that many of the target genes were involved in regulation of the cell cycle, cell migration, apoptosis, and drug responses. To confirm the microarray data, qPCR analysis was conducted for 10 selected genes based on previously reported associations with breast cancer and GO analysis. In conclusion, berberine exhibits inhibitory effects on breast cancer cells proliferation, which is likely mediated by alteration of gene expression profiles.

      • Preferential Induction of CYP1A1 over CYP1B1 in Human Breast Cancer MCF-7 Cells after Exposure to Berberine

        Estrogens are considered the major breast cancer risk factor, and the carcinogenic potential of estrogens might be attributed to DNA modification caused by derivatives formed during metabolism. $17{\beta}$-estradiol ($E_2$), the main steroidal estrogen present in women, is metabolized via two major pathways: formation of 2-hydroxyestradiol (2-OH $E_2$) and 4-hydroxyestradiol ($4-OH\;E_2$) through the action of cytochrome P450 (CYP) 1A1 and 1B1, respectively. Previous reports suggested that $2-OH\;E_2$ has putative protective effects, while $4-OH\;E_2$ is genotoxic and has potent carcinogenic activity. Thus, the ratio of $2-OH\;E_2/4-OH\;E_2$ is a critical determinant of the toxicity of $E_2$ in mammary cells. In the present study, we investigated the effects of berberine on the expression profile of the estrogen metabolizing enzymes CYP1A1 and CYP1B1 in breast cancer MCF-7 cells. Berberine treatment produced significant induction of both forms at the level of mRNA expression, but with increased doses produced 16~ to 52~fold greater induction of CYP1A1 mRNA over CYP1B1 mRNA. Furthermore, berberine dramatically increased CYP1A1 protein levels but did not influence CYP1B1 protein levels in MCF-7 cells. In conclusion, we present the first report to show that berberine may provide protection against breast cancer by altering the ratio of CYP1A1/CYP1B1, could redirect $E_2$ metabolism in a more protective pathway in breast cancer MCF-7 cells.
