Role of Autophagy in Liver Injury

Autophagy is a genetically programmed, evolutionarily conserved intracellular degradation pathway involved in the trafficking of long-lived proteins and cellular organelles to the lysosome for degradation to maintain cellular homeostasis. It has been generally thought that autophagy serves as a cell survival pathway by removing damaged proteins and organelles such as mitochondrial and lipid droplets. We found that mice exposed to either acute alcohol or acetaminophen (APAP) induced autophagy process in the liver in both models. Autophagy serves as an adaptive mechanism to selectively remove alcohol-induced damaged mitochondria and excess lipid droplets. Pharmacological activation of autophagy protects against alcohol-induced steatosis and APAP-induced liver injury. The role of Parkin-mediated selective mitophagy and the role of p62 in selective autophagy for APAP-adducts will be discussed during the presentation.

For several years, temperatures in the Korean peninsula have gradually increased due to climate change, resulting in a changing environment for growth of crops and vegetables. An associated consequence is that emerging species of insect vector have caused increased viral transmission. In Jeju Island, Korea, occurrences of viral disease have increased. Here, we report characterization of five newly collected turnip mosaic virus (TuMV) isolates named KBJ1, KBJ2, KBJ3, KBJ4 and KBJ5 from a survey on Jeju Island in 2017. Full-length cDNAs of each isolate were cloned into the pJY vector downstream of cauliflower mosaic virus 35S and bacteriophage T7 RNA polymerase promoters. Their fulllength sequences share 98.9-99.9% nucleotide sequence identity and were most closely related to previously reported Korean TuMV isolates. All isolates belonged to the BR group and infected both Chinese cabbage and radish. Four isolates induced very mild symptoms in Nicotiana benthamiana but KBJ5 induced a hypersensitive response. Symptom differences may result from three amino acid differences uniquely present in KBJ5; Gly(382)Asp, Ile(891)Val, and Lys(2522)Glu in P1, P3, and NIb, respectively.

A full-length cDNA of cyclin B was isolated from ovary of the mud crab (Scylla paramamosain) in this study. This transcript encodes a polypeptide of 401 amino acids, which is highly homologous to cyclin B protein family. Reverse transcription-PCR (RT-PCR) showed that cyclin B mRNA was expressed at highest levels in ovary of the mud crab. During the ovarian maturation process, realtime RT-PCR revealed that the abundance of cyclin B mRNA increased from the second stage (early-developing stage) to the fourth stage (nearly-ripe stage) and reached the peak level at the fifth stage (ripe stage). This result indicates the identified cyclin B gene might be related to the cell proliferation in ovary, both mitotically and meiotically. Immunohistochemistry showed that cyclin B protein was localized in the cytoplasm of prophase oocytes at the second stage while enriched in the nuclei of pro-metaphase oocytes at the fourth stage. It suggests the tested cyclin B protein might play different roles in ovary at the two stages.

The nonlinear dynamics of functionally graded graphene nanoplatelets (GPLs) reinforced composite plates are studied based on the first-order shear deformation theory (FSDT). The ordinary differential equation of the plate is derived by Hamilton principle and Galerkin method. The multi-scale method is used to analyze the jump forces. The effects of GPL distribution pattern, weight fraction, total number of layers and plate size on the resonance peak under sweep excitations, the maximal amplitude and attenuation time under impact loads, the jump force under harmonic excitations are quantitatively studied. The highest nonlinearity is found for the plate with uniform GPL distribution. The change of the number of layers has no effect on the nonlinearity but affects the natural frequency. The size of the plate and weight fraction of GPL both influence the natural frequency and nonlinearity, thereby influencing the dynamic characteristics.
Staged Improvement in Awareness of Disease for Elderly Cancer Patients in Southern China

Background: In mainland China, awareness of disease of elderly cancer patients largely relies on the patients' families. We developed a staged procedure to improve their awareness of disease. Materials and Methods: Participants were 224 elderly cancer patients from 9 leading hospitals across Southern China. A questionnaire was given to the oncologists in charge of each patient to evaluate the interaction between family and patients, patient awareness of their disease and participation in medical decision-making. After first cycles of treatment, increased information of disease was given to patients with cooperation of the family. Then patient awareness of their disease and participation in medical decision-making was documented. Results: Among the 224 cancer elderly patients, 26 (11.6%) made decisions by themselves and 125 (55.8%) delegated their rights of decision-making to their family. Subordinate family members tended to play a passive role in decision-making significantly. Patients participating more in medical decision-making tended to know more about their disease. However, in contrast to the awareness of disease, patient awareness of violation of medical recommendations was reversely associated with their participation in medical decision-making. Improvement in awareness of diagnosis, stages and prognosis was achieved in about 20% elderly cancer patients. About 5% participated more actively in medical decision-making. Conclusions: Chinese elderly cancer patient awareness of disease and participation in medical decision-making is limited and relies on their family status. The staged procedure we developed to improve patient awareness of disease proved effective.
A reliability analysis method for rock slope controlled by weak structural surface

Catastrophic landslides maybe occur in rock slope due to the effect of strong earthquakes or heavy rainfall. The stability of rock slope is usually controlled by different scales of weak structural surfaces, which are uncertain and randomly exist in the rock slope. According to the geological characteristics of rock slope, two typical failure modes – plane and wedge are possible. A second-order second-moment (SOSM) method is presented to calculate the reliability index and the failure probability of rock slope, which is an improvement over the first-order second-moment (FOSM) method, and performance functions are built up with the classic limit equilibrium method. The presented method is applied to analyze the failure probability of two rock slopes at the Jinping I Hydropower Station and is compared with the Monte Carlo method and the FOSM method. The computed results show that for plane failure, the reliability index and the failure probability determined by the presented method are 0.563 and 28.7%, respectively, and the reliability index and the failure probability determined by Monte Carlo method are 0.677 and 24.9%, respectively. However, for the FOSM method, the reliability index and failure probability are –0.025 and 51.0%, respectively. For both plane failure and wedge failure, the difference between the presented method and the Monte Carlo method is very small, but the failure probability of plane failure determined by FOSM method is larger than that of the other two methods. The presented method can provide a useful tool to evaluate the failure probability of rock slope.

Background: This retrospective study was aimed to investigate the efficacy of prophylactic agents in hepatocellular carcinoma (HCC) patients receiving TACE and compare the difference between lamivudine and entecavir. Materials and Methods: A consecutive series of 203 HBV-related HCC patients receiving TACE were analyzed including 91 patients given prophylactic agents. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 IU/ml higher than the nadir level, hepatitis flares due to HBV reactivation and progression free survival (PFS) were the main endpoints. Results: Some 48 (69.6%) reached virologic response. Prophylaxis significantly reduced virologic events (8.8% vs 58.0%, p=0.000) and hepatitis flares (1.1% vs 13.4%, p=0.001). Patients presenting undetectable HBV DNA levels displayed a significantly improved PFS as compared to those who never achieved undetectable HBV DNA. Prophylaxis and e-antigen positivity were the only significant variables associated with virologic events. In addition, prophylaxis was the only independent protective factor for hepatitis flares. Liver cirrhosis, more cycles of TACE, HBV DNA negativity, a lower Cancer of the Liver Italian Program score, non-metastasis and no hepatitis flares were protective factors for PFS. Prophylactic lamivudine demonstrated similar efficacy as entecavir. Conclusions: Prophylactic agents are efficacious for prevention of HBV reactivation in HCC patients receiving TACE. Achievement of undetectable HBV DNA levels displayed a significant capability in improving PFS. Moreover, persistent tumor residual lesions, positive HBV DNA and hepatitis B flares might be causes of tumor progression in these patients.
