Comparison of stomach microRNA transcriptomes of Tibetan and Yorkshire pigs by deep sequencing

MiRNAs regulate the expression of target genes in diverse cellular processes and hence play important roles in different physiological processes, yet little is known about the stomach microRNAome (miRNAome) of the Tibetan pig. The objective of this experiment was to investigate differentially expressed stomach miRNAs participating in digestion. Firstly, we isolated total RNA by Trizol reagent from three Tibetan and three Yorkshire purebred pigs stomach samples at 90-day-old. Secondly, a comprehensive analysis of Tibetan and Yorkshire pig stomach miRNAomes was performed by small RNA sequencing in the Illumina HiSeq 2000 system. Finally, SYBR Green Real-time RT-PCR was performed to validate the differentially expressed miRNAs. We identified 318 unique miRNAs, 260 were co-expressed in both libraries, 17 and 31 miRNAs were specifically expressed in Tibetan and Yorkshire pigs respectively. Fifty six differentially expressed miRNAs were identified by the identifying differentially expressed genes 6 (IDEG6). Kyoto encyclopedia of genes and genomes analysis revealed that some of the differentially expressed miRNAs were associated with protein and fat digestion. Two differentially expressed miRNAs (miR-214-3p and ssc-un39) participating in the digestion of lipid were identified. Additionally, qRT-PCR results suggested that a higher expression of miR-214-3p in the Tibetan pig stomach could lead to relatively lower expression of calcium-dependent phospholipase A2, which is an enzyme important for the digestion of glycerol phospholipid. This study has delineated the different stomach miRNAs expression patterns of Tibetan and Yorkshire pigs, which would help explain the regulatory mechanisms of miRNAs in digestion of Tibetan pigs, and contribute to utilize a the unique digestion merits of Tibetan pig in future porcine hybridization breeding.

This paper investigates the issue of performance testing on web browsing environments. Among the typical non-functional characteristics, index of link validity will be deeply explored. A framework to certify link correctness in web site is proposed. All possible navigation paths are first formulated to represent a usage model with the Markov chain property, which is then used to generate test script file statistically. With collecting any existing failure information followed by tracing these testing browsed paths, certification analysis may be performed by applying Markov chain theory. The certification result will yield some significant information such as: test coverage, reliability measure, confidence interval, etc. The proposed mechanism may provide not only completed but also systemic methodologies to find any linking errors and other web technologies errors. Besides, an actual practice of the proposed approach to a web-based system will be demonstrated quantitatively through a certification tool.

We assessed the ability of muscle-derived stem cells (MDSC) to differentiate into smooth muscle cells (SMC) and their potential to promote the regeneration of smooth muscle with a vessel extracellular matrix (VECM)for tissue engineering of the ureter. MDSC were isolated,proliferated, and identified by flow cytometry. SMC phenotype differentiation was induced with a smooth muscle induction medium. Gene expression was evaluated by real-time quantitative polymerase chain reaction (PCR)and Western blot studies. The VECM was obtained by a decellularization process, and cytotoxic effects were evaluated by exposing the induced cells to a VECM extract. The induced cells were seeded onto VECM in vitro for 1 week, and then the compound grafts were used for ureter reconstitution in vivo. The grafts were obtained for histological studies at 2, 4, 8, and 16 weeks post-operation. Intravenous urography was used to evaluate renal function and ureteral patency. Flow cytometry demonstrated that the MDSC expressed Sca-1 and desmin, but did not express CD45. After induction, SMC phenotype gene expression was confirmed in the induced cells by real-time quantitative PCR and Western blot studies. VECM exhibited a nontoxic effect on the induced cells in vitro. At 16 weeks postoperation,a histological evaluation showed that multilayered urothelium and organized muscle fiber bundles had formed in the grafts. Intravenous urography demonstrated no evidence of ureteral stricture or hydroureteronephrosis. These results demonstrate that MDSC can be induced into SMC and that this was useful for promoting regeneration of smooth muscles for ureter tissue engineering. This paper illustrates a prototyping framework of the documentation-standards retrieval system via the data mining approach for enhancing software development quality. We first present an approach for designing a retrieval algorithm based on data mining, with the three basic technologies of machine learning, statistics and database management, applied to this system to speed up the searching time and increase the fitness. This approach derives from the observation that data mining can discover unsuspected relationships among elements in large databases. This observation suggests that data mining can be used to elicit new knowledge about the design of a subject system and that it can be applied to large legacy systems for efficiency. Finally, software development quality will be improved at the same time when the project managers retrieving for the documentation standards.
Preparation of BiPO4-Polyaniline Hybrid and its Enhanced Photocatalytic Performance

Recently, BiPO4 has been proved to be an efficient photocatalyst which exhibits more attractive activity than that of TiO2-P25 under UV irradiation. However, photocatalytic oxidation-reduction ability of BiPO4 under visible light still needs to be improved. Conjugated polymer just like polyaniline was demonstrated to be a new and efficient visible co-photocatalyst in the field of pollutant governance. As far as we know, the attempt to enhance the photochemical ability of BiPO4 by employing polyaniline as a co-photocatalyst is still very limited. Hence, in order to testify the potential enhanced performance of such heterostructure, BiPO4-polyaniline heterostructure was prepared via hydrothermal method and hybridization in this paper. The experimental results indicated that several typical pollutants such as organic dyes and heavy metal ions can be effectively removed by BiPO4-polyaniline under visible light. The enhanced removal mechanism was attributed to the high separation efficiency of photo-induced electron–hole pairs and more efficient absorption of visible light after the loading of polyaniline on the surface of BiPO4 as an out-layer compared with single BiPO4.

The electron emission characteristics, current emission stability, and uniformity of screen-printed carbon nanotube were enhanced by introducing a novel dual-particle-containing multicomponent-blending carbon nanotube cathode via the comprehensive combination of baking in the air and sintering in the inert gas atmosphere. The dual-particle-containing multicomponent-blending carbon nanotube cathode’s design and manufacturing improvements ensured its steeper emission curve variation amplitude, larger maximum emission current, better electron emission current of 806.7 μA was only 2.56 V/μm. When it was increased from 2.78 to 2.91 V/μm, the enhancing current emission amplitude of the proposed cathode was increased by 600.7 μA versus 324.1 μA of the conventional non-blending carbon nanotube cathode. The maximum emission current of the proposed cathode was 3762.5 μA. After the vacant static state of 72 h, the measured emission current had no obvious attenuation, and the fl uctuation rate of emission current did not exceed 5%. The backlight unit with dualparticle-containing multicomponent-blending carbon nanotube cathode was sealed, and the obtained emission image had high luminescence brightness and uniformity.
