        Rapid Identification of Bifidobacteria in Dairy Products by Gene-targeted Species-specific PCR Technique and DGGE

        In this paper, a rapid and reliable gene-targeted species-specific polymerase chain reaction (PCR) technique based on a two-step process was established to identify bifidobacteria in dairy products. The first step was the PCR assay for genus Bifidobacterium with genus specific primers followed by the second step, which identified the species level with species-specific primer mixtures. Ten specific primer pairs, designed from nucleotide sequences of the 16-23S rRNA region, were developed for the Bifidobacterium species including B. angulatum, B. animalis, B. bifidum, B. breve, B. catenulatum, B. infantis, B. longum, B. minimum, B. subtile, and B. thermophilum. This technique was applied to the identification of Bifidobacterium species isolated from 6 probiotic products, and four different Bifidobacterium spp. (B. bifidum, B. longum, B. infantis, and B. breve) were identified. The findings indicated that the 16S-23S rDNA gene-targeted species-specific PCR technique is a simple and reliable method for identification of bifidobacteria in probiotic products. PCR combined with Denaturing Gradient Gel Electrophoresis (DGGE) for identification of the bifidobacteria was also evaluated and compared with the gene-targeted species-specific technique. Results indicated that for fermented milk products consistency was found for both species-specific PCR and PCR-DGGE in detecting species. However, in some lyophilized products, the bands corresponding to these species were not visualized in the DGGE profile but the specific PCR gave a positive result.

      • Accurate Logic-Level Power Simulation Using Glitch Filtering and Estimation

        Power estimation tool is needed to be more fast and accurate when the power consumption is the chief concern during the chip design. Logic-level simulator is a good choice to estimate the power consumption of a chip design. In this paper we attempt to improve the accuracy of a logic-level simulator using glitch filtering and estimation techniques. We use logic-level simulator to filter some glitches and estimate the glitch power. We use slope of transition and time interval of two consecutive transitions to decide whether these transitions is partial glitches or full transitions. We estimate the transition power of each transition event and summed them up. The power simulation error is reduced from 35.8% to 7.9% referring to the Spice simulation.

        Development and Characterization of a Specific Anti-Caveolin-1 Antibody for Caveolin-1 Functional Study in Human, Goat and Mouse

        Caveolin-1 of the caveolin family of proteins regulates mammary gland development and has been shown to play a contradictory role in breast tumor progression. A specific anti-Caveolin-1 antibody will be useful for functional study of Caveolin-1 in different tissues. In this study, we generated a rabbit polyclonal antibody that specifically recognizes the N-terminal amino acids 50-65 of Caveolin-1. This polyclonal antibody specifically reacted with Caveolin-1 extracted from cells of different species, including human epithelial A431 cells, goat primary mammary epithelial cells and mice fibroblast NIH 3T3 cells, by Western blotting. Endogenous Caveolin-1 protein expressing in cells and normal human tissues was detected by this polyclonal antibody using immunocytofluorescent and immunohistochemical staining, respectively. Furthermore, an apparent decrease in Caveolin-1 expression in tumorous breast and colon tissues was detected by this polyclonal antibody. In conclusion, we have identified amino acids 50-65 of Caveolin-1, which contains an epitope that is specific to Caveolin-1 and is conserved in the human, goat and mouse. In future, this anti-Caveolin-1 antibody can be used to examine the progression of breast and colon cancers and to study functions of Caveolin-1 in human, goat and mouse cells.

        Multi-Exchange Neighborhood Search Heuristics for the Multi-Source Capacitated Facility Location Problem

        We present two local-search based metaheuristics for the multi-source capacitated facility location problem. In such a problem, each customer’s demand can be supplied by one or more facilities. The problem is NP-hard and the number of locations in the optimal solution is unknown. To keep the search process effective, the proposed methods adopt the following features: (1) a multi-exchange neighborhood structure, (2) a tabu list that keeps track of recently visited solutions, and (3) a multi-start to enhance the diversified search paths. The transportation simplex method is applied in an efficient manner to obtain the optimal solutions to neighbors of the current solution under the algorithm framework. Two in-and-out selection rules are also proposed in the algorithms with the purpose of finding promising solutions in a short computational time. Our computational results for some of the benchmark instances, as well as some instances generated using a method in the literature, have demonstrated the effectiveness of this approach.
