Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.
Ubiquitin Ligases in Cholesterol Metabolism

To maintain cholesterol homeostasis, the processes of cholesterol metabolism are regulated at multiple levels including transcription, translation, and enzymatic activity. Recently, the regulation of protein stability of some key players in cholesterol metabolism has been characterized. More and more ubiquitin ligases have been identified including gp78, Hrd1, TRC8, TEB4, Fbw7, and inducible degrader of low density lipoprotein receptor. Their working mechanisms and physiological functions are becoming revealed. Here, we summarize the structure, substrates and function of these ubiquitin ligases. Their potential application in drug discovery is also discussed.
A Scheme to Improve SOS Security against DDoS Attack Using Active Network Technology

Secure overlay services (SOS) architecture has been proposed to provide reliable communication between clients and a target under DDoS attacks. The SOS architecture employs a set of overlay nodes arranged in three hierarchical layers that controls access to the target. Although the architecture is novel and works well under simple congestion based attacks, we observe that when the attacker uses same malicious traffic raid the SOS nodes, the SOS system can not responses this kind of repeating action, and wastes the chance to collect the information of attacker. We propose a scheme to improve the secure overlay services (SOS) against the DDoS attack.

Isotropic pitch-based carbon fiber was successfully prepared from tetrahydrofuran-soluble fraction of coal tar pitch cocarbonization with petrolatum by air-blowing. The effects of reaction temperature and time, amount of petrolatum added on the composition and spinning properties of resultant pitches were investigated. It indicated that petrolatum could effectively improve the softening point, aromaticity, hydrogen content and molecular weight of the resultant pitches by promoting cross-linking and dehydrogenation polymerization reactions at low air-blowing temperature. Moreover, more aliphatic and naphthenic structures had been introduced into resultant pitches as addition of petrolatum and also inhibited the generation of quinoline-insoluble particles. The obtained green fibers were facile to be stabilized and carbonized and the resultant carbon fibers showed fully isotropic and finer, uniform diameter with smooth surface and higher tensile strength of up to 0.92 GPa. It provided a facile chemical modification method for isotropic pitch-based carbon fiber production.

Because of its greater biodiversity, ASEAN has long been a source and transit hub for transnational wildlife trafficking. Although several international and domestic wildlife enforcement mechanisms have been established, the scope and effectiveness of international legal cooperation and policy coordination between China and ASEAN remain limited. Governments have been hampered in their efforts to combat wildlife trafficking by the difficulties of investigation and prosecution in overseas jurisdictions and the maintenance of illicit supply chains by organized crime groups. This paper reviews the main trends of legal and illegal wildlife trade between China and ASEAN, explores the existing legal and policy frameworks of ASEAN countries and China, and discusses the effectiveness of cooperation between China and ASEAN countries by referring to the United Nations Convention against Transnational Organized Crime (UNTOC). Finally, put forward the legal cooperation between China and ASEAN countries in combating transnational illegal wildlife trafficking.

With the advantages of biocatalytic method, enzymes have been excavated for the synthesis of chiral amino acids by the reductive amination of ketones, offering a promising way of producing pharmaceutical intermediates. In this work, a robust phenylalanine dehydrogenase (PheDH) with wide substrate spectrum and high catalytic efficiency was constructed through rational design and active-site-targeted, site-specific mutagenesis by using the parent enzyme from Bacillus halodurans. Active sites with bonding substrate and amino acid residues surrounding the substrate binding pocket, 49L-50G-51G, 74M,77K, 122G-123T-124D-125M, 275N, 305L and 308V of the PheDH, were identified. Noticeably, the new mutant PheDH (E113D-N276L) showed approximately 6.06-fold increment of kcat/Km in the oxidative deamination and more than 1.58-fold in the reductive amination compared to that of the wide type. Meanwhile, the PheDHs exhibit high capacity of accepting benzylic and aliphatic ketone substrates. The broad specificity, high catalytic efficiency and selectivity, along with excellent thermal stability, render these broad-spectrum enzymes ideal targets for further development with potential diagnostic reagent and pharmaceutical compounds applications.
Synthesis of ditrimethylolpropane acrylate with low functionality for UV-curable coatings

An UV-curable acrylic resin, ditrimethylolpropane acrylale (DTMPA), with low functionality was synthesized by esterification of ditrimethylolpropane and acrylic acid. The influences of the synthetic process, such as catalyst, acid/alcohol ratio, inhibitor, solvent, reaction time, and temperature were discussed in detail. The chemical structure of the DTMPA was characterized by FT-IR, 1H NMR, and 13C NMR spectra. The optimal synthesis conditions of the DTMPA were determined, and the resulting product had a yield of 86.5% and pale color. The results of a UV-curing test indicate that the DTMPA has a higher curing speed than that of trimethylolpropane triacrylate.
