This experiment was conducted to investigate the leaf reflectant color degree affecting the chromosome number of regenerated plants from midrib of haploid plants derived from anther culture of Nicotiana tabacum. Plants regenerated from midrib of lower leaves of haploid included diploid, haploid and tetraploid. Those regenerated from middle and upper leaves included diploid and haploid. The frequency of diploid among the regenerated plants from midrib of haploid leaves was correlated positively with lightness (r=0.82) and yellowness-blueness degree (r=0.87) of the leaf measured by color difference meter, and was correlated negatively with rednessgreenness degree (r=0.92) of the leaf.

This study was conducted to obtain the information for breeding of aromatic flue-cured tobacco, and to select cis-abienol producing lines from the cross of NC82×Hyangcho. Parental genotypes were Hyangcho which produces cis-abienol and NC82 which doesn’t produce cis-abienol on the leaf surface. The parents, F₁, F₂, BC₁, and BC₂ generation were evaluated for cis-abienol production to determine the inheritance of cis-abienol on the leaf surface. The presence of cis-abienol on the leaf surface was determined using thin layer chromatography(TLC) method. Segregation patherns from F₂ and BC₂ generation revealed that presence or absence of cis-abienol on the leaf surface was controlled by alleles at a single locus. Cis-abienol was produced on leaves of the homozygous dominant and heterozygous genotype plant, while the homozygous recessive genotype plants did not produce cis-abienol.

This experiment was conducted to investigate some of the factors affecting the number of haploids derived from N. africana method. The number of haploids produced by N. africana method was influenced by the growing condition of the paternal plants rather than maternal plants. Thus, paternal plants grown at 30, 25, 18 and 26-22-18℃ produced 2.89, 1.40, 0.20 and 0.53 haploids per capsule, respectively, while maternal plants grown at above temperature and in the field produced 0.36 to 0.82 haploids per capsule. There was no significant difference in the number of haploids produced by N. africana method between maternal plant starved and well fed with fertilizer. More maternal haploids were obtained from the instant pollination of paternal pollen after emasculation of maternal plant flower.

AFLP(Amplified Fragment Length Polymorphism) analysis was conducted to cultivars of tobacco, Nicotiana tabacum in order to select the cultivar-specific markers. AFLP results using 12 primer sets revealed genetic diversity among 12 field grown tobacco cultivars. Polymorphic fragments amplified by PCR was purified and cloned to identify their nucleotide sequences. From the sequences of them, 40 primer sets were designed to select cultivar-specific markers. When genomic DNA isolated from tobacco were used as PCR template, a set of primers, BrSF/BrSR showed Burley-specific band patterns. The results indicate that AFLP technique used in this experiments is useful for identifying tobacco cultivars in a rapid manner.

This experiment was conducted to investigate the variability of the plants(somaclones) regenerated from mesophyll protoplast of N. tabacum. Root tip chromosome numbers were determined in 137 unselected clones regenerated from mesophyll protoplasts of the flue-cured tobacco variety Bright Yellow 4. Seventy-seven percent of the plants retained the parental chromosome number of 2n=48 and the remaining plants were tetraploid. Randomly selected 28 clones among above diploids were analyzed for variability. Mean value of all investigated characters for 28 clones were not different significantly from parental variety. however, some clones differed significantly from parental variety in leaves per plant, days to flower, nicotine, total sugar, and nitrogen contents.
