        Higher order fully discrete scheme combined with $H^1$-Galerkin mixed finite element method for semilinear reaction-diffusion equations

        We first apply a first order splitting to a semilinear reactiondiffusion equation and then discretize the resulting system by an H1- Galerkin mixed finite element method in space. This semidiscrete method yields a system of differential algebraic equations (DAEs) of index one. A priori error estimates for semidiscrete scheme are derived for both differential as well as algebraic components. For fully discretization, an implicit Runge-Kutta (IRK) methods is applied to the temporal direction and the error estimates are discussed for both components. Finally, we conclude the paper with a numerical example

      • A Novel Mutation in the DNA Binding Domain of NFKB is Associated with Speckled Leukoplakia

        Background: Activation and inactivation of nuclear factor of kappa light chain gene enhancer in B cells (NFKB) is tightly regulated to ensure effective onset and cessation of defensive inflammatory signaling. However, mutations within NFKB, or change in activation and inactivation molecules have been reported in a few cancers. Although oral squamous cell carcinoma is one of the most prevalent forms of cancer in India, with a development associated with malignant transformation of precancerous lesions, the genetic status of NFKB and relative rates of change in oral precancerous lesions remain unknown. Hence in the present study we investigated all twenty four exons of NFKB gene in two precancerous lesions, namely oral submucous fibrosis (OSMF) and oral leukoplakia (OL) to understand its occurrence, incidence and assess its possible contribution to malignant transformation. Materials and Methods: Chromosomal DNA isolated from twenty five each of OSMF and OL tissue biopsy samples were subjected to PCR amplification with intronic primers flanking twenty four exons of the NFKB gene. The PCR amplicons were subsequently subjected to direct sequencing to elucidate the mutation status. Results: Sequence analysis identified a novel heterozygous mutation, c.419T>A causing substitution of leucine with glutamine at codon 140 (L140Q) in an OL sample. Conclusions: The identification of a substitution mutation L140Q within the DNA binding domain of NFKB in OL suggests that NFKB mutation may be relatively an early event during transformation. To the best of our knowledge, this study is the first to have identified a missense mutation in NFKB in OL.

        Performance Comparison of a Vapor-Adsorption Cycle-Based Gas Turbine Inlet Air Cooling System for Different Refrigerants

        Presently, to enhance the thermal efficiency of a gas turbine power plant, turbine inlet air cooling (TIAC) is the widely used technique. The conventional refrigeration methods like vapor compression refrigeration and evaporative cooling need electric power, hence absorption and adsorption refrigeration systems are attractive options as they can be powered using the waste heat energy of the exhaust gases. Adsorption system has advantages over absorption system like scalability, requirement of lower heat source temperature, absence of corrosion and crystallization. This paper focuses on the thermodynamic analysis of waste heat powered adsorption chiller used for the cooling of intake air to enhance the net power output of the gas turbine power plant. This paper also presents a comparative analysis of the vapor-adsorption cycle-based TIAC system for four different refrigerants viz. HFC-134a, carbon dioxide, ethanol and ammonia with the motive of finding a substitute refrigerant for HFC-134a which has a high global warming potential (GWP). The adsorption chiller is mathematically modeled in MATLAB with activated carbon as the adsorbent and each one of carbon dioxide, ethanol and ammonia as the adsorbate. The variation of the coefficient of performance (COP) and specific cooling effect (SCE) with varying adsorption temperatures is presented for each pair. The net power output and primary energy rate (PER) improvement of the gas turbine power plant at different ambient temperatures are also discussed. It is observed that ammonia can improve the power plant performance significantly better compared to the other three refrigerants at ambient temperatures less than 40 ∘ ∘C.

      • BIOLOGICAL ACTIVITIES OF L - THEANINE (SUNTHEANINE™), AN AMINO ACID OF GREEN TEA, IN HUMANS

        L-theanine is a unique amino acid, found almost solely in tea plants. It is the main component responsible for the exotic taste of green tea. In our studies of L-theanine, we have found a variety of biological activities including relaxation and the alleviation of PMS.<br/> In general, animals generate very weak electric pulses on the surface of the brain, called brain waves. Brain waves are classified into four types, namely α, β, δ and θ-waves, based on their frequency. Brain waves correlate with individual mental conditions. For example, generation of α-waves is considered an index of relaxation. In human volunteers, a-waves were generated on the occipital and parietal regions of the brain surface within 40 minutes after the oral administration of 50 or 200 ㎎ Suntheanine™ without causing drowsiness.<br/> Premenstrual Syndrome (PMS) is a symptom unique to women which appears in the luteal phase from the ovulation period through the first day of menstruation. It possesses characteristics of having a peak just prior to menstruation and disappearing 1 - 2 days following the start of menstruation. Symptoms of PMS are generally categorized as mental, physical and social symptoms. When comparing the reported Symptoms of PMS by the methods of MDQ score, the Suntheanine™ group was found to have a lower incidence of PMS symptoms, including physical, mental and social symptoms. Overall, a significant alleviation of PMS symptoms by the administration of 200 ㎎ Suntheanine™ was observed.<br/> With the successful industrial production of L-theanine, we are now able to supply Suntheanine™, offering a tremendous opportunity for designing functional foods targeting relaxation and the alleviation of PMS.

      • Mutation Analysis of the Dimer Forming Domain of the Caspase 8 Gene in Oral Submucous Fibrosis and Squamous Cell Carcinomas

        Background: Missense and frame-shift mutations within the dimer forming domain of the caspase 8 gene have been identified in several cancers. However, the genetic status of this region in precancerous lesions, like oral submucous fibrosis (OSMF), and well differentiated oral squamous cell carcinomas (OSCCs) in patients from southern region of India is not known, and hence the present study was designed to address this issue. Materials and Methods: Genomic DNA isolated from biopsy tissues of thirty one oral submucous fibrosis and twenty five OSCC samples were subjected to PCR amplification with intronic primers flanking exon 7 of the caspase 8 gene. The PCR amplicons were subsequently subjected to direct sequencing to elucidate the status of mutation. Results: Sequence analysis identified a frame-shift and a novel missense mutation in two out of twenty five OSCC samples. The frame-shift mutation was due to a two base pair deletion (c.1225_1226delTG), while the missense mutation was due to substitution of wild type cysteine residue with phenylalanine at codon 426 (C426F). The missense mutation, however, was found to be heterozygous as the wild type C426C codon was also present. None of the OSMF samples carried mutations. Conclusions: The identification of mutations in OSCC lesions but not OSMF suggests that dimer forming domain mutations in caspase 8 may be limited to malignant lesions. The absence of mutations in OSMF also suggests that the samples analyzed in the present study may not have acquired transforming potential. To the best of our knowledge this is the first study to have explored and identified frame-shift and novel missense mutations in OSCC tissue samples.
