        Genotoxic Evaluation of Mikania laevigata Extract on DNA Damage Caused by Acute Coal Dust Exposure

        In the present article, we report data on the possible antigenotoxic activity of Mikania laevigata extract (MLE) after acute intratracheal instillation of coal dust using the comet assay in peripheral blood, bone marrow, and liver cells and the micronucleus test in peripheral blood of Wistar rats. The animals were pretreated for 2 weeks with saline solution (groups 1 and 2) or MLE (100mg/kg) (groups 3 and 4). On day 15, the animals were anesthetized with ketamine (80mg/kg) and xylazine (20mg/kg), and gross mineral coal dust (3mg/0.3mL saline) (groups 2 and 4) or saline solution (0.3mL) (groups 1 and 3) was administered directly in the lung by intratracheal administration. Fifteen days after coal dust or saline instillation, the animals were sacrificed, and the femur, liver, and peripheral blood were removed. The results showed a general increase in the DNA damage values at 8 hours for all treatment groups, probably related to surgical procedures that had stressed the animals. Also, liver cells from rats treated with coal dust, pretreated or not with MLE, showed statistically higher comet assay values compared to the control group at 14 days after exposure. These results could be expected because the liver metabolizes a variety of organic compounds to more polar by-products. On the other hand, the micronucleus assay results did not show significant differences among groups. Therefore, our data do not support the antimutagenic activity of M. laevigata as a modulator of DNA damage after acute coal dust instillation.

        Awake craniotomy removal of a corticospinal tract developmental venous anomaly hemorrhage: A case report

        Developmental venous anomalies (DVAs) are composed of mature venous vessels that lack malformed or neoplastic elements. Although the hemorrhage risk is considered negligible, some patients may have neurological symptoms attributable to acute infarction or intracranial hemorrhage secondary to thrombosis, in the absence of a coexisting cavernous malformation. We report the case of a 42-year-old patient who presented with acute left-hand paresis secondary to a subcortical hemorrhage. This bleeding originated from a DVA in the corticospinal tract area and was surgically drained through an awake craniotomy. To accomplish this, we used a trans-precentral sulcus approach. After the complete removal of the coagulum, small venous channels appeared, which were coagulated. No associated cavernoma was found. Although the main DVA trunk was left patent, no signs of ischemia or venous infarction were observed after coagulating the small venous channels found inside the hematoma cavity. Two weeks after the procedure, the patient’s hand function improved, and he was able to resume desktop work. DVA-associated hemorrhage within the cortico-spinal tract could be safely removed with modern awake mapping techniques. This technique allowed the patient to rapidly improve his hand function.

        Comparative evaluation of shear bond strength of orthodontic brackets bonded to three-dimensionally- printed and milled materials after surface treatment and artificial aging

        Objective: This study aimed to evaluate the shear bond strength (SBS) of orthodontic brackets bonded to three-dimensionally (3D)-printed materials after various surface treatments and artificial aging compared with that bonded to computer-aided design/computer-aided manufacturing (CAD-CAM) polymethyl methacrylate (PMMA)-milled materials. Methods: Eighty cylindrical specimens were 3D printed and divided into the following four subgroups (n = 20 each) according to the surface treatment and artificial aging procedure. Group A, sandblasted with 50 μm aluminum oxide particles (SA) and aging; group B, sandblasted with 30 μm silica-coated alumina particles (CO) and aging; group C, SA without aging; and group D, CO without aging. For the control group, 20 CAD-CAM PMMA-milled cylindrical specimens were sandblasted with SA and aged. The SBS was measured using a universal testing machine (0.25 mm/min), examined at ×2.5 magnification for failure mode classification, and statistically analyzed (p = 0.05). Results: The retention obtained with the 3D-printed materials (groups A–D) was higher than that obtained with the PMMA-milled materials (control group). However, no significant difference was found between the study and control groups, except for group C (SA without aging), which showed significantly higher retention than the control group (PMMA-SA and thermocycling) (p = 0.037). Study groups A–D predominantly exhibited a cohesive specimen mode, indicating specimen fracture. Conclusions: Orthodontic brackets bonded to 3D-printed materials exhibit acceptable bonding strengths. However, 3D-printed materials are prone to cohesive failure, which may result in crown fractures.

        Characterization of Expression and Function of the Formins FHOD1, INF2, and DAAM1 in HER2-Positive Breast Cancer

        Purpose: Human epidermal growth factor receptor 2 (HER2)-targeted therapies, such as trastuzumab, benefit patients with HER2-positive metastatic breast cancer; however, owing to traditional pathway activation or alternative signaling, resistance persists. Given the crucial role of the formin family in shaping the actin cytoskeleton during cancer progression, these proteins may function downstream of the HER2 signaling pathway. Our aim was to uncover the potential correlations between formins and HER2 expression using a combination of public databases, immunohistochemistry, and functional in vitro assays. Methods: Using online databases, we identified a negative prognostic correlation between specific formins mRNA expression in HER2-positive cancers. To validate these findings at the protein level, immunohistochemistry was performed on HER2 subtype breast cancer tumors to establish the links between staining patterns and clinical characteristics. We then knocked down individual or combined formins in MDA-MB-453 and SK-BR-3 cells and investigated their effects on wound healing, transwell migration, and proliferation. Furthermore, we investigated the effects of erb-b2 receptor tyrosine kinase 2 (ERBB2)/HER2 small interfering RNA (siRNA)-mediated knockdown on the PI3K/Akt and MEK/ERK1 pathways as well as on selected formins. Results: Our results revealed that correlations between INF2, FHOD1, and DAAM1 mRNA expression and ERBB2 in HER2-subtype breast cancer were associated with worse outcomes. Using immunohistochemistry, we found that high FHOD1 protein expression was linked to higher histological grades and was negatively correlated with estrogen and progesterone receptor positivity. Upon formins knockdown, we observed effects on wound healing and transwell migration, with a minimal impact on proliferation, which was evident through single and combined knockdowns in both cell lines. Notably, siRNA-mediated knockdown of HER2 affected FHOD1 and INF2 expression, along with the phosphorylated Akt/MAPK states. Conclusion: Our study highlights the roles of FHOD1 and INF2 as downstream effectors of the HER2/Akt and HER2/MAPK pathways, suggesting that they are potential therapeutic targets in HER2-positive breast cancer.

        Selection of a carrying agent for obtaining radioactive methyliodide vapors under dynamic conditions

        A method for preparing "reagent" for radioactive methyliodide vapors production using an isotopic exchange reaction has been developed. Based on the obtained data of the isotopic exchange efficiency and hydraulic resistance, white fused alumina (700-840 ㎛) was selected as the carrying agent material for "reagent" production. The radioiodine isotopic exchange dependences on such parameters as temperature, gas flow velocity, and the methyliodide concentration in it were determined. Optimal conditions have been selected to achieve 85% of the isotopic exchange rate in 1 h of the experiment. The obtained data allowed to develop an approach to the test of iodine filters for nuclear power plants and to determine their efficiency.

        Gastric cancer (GC) remains a virtually incurable disease when metastatic and requires early screening tools for detection of early tumor stages. Therefore, finding effective strategies for prevention or recurrence of GC has become a major overall initiative. RNA-interference (RNAi) is an innovative technique that can significantly regulate the expression of oncogenes involved in gastric carcinogenesis, thus constituting a promising epigenetic approach to GC therapy. This review presents recent advances concerning the promising biomolecular mechanism of RNAi for GC treatment.

        Speckle Tracking Imaging in Patients with Pulmonary Hypertension

        BACKGROUND: Right ventricular (RV) systolic dysfunction is a strong predictor of mortality in pulmonary hypertension (PH). The goal of this study was to investigate whether right atrium (RA) and RV myocardial strain related to PH using speckle tracking echocardiography provide a superior estimation of RV systolic function than 2-dimensional (2D)-echo. METHODS: This cross-sectional study analyzed 22 patients with a diagnosis of PH stratified by right heart catheterization, and they were compared to a control group of 22 age- and sex-matched healthy subjects. RESULTS: Global longitudinal peak systolic strain measured in the RV free wall from the apical 4 chamber view was −15% vs. −14.5% when measured from the subcostal view (p = 0.99). Mean longitudinal strain during reservoir phase, and longitudinal strain rate during atrial reservoir and passive conduit function was significantly impaired measured in the right atrial free wall in patients with PH. CONCLUSIONS: This study showed impaired LV contractility in patients with PH assessed by speckle tracking strain. RV free wall longitudinal strain does not correlate with any of the measurements of RV systolic function obtained by 2D echocardiography. A major strength of RV longitudinal strain is its ability to assess the RV function without the limitations of 2D parameters. The subcostal RV strain is a feasible and accurate alternative to conventional RV strain from the apical view in patients with poor acoustic apical 4-chamber windows. The RA strain and strain rates values may be a valuable additive to assess right-sided heart function.
