        In vivo analysis of covering materials composed of biodegradable polymers enriched with flax fibers

        Background: The objective of this study was to investigate the in vivo effect of bioactive composites with poly(lactic acid) (PLA) or polycaprolactone (PCL) as the matrix, reinforced with bioplastic flax fibers, on the surrounding muscle tissue. Methods: Materials of pure PLA and PCL and their composites with flax fibers from genetically modified plants producing poly-3-hydroxybutyrate (PLA-transgen, PCL-transgen) and unmodified plants (PLA-wt, PCL-wt) were placed subcutaneous on the M. latissimus dorsi for four weeks. Results: The analysis of histological samples revealed that every tested material was differently encapsulated and the capsule thickness is much more pronounced when using the PCL composites in comparison with the PLA composites. The encapsulation by connective tissue was significantly reduced around PCL-transgen and significantly increased in the cases of PLA-transgen and PLA-wt. In the collected muscle samples, the measured protein expression of CD45, lymphocyte common antigen, was significantly increased after the use of all tested materials, with the exception of pure PCL. In contrast, the protein expression of caveolin-1 remained unchanged after treatment with the most examined materials. Only after insertion of PLA-wt, a significant increase of caveolin-1 protein expression was detected, due to the improved neovascularization. Conclusion: These data support the presumption that the new bioactive composites are biocompatible and they could be applicable in the medical field to support the regenerative processes.

        Production of recombinant colicin M in Nicotiana tabacum plants and its antimicrobial activity

        Antibiotic-resistant microorganisms causing a life-threatening infection pose a serious challenge for modern science. The rapidly growing number of incidents for which the use of standard antibiotics is ineffective forces us to develop new alternative methods of killing microorganisms. Antimicrobial proteins and peptides (AMPs) can be promising candidates to solve this problem. Colicin-M is one of the representatives of this group and is naturally produced by Escherichia coli acting on other closely related bacterial strains by disrupting their outer cell membrane. This bacteriocin has huge potential as a potent antimicrobial agent, especially, since it was recognized by the FDA as safe. In this work, we present the expression of colicin M in model transgenic Nicotiana tabacum L. plants. We demonstrate that purified colicin retains its antibacterial activity against the control Escherichia coli strains and clinical isolates of Escherichia coli and Klebsiella pneumoniae. Our results also show that plant-derived ColM is not toxic for L929 and HeLa cell line, which allows us to suppose that plant-based expression could be an alternative production method of such important proteins.

        Grafizacja polszczyzny u progu ery typograficznej (lata 1521 i 1522) - między konwencją a kreacją

        Graphization besides intellectualization and standardization is one of the most crucial factors influencing developing of a standard language. Beginnings of the long-drawn-out process of graphization of Polish date back to year 1136. Rules of the adaptation of the Latin alphabet to the phonetics (phonology) requirements of the Polish language of the Old Polish era that ended in the beginning of the 16th century evolved and were characterized by a great degree of variation. Yet an appearance of the printed book, that was considered as a good for sale, forced into developing the graphic system being characterized by a possibly great degree of standardization. A spelling system, conceived by editors working for the Hieronim Wietor's printing house In years 1521 and 1522, on the threshold of the Polish typographical era, was a compromise between modern solutions (Latin letters modified with diactritics, e.g. ę, ą, ó, ś, ż, ł) and traditional solutions derived from manuscripts (digraphs, e.g. cz, dz, rz). The main ideas of the Hieronim Wietor's publishing house spelling system are in force until today.

        Chlorophyll a Fluorescence Parameters of Hulled and Hull-less Barley (Hordeum vulgare L.) DH Lines Inoculated with Fusarium culmorum

        Barley worldwide is affected seriously by Fusarium seedling blight (FSB) and Fusarium head blight (FHB) diseases caused by the Fusarium species. The objective of this study was to facilitate the resistance of hulled and hull-less barley at different growth stages to F. culmorum according to direct parameters: disease rating (DR), fresh weight of leaves and roots, kernel weight per spike, kernel number per spike, plump kernels, and indirect parameters - chlorophyll a fluorescence (CF). Plate assay, greenhouse and field tests were performed on 30 spring barley doubled haploid (DH) lines and their parents infected with Fusarium culmorum. Direct parameters proved that hulled genotypes show less symptoms. Most studied chlorophyll a fluorescence (CF) parameters (apart from DIo/CS – amount of energy dissipated from PSII for laboratory test, TRo/CS – amount of excitation energy trapped in PSII reaction centers, ETo/CS – amount of energy used for electron transport and RC/CS – number of active reaction centres in the state of fully reduced PSII reaction center in field experiment) were significantly affected by F. culmorum infection. In all experiments, hulled genotypes had higher values of CF parameters compared to hull-less ones. Significant correlations were detected between direct and indirect parameters and also between various environments. It was revealed that ABS/CS, TRo/CS, and RC/CS have significant positive correlation in greenhouse test and field experiment. Significant correlations suggest the possibility of applying the CF parameters in selection of barley DH lines resistant to F. culmorum infection.

        Polskie czasowniki wnioskowania – stan badań, hipotezy i perspektywy

        This work is intended by the author's part and also the prelude to a larger whole. Of concern in the article is the class of predicates inference, in particular a substantial part of it, namely verbs of inference – from the most representative for the whole of this class expression: [a] wywnioskował z [czegośp], że [q]. Inference predicates interest me primarily because of its importance: systemic (potential), synchronous (real) and local (Polish). Goal that I set myself (in the near term), is reduced to the reconstruction of the semantic representation of selected expressions (representatives of the class in question), and more precisely to formulate their dictionary definitions (lexicographical portraits) in terms of semantic components (as necessary conditions separately and jointly sufficient) The method by which I reach, meaning interesting question on my unit, part of a program of structuralist and refers to two more specific concepts: reducing component semantics and natural semantic metalanguage. Of course, this paper will confine itself to outline the general characteristics (pan) of that class and formulate a series of initial hypotheses, which deals with the verification of some time in the cycle of works devoted to particular verbs inference.

        Clostridium botulinum spores in Polish honey samples

        The aim of this study was an examination of 240 multifloral honey samples collected from Polish apiaries to determine Clostridium botulinum occurrence. Honey was collected from apiaries directly after the extraction process. Samples were inoculated by using the dilution and centrifugation method. Suspected isolates were examined by using mouse bioassay, polymerase chain reaction (PCR), and real-time PCR methods. C. botulinum type A and B strains were detected in 5 of 240 examined honey samples (2.1%). Bacterial strains were also detected that were phenotypically similar to C. botulinum but that did not exhibit the ability to produce botulinum toxins and did not show the presence of the botulinum cluster (ntnh and bont genes) or expression of the ntnh gene. The methods used in the examination, especially the expression analysis of ntnh gene, enabled specific analysis of suspected strains and could be used routinely in environmental isolate analyses of C. botulinum occurrence.

      • New Current Controller for Inverter Fed Medium Voltage Drives with LC Filter

        In high power medium voltage drives, the switching frequency of power semiconductor devices is restricted because of high switching losses. Such drives often include an LC output filter, introducing a resonant circuit that may be excited for instance by fast control transients. One way to avoid such oscillations is using damping control schemes, which in turn usually require switching frequencies well above the filter resonance. This paper proposes a new predictive stator current controller that avoids the excitation of the filter resonance, performs active damping and allows fast current control while maintaining low switching frequency. Simulation results of a 2.4 ㎸ induction motor drive and experimental results obtained from a 55 ㎾ prototype a.c. drive demonstrate feasibility and good dynamic performance of the proposed control approach.

        Application of chloroplast promoters of Cyanidioschyzon merolae for exogenous protein expression

        The ability to transform the chloroplast of Cyanidioschyzon merolae was limited by lack of confirmed and reliablepromoter sequences (among other reasons), capable of delivering stable or modulated DNA transcription followed byprotein synthesis. Our research has confirmed the applicability of three selected chloroplast promoters in C. merolaechloroplast overexpression of the exogenous protein (i.e., chloramphenicol acetyltransferase) and genetic transformation. These results might facilitate further research on genetically modified strains of C. merolae to envisage yet unknownaspect of cellular and plastic physiology as well as C. merolae potential applications as bio-factories or sources of usefulchemicals.
