        Increased Cysteine Accumulation is Essential for Salt Stress Tolerance in Arabidopsis Halotolerance 2-Like (AHL)-Overexpressing Transgenic Plants

        Cysteine (Cys) regulates plant growth, development, and various abiotic stress responses. However, the knowledge gained from genetic and molecular studies on the role of Cys metabolism in salinity response remains limited. Here, we introduce a sulfate metabolism-related component, Arabidopsis Halotolerance 2-like (AHL), which is involved in salt stress adaptation. AHL expression was experimentally induced under salt stress conditions using AHL promoter-β-glucuronidase transgenic plants. Phenotypic analysis revealed that the survival rate of AHL-overexpressing (AHL-OE) lines was greater than that of wild-type (WT) plants under high-salinity conditions, while ahl mutant and AHL-RNAi seedlings were more sensitive than WT seedlings. Accumulation of Cys and proline was increased in AHL-OE transgenic lines under salinity stress conditions, but malondialdehyde and hydrogen peroxide levels were lower in AHL-OE than in WT, ahl, and AHL-RNAi plants. In addition, the transcription levels of genes associated with Cys biosynthesis and sulfate metabolism were higher in AHL-OE than ahl and AHL-RNAi seedlings after salt stress treatment. Moreover, exogenous application of Cys could rescue the salt stress-induced sensitive phenotypes of ahl and AHL-RNAi lines, and return them to the WT phenotype. Taken together, our findings indicate that AHL positively regulates salinity response in Arabidopsis seedlings by modulating a Cys-dependent pathway.

        A novel meshfree model for buckling and vibration analysis of rectangular orthotropic plates

        The present work mainly reports a significant development of a novel efficient meshfree method for vibration and buckling analysis of orthotropic plates. The plate theory with orthotropic materials is followed the Kirchhoff’s assumption in which the only deflection is field variable and approximated by the moving Kriging interpolation approach, a new technique used for constructing the shape functions. The moving Kriging technique holds the Kronecker delta property, thus it makes the method efficiently in imposing the essential boundary conditions and no special techniques are required. Assessment of numerical results is to accurately illustrate the applicability and the effectiveness of the proposed method in the class of eigenvalue problems.

        Genetic parameters and litter trait trends of Danish pigs in South Vietnam

        Objective: The objective of this study was to estimate the genetic parameters and various litter trait trends of Danish pigs in South Vietnam, including the number born alive (NBA), number weaned (NW), and litter weight at the 21st day (LW21). Methods: Records of 936 Yorkshire sows with 3361 litters and 973 Landrace sows with 3161 litters were used to estimate the variance components, genetic parameters, and trends of NBA, NW, and LW21. The restricted maximum likelihood method was applied using VCE6 software to obtain the variance components and genetic parameters. Thereafter, the best linear unbiased prediction procedure with an animal model was applied using PEST software to estimate the breeding values of the studied traits. Results: The heritability estimates were low, ranging from 0.12 to 0.21 for NBA, 0.03 to 0.04 for NW, and from 0.11 to 0.13 for LW21. The genetic correlation between the NBA and NW was relatively strong in both breeds, at 0.77 and 0.60 for Yorkshire and Landrace, respectively. Similarly, the genetic correlation between the NW and LW21 was considerably stronger in Landrace pigs (0.71) than in Yorkshire pigs (0.48). The estimates of annual genetic progress were 0.0431, 0.0233, and 0.0461 for NBA, NW, and LW21 in Landrace pigs and 0434, 0.0202, and 0.0667 for NBA, NW, and LW21 in Yorkshire pigs, respectively. Conclusion: The positive genetic trends estimated for the additive genetic values of the selected traits indicated that the current breeding system has achieved favorable results.         Original Articles : Abalone Haliotis discus hannai Intestine Digests with Different Molecule Weights Inhibit MMP-2 and MMP-9 Expression in Human Fibrosarcoma Cells

        The abalone Haliotis discus hannai, is one of the economically important species in the fisheries industry. Abalone intestines are one of the by-products of its processing. To investigate its bioactive potential, abalone intestine was digested using an in vitro gastrointestinal (GI) digestion system containing pepsin, trypsin, and α-chymotrypsin. The abalone intestine G1 digests (AIGIDs) produced by the GI digestion system were fractionated into AIGID I (> 100kDa), AIGID II (10-100kDa), and AIGID III (1-10kDa) using an ultrafiltration membrane system. Of the three digests, AIGID II and AIGID III exhibited inhibitory effects against matrix metalloproteinase-2 and -9 (MMP-2, MMP-9) in HT1080 human fibrosarcoma cells. Both fractions potently inhibited gelatine digestion by MMP-2 and MMP-9 treated with phorbol 12-myristate 13-acetate (PMA) and migration of HT1080 cells in dose dependently. Furthermore, AIGID II and III attenuated expression of p65, a component of nuclear transcription factor kappa B. These results indicate that of the abalone intestine digests inhibit MMP-2 and MMP-9. Thus, the AIGIDs or their active components may have preventive and therapeutic potential for diseases associated with MMP-2 and MMP-9 activation in fibrosarcoma cells.

      • Bubble bursting in bioreactor for animal cell culture

        Due to absence of cell wall structures, animal cells are very sensitive to extensional stress that has great influence on cell damage. Understanding the bubble bursting mechanism at free surface, including maximum extensional stress generation and bubble diameter, is a key in bioreactor systems. The maximum extensional stress must be smaller than the critical one for a given animal cell type. This study will carry out the numerical investigation of bubble bursting process on the free surface, including extensional stress generation, in considering physical and geometric parameters: bubble diameter, surface tension. The coupling between volume of fluid and level-set methods is employed to descript the evolution of gas-water interfaces.

        Effects of Psidium guajava Leaf Extract on Apoptosis Induction Through Mitochondrial Dysfunction in HepG2 Cells

        The anticancer activity of guava (Psidium guajava L.) leaf extract (GLE) occurs via the induction of apoptosis in cancer cells. However, the mechanism behind GLE-induced apoptosis in the human hepatocellular carcinoma cell line HepG2 remains unclear. In the present study, we investigated the apoptotic effects and mechanism of action of GLE in cultured HepG2 cells. The results showed that GLE induced reactive oxygen species (ROS) synthesis and disrupted the mitochondrial membrane potential (ΔΨm). Moreover, GLE increased the expression of apoptotic pathway proteins, such as the cleaved forms of caspase-3, -8, and -9; the translocation of Bax and cytochrome c (cyt-c) from the mitochondria to the cytosol; and the downregulation of Bcl-2. In addition, p53 protein expression was increased upon GLE treatment. These observations indicate that the GLE-induced apoptosis in HepG2 cells is mediated by mitochondrial ROS generation, followed by caspase activation and cyt-c release, suggesting that GLE may be a promising candidate for the development of novel drugs for the treatment of liver cancers.

        A Finite-time Adaptive Fault Tolerant Control Method for a Robotic Manipulator in Task-space With Dead Zone, and Actuator Faults

        This paper expresses a novel adaptive fault-tolerant control (FTC) method for a robotic manipulator with dead zones and actuator faults in the task space. To specify, the actuator faults are the partial loss of effectiveness of actuators. The partial loss of effectiveness and the break-points of dead zone are estimated by the novel adaptive laws. Establishing an anti-singularity terminal sliding surface ensures the finite time convergence of the tracking errors to zero. The correctness of this method is proved by the Lyapunov criterion. Despite the fact that the proposed adaptive FTC method belongs to the passive group, it does not require any robust component which makes largemagnitude chattering phenomena to deal with the actuator faults. Instead, they are handled by a unique technique. The comparative simulation results illustrate the merit and efficiency of the proposed method.

        Effect of pre-tensioned loads to vibration at the ladder tip in raising and lowering processes on a turntable ladder

        A turntable ladder is an indispensable vehicle in fire and rescue work. Vibration on the top of the ladder is considerable when the ladder set is maximally extended. It induces the discomfort and unsafety for beings in the rescue cage. Aiming at the reduction of the vibration, in this study, one additional steel wire rope is assumedly added into each handrail of the ladder set. The steel wire ropes are suggested to be pre-tensioned before the turntable ladder works. This study is based on an available ladder set structure which includes three ladder sections with different area moments of inertia. Additionally, a ladder structure model is established as a multibody system dynamic model. The changes of kinetic parameters in the raising and lowering processes of the ladder set are investigated. The obtained results can be the fundamentals for modeling, experiments and applications of turntable ladders.
