Characterization and expression analysis of dirigent family genes related to stresses in Brassica

The dirigent (DIR) genes are playing a vital role in enhancing stress resistance in different crop plants. In this study, we collected 29 DIR like genes, two from a Brassica rapa cv. Osome full length cDNA library and 27 from the B. rapa database designated as B. rapa Dirigent (BrDIR) like genes. Sequence analysis and a comparison study of these genes confirmed that seven were dirigent and the remaining 22 were dirigent like genes. Expression analysis revealed an organ specific expression of these genes. BrDIR2 showed differential responses after Fusarium oxysporum f.sp. conglutinans infection in cabbage. Four Brassica oleracea dirigent like genes highly homologous to BrDIR2 also showed similar responses in cabbage plants infected with this fungus. Moreover, several BrDIR like genes showed significant responses after water, ABA and cold stress treatments in Chinese cabbage. Under water stress, most responsive genes showed the highest expression at 24 h, at which time the acid soluble lignin content of samples under the same stress condition were also highest, indicating a possible relationship between BrDIR like genes and lignin content. Taken together, our results indicate a protective role of BrDIR genes against biotic and abiotic stresses in Brassica.
Antimicrobial and lipid peroxidation inhibition activity of Oxystelma esculentum (Asclepiadaceae)

The aerial parts of methanol extract of Oxystelma esculentum (MEOE) (Asclepiadaceae) was evaluated for in vitro lipid peroxidation and antimicrobial activity. Lipid peroxidation was assayed by the change in optical density of the various concentrations (20 - 320 µg/ml) and the percentage inhibition was estimated. Ascorbate/FeSO4-induced peroxidation was inhibited by MEOE and standard antioxidants such as BHA, BHT and the percentage inhibition of the methanol extract was increased with dose dependent manner. The IC50 value of the MEOE, BHA and BHT for lipid peroxidation was found to be 135.24 µg/ml, 25.62 µg/ml and 17.13 µg/ml, respectively. The antimicrobial activity of MEOE was determined by disc diffusion method with three gram-positive, five gram-negative and two fungal microorganisms. MEOE exhibited the antimicrobial activity against the tested microorganisms except Salmonella typhimurium (MTCC 98). In present study, it is concluded that MEOE has significant effect in the inhibition of lipid peroxidation and possesses good antimicrobial activity.

The oxidative decarboxylation of phenylsulfinylacetic acid (PSAA) by Cr(VI) in 20% acetonitrile -80% water (v/v) medium follows overall second order kinetics, first order each with respect to [PSAA] and [Cr(VI)] at constant [$H^+$] and ionic strength. The reaction is acid catalysed, the order with respect to [$H^+$] is unity and the active oxidizing species is found to be $HCrO_3^+$. The reaction mechanism involves the rate determining nucleophilic attack of sulfur atom of PSAA on chromium of $HCrO_3^+$ forming a sulfonium ion intermediate. The intermediate then undergoes ${\alpha}$,${\beta}$-cleavage leading to the liberation of $CO_2$. The product of the reaction is found to be methyl phenyl sulfone. The operation of substituent effect shows that PSAA containing electron-releasing groups in the meta- and para-positions accelerate the reaction rate while electron withdrawing groups retard the rate. An excellent correlation is found to exist between log $k_2$ and Hammett ${\sigma}$ constants with a negative value of reaction constant. The ${\rho}$ value decreases with increase in temperature evidencing the high reactivity and low selectivity in the case of substituted PSAAs.

In the Indian traditional medicine, Lippia nodiflora (Verbenaceae) whole plant is claimed to possess powerful diuretic activity. However, the diuretic potential of this plant is not yet investigated. The aim of this study was to evaluate the diuretic potential of methanol extract of Lippia nodiflora (MELN) in rats. Control (0.9% saline solution, 25 ml/kg, b.w) or urea (1 g/kg b.w) or frusemide (5 mg/kg b.w) and different concentrations of MELN (200 and 400 mg/kg b.w) were intraperitoneally administered (n = 6 per each treatment group) to hydrated rats and their urine output was monitored over a period of 5 h and 24 h after drug administration. The diuretic responses with its electrolyte excretion potency of the extract were highly remarkable in comparison with control animals. The extract at doses of 200 and 400 mg/kg shows a significant increase in volume of urine with increase in $Na^{+}$, $Ca^{2+}$ and $Cl^{-}$ excretion accompanied by the excretion of $K^{+}$ in dose dependent manner. This study suggests that the active component(s) in MELN had similar diuretic effect to that of frusemide. These results validate the traditional use of Lippia nodiflora as a diuretic agent.

Parkinson’s disease (PD) is the second most common neurodegenerative disorder. It is usually seen in those above 50 years old. Current medical treatments only provide symptomatic relief but cannot cure the disease. There are claims that PD can be cured by stem cell transplant. The present study is aimed to assess the clinical potency and safety of stem cell in treating PD. A total of eleven articles were included for analysis, with four randomised control trials (RCTs), five non-RCTs and 2 follow up studies. All the four non-RCTs showed improvement of Unified Parkinson’s Disease Rating Scale with no adverse events. However, results from RCTs showed no significant differences in the rating score among the transplant group and the Sham surgery group. The secondary analysis of one study showed a significant improvement of the rating score in those patients aged 60 and younger. Transplant group also associated with an overall higher incidence of adverse events. In conclusion, the RCTs and non-RCTs produced opposite results. When the studies were performed as non-RCTs in small number of patients, they showed promising result in the patients. It could say that currently the use of stem cell/progenitor cells in treating PD need much research despite having the implanted stem cell to be able to survive and integrated. The survival of implanted dopamine neurons in the striatum, however, does not indicate a success in correcting PD symptoms. Further investigations will shed light on the application and mechanism of action of stem cells in treating PD.

The methanol extract of the root of Artanema sesamoides Family Scrophuilariaceae (MEAS) was investigated for possible analgesic and anti-inflammatory effects in animals. Three models were used to study the extract effects on nociception, which were acetic acid-induced writhing response, hot-plate method and the tail flick test in mice. The antiinflammatory effects were evaluated using carrageenan, dextran, histamine and serotonin induced rat paw oedema (acute) and cotton pellet induced granuloma (chronic) models in rats. Results of the study revealed that the extract exhibited significant (P < 0.001) analgesic effect at a dose of 50, 100 and 200 mg/kg b.w p.o in mice in all the models. In acute model, the MEAS also exhibited significant (P < 0.001) antiinflammatory effect in all the above mentioned doses. In chronic model (cotton pellet induced granuloma) the MEAS 200 mg/kg and indomethacin 10 mg/kg showed that inhibition of granuloma formation 25.0% and 47.7% respectively (P < 0.001). The MEAS and indomethacin were effectively preventing the transudation of the fluid. Thus, the present study revealed that the methanol extract of the root of Artanema sesamoides exhibited significant analgesic and antiinflammatory activity.
