Absence of an Essential Thiol in Human Glutaminyl Cyclase: Implications for Mechanism

We have partially sequenced glutaminyl cyclases from several mammalian and one avian species and found that the two cysteine residues of the human glutaminyl cyclase are completely conserved. The mammalian glutaminyl cyclase has been reported to possess reactive thiols (Busby, Jr, et aI., 1987, J BioI Chern 262, 8532-8536). Mutagenesis of these cysteine residues, however, resulted in only a slight decrease in enzyme activity. Likewise, the recombinant human enzyme was completely resistant to attempted chemical modification of the putative reactive thiols. Although the human glutaminyl cyclase did not appear to have reactive thiols, it was sensitive to diethylpyrocarbonate and acetylimidazole, indicating the presence of functionally important histidine and tyrosine residues which could act as acid/base catalysts. Almost identical deuterium solvent isotope effect (1.2 vs 1.3) upon the reaction by the human and papaya enzymes, respectively, provides an evidence both animal and plant glutaminyl cyclases catalyze pyroglutamyl-peptide formation by intramolecular cyclization.

Protecting the age-old culture and body of knowledge of indigenous people has become a topical issue. Three options are usually canvassed for such — a typical western intellectual model, a typical customary law model and a sui generis model. More often than not, these options still mirror intellectual property paradigms and the reason is not far-fetched. Intellectual property generally has to do with property rights, innovation and ownership. Hence, indigenous people’s claim to ownership of their knowledge and innovations are usually associated with the canons of intellectual property. However, several views and arguments abound on the practicability of protecting indigenous knowledge with intellectual property principles. The literature addresses themes such as catering for the spiritual elements of indigenous knowledge, originality requirement, public domain concerns, among others. Instructively, little or nothing has been done on analyzing the philosophical undertone behind these arguments. By way of contributing to knowledge, this paper presents five underlying perspectives that frame divergent views in the debate on protecting indigenous knowledge — subjective descriptions of property, views on authorship, views of cultural orthodoxy versus cultural pluralism, Vermeylen’s legalistic and anthropological approaches and optimism of the modern era. Like a matrix, set of theoretical principles and founding philosophies of thoughts, these inter-related perspectives directly and inadvertently shape various articulations, propositions and deliberations on the subject. This paper engages this discourse outlining precisely what these underlying perspectives are, whilst also noting their relative strength and flaws. It is expected that this will present future researchers with a clear philosophical categorization of arguments in the debate on indigenous knowledge protection.

This study analyzed the presence and effect of sexual appeal of plus-size models using Victoria's Secret's Instagram content. A content analysis using roBERTa deep-learning model found that, plus-size models’ sexual appeals increased number of likes and comments, but explicit sexual appeals resulted in less positive sentiments than thin models’.
Application of Wavelet-Based RF Fingerprinting to Enhance Wireless Network Security

This work continues a trend of developments aimed at exploiting the physical layer of the open systems interconnection (OSI) model to enhance wireless network security. The goal is to augment activity occurring across other OSI layers and provide improved safeguards against unauthorized access. Relative to intrusion detection and anti-spoofing, this paper provides details for a proof-of-concept investigation involving "air monitor" applications where physical equipment constraints are not overly restrictive. In this case, RF fingerprinting is emerging as a viable security measure for providing device-specific identification (manufacturer, model, and/or serial number). RF fingerprint features can be extracted from various regions of collected bursts, the detection of which has been extensively researched. Given reliable burst detection, the near-term challenge is to find robust fingerprint features to improve device distinguishability. This is addressed here using wavelet domain (WD) RF fingerprinting based on dual-tree complex wavelet transform (DT-$\mathbb{C}WT$) features extracted from the non-transient preamble response of OFDM-based 802.11a signals. Intra-manufacturer classification performance is evaluated using four like-model Cisco devices with dissimilar serial numbers. WD fingerprinting effectiveness is demonstrated using Fisher-based multiple discriminant analysis (MDA) with maximum likelihood (ML) classification. The effects of varying channel SNR, burst detection error and dissimilar SNRs for MDA/ML training and classification are considered. Relative to time domain (TD) RF fingerprinting, WD fingerprinting with DT-$\mathbb{C}WT$ features emerged as the superior alternative for all scenarios at SNRs below 20 dB while achieving performance gains of up to 8 dB at 80% classification accuracy.
