        Molecular cloning and characterization of novel human JNK2 (MAPK9) transcript variants that show different stimulation activities on AP-1

        The c-Jun NH2-terminal kinase (JNK) signaling pathway participates in many physiological functions. In the current study we reported the cloning and characterization of five novel JNK2 transcript variants, which were designated as JNK2 α3, JNK2 α4, JNK2 β3, JNK2 γ1 and JNK2 γ2, respectively. Among them, JNK2 α4 and JNK2 γ2 are potential non-coding RNA because they contain pre-mature stop codons. Both JNK2α3 and JNK2β3 contain an intact kinase domain, and both encode a protein product of 46 kDa, the same as those of JNK2α1 and JNK2β1. JNK2γ1 contains a disrupted kinase domain and it showed a disable function. When over-expressed in mammalian cells, JNK2α3 showed higher activity on AP-1 than that of JNK2β3 and JNK2γ1. Furthermore, JNK2α3 and JNK2β3 showed different levels of substrate phosphorylation, although they both could promote the proliferation of 293T cells. Our results further demonstrate that JNK2 isoforms preferentially target different substrates and may regulate the expression of various target genes. [BMB reports 2010; 43(11): 738-743]

        Human HS1BP3 induces cell apoptosis and activates AP-1

        In the present study, we characterized the function of HS1-binding protein 3 (HS1BP3), which is mutated in essential tremor and may be involved in lymphocyte activation. We found that HS1BP3 localized to the mitochondria and endoplasmic reticulum partially. Overexpression of HS1BP3 induced apoptosis in HEK293T and HeLa cell lines. When these cell lines were transfected with HS1BP3, they exhibited nuclear DNA condensation, externalization of phosphatidylserine (PS), and cleavage of poly ADP ribose polymerase (PARP). Furthermore, suppression of HS1BP3 or HS1 expression attenuates HS1BP3 induced apoptosis. In addition, HS1BP3 enhanced activator protein 1 (AP-1)-mediated transcription in a dose-dependent manner. Therefore, we conclude that HS1BP3 regulates apoptosis via HS1 and stimulates AP-1-mediated transcription. [BMB reports 2011; 44(6): 381-386]

      • Effect of cation ratio and order on magnetic circular dichroism in the double perovskite Sr<sub>2</sub>Fe<sub>1+</sub> <i> <sub>x</sub> </i>Re<sub>1-</sub> <i> <sub>x</sub> </i>O<sub>6</sub>

        <P><B>Abstract</B></P> <P>Superexchange-based magnetic coupling of the two <I>B</I>-site cations in rock-salt-ordered double perovskite oxides is extremely sensitive to the cation ratio and degree of order. However, as a result of the limited spatial resolution of most magnetic characterization techniques, it is challenging to establish a direct relationship between magnetic properties and structure in these materials, including the effects of elemental segregation and cation disorder. Here, we use electron energy-loss magnetic chiral dichroism together with aberration-corrected electron microscopy and spectroscopy to record magnetic circular dichroism (MCD) spectra at the nm scale, in combination with structural and chemical information at the atomic scale from the very same region. We study nanoscale phases in ordered Sr<SUB>2</SUB>[Fe][Re]O<SUB>6</SUB>, ordered Sr<SUB>2</SUB>[Fe][Fe<SUB>1/5</SUB>Re<SUB>4/5</SUB>]O<SUB>6</SUB> and disordered Sr[Fe<SUB>4/5</SUB>Re<SUB>1/5</SUB>]O<SUB>3</SUB> individually, in order to understand the role of cation ratio and order on local magnetic coupling. When compared with ordered Sr<SUB>2</SUB>[Fe][Re]O<SUB>6</SUB>, we find that antiferromagnetic Fe<SUP>3+</SUP>-O<SUP>2−</SUP>-Fe<SUP>3+</SUP>superexchange interactions arising from an excess of Fe suppress the MCD signal from Fe cations in ordered Sr<SUB>2</SUB>[Fe][Fe<SUB>1/5</SUB>Re<SUB>4/5</SUB>]O<SUB>6</SUB>, while dominant Fe<SUP>3+</SUP>-O<SUP>2−</SUP>-Fe<SUP>3+</SUP>antiferromagnetic coupling in disordered Sr[Fe<SUB>4/5</SUB>Re<SUB>1/5</SUB>]O<SUB>3</SUB> leads to a decrease in MCD signal down to the noise level.       • Control of Cascaded H-Bridges STATCOM based on the Power Flow Management

        As smart grid techniques are getting more positive perspective and expected to grow in the future, the STATic synchronous COMpensator (STATCOM) has become one of the most important roles to manage the power flow and regulate the grid voltage. The Modular Multilevel Cascaded Converter (MMCC) based on single star bridge cell (SSBC) is very suitable for STATCOM application at the medium-voltage level in the power system. The dc capacitors voltage balancing control is a critical issue to secure the stable operation of MMCCSSBC converter. Four different control methods based on power flow analysis have been considered. The performance of each controller is evaluated under steady state or even Low Voltage Ride Through (LVRT) operation. Each controller has its power flow management and its feature. Experimental test results are given to verify these control methods can well regulated the dc bus voltages under the unbalanced grid voltages.

        Surface Plasmon Resonance in Metallic Nanocylinder Array

        Localized surface plasmon resonances of noble-metal nanoparticles lead to high enhancements of local electromagnetic fields near the nanoparticle surface and play a key role in surface-enhanced Raman scattering, surface-enhanced fluorescence, near-field microscopy, and plasmonic devices. Recently, research results have shown that electromagnetic energy can transfer along a metallic nanoparticle chain because of coupled plasmon modes. In this paper, finite-difference time-domain method was used to study the surface plasmon resonances in coupled silver nanocylinder (nanoparticle) arrays and slab-like periodic modulation structures. From the far-field transmittance and reflectance, three different resonance modes were excited for the periodic nanocylinder arrays and only one resonance was excited for the periodic modulation structure. The near-field intensity distributions exhibited highly enhanced fields at the center of the gaps for nanoparticle arrays and at the caves of the periodic modulation structure.

        Can herbal extracts be used as skin penetrating agent

        Swollen and painful extremities have been treated with herbal baths in attempts to alleviate the symptoms. A popular herbal bath used in China, contains a component called ‘bone-penetrating herb’ which is believed to facilitate the penetration of herbal substances across the skin to the swollen site, so that pain and swelling will be improved. A search from the Chinese pharmacopedia revealed that 22 different herbs have been traditionally used as ‘bone-penetrating herb’. Five of these herbs were available in market and were chosen for experimental studies. Standard diffusion experiments were done to identify the most effective herb among the five, in the penetration facilitation. Glechoma longituba at a concentration of 20% was found to give the best results in the facilitation of Bromophenol blue diffusion across artificial and biological membranes. When compared with one commonly used diffusion facilitator, viz. azone, azone was found to be more effective than glechoma longituba. The encouraging observations support future studies on the basic science behind the use of herbal components as topical agents to treat pain and swelling.

        Research on Traditional Chinese Medicine harmonising two approaches

        While full recognition of the practical value of Traditional Chinese Medicine is being endorsed, the current stand on the research methodology of this field should be worked out. Since modern medicine has already developed a logical system of research methodology basing on the principles of deduction, any research on any system of medicine need to take reference to what is most popularly used and commonly recommended. The best way to approach research on Chinese Medicine, therefore, would be one that would take full reference to the methodology being used in modern medicine, while at the same time respecting the traditional approach. This would enable traditional medicine to be elevated to the level of general modern recognition. Nevertheless, innate problems in traditional medicine are making its research difficult. The problems lie in difficulties to achieve uniform herb supply, principles of randomization and placebo arrangements, uncertain chemical structures and toxicology etc. A practical approach centered on carefully planned evidence-based clinical trials, with parallel studies on biological activities and herb authentication is being recommended.
