Enhancing the Enzymatic Activity of the Multifunctional β-Glycosyl Hydrolase (Cel44C-Man26A_P558) from Paenibacillus polymyxa GS01 Using DNA Shuffling

본 연구자들은 이전에 cellulase, xyalnase 및 lichenase의 다기능 효소활성을 지니는 절단된 Cel44C-$Man26A_{P558}$의 ${\beta}$-glycosyl hydrolase를 보고하였다. 본 연구에서는 절단된 Cel44C-$Man26A_{P558}$ 효소의 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키기 위해 DNA shuffling을 시도하였다. DNA shuffling에 의해 단일변이(P438A)를 가진 M2Cel44C-$Man26A_{P558}$와 이중변이(A273T 및 P438A)를 가진 M21Cel44C-$Man26A_{P558}$를 얻었다. 이중변이를 가진 M21Cel44C-$Man26A_{P558}$은 단일변이를 가진 M2Cel44C-$Man26A_{P558}$ 보다 효소활성이 낮게 나타났으나, M2Cel44C-$Man26A_{P558}$와 M21Cel44C-$Man26A_{P558}$은 대조구인 Cel44C-$Man26A_{P558}$ 보다 약 1.3에서 2.2배 정도 높은 효소활성을 나타내었다. 특히, 단일변이를 가진 M2Cel44C-$Man26A_{P558}$는 대조구인 Cel44C-$Man26A_{P558}$보다 cellulase, xylanase 및 lichenase 효소활성이 약 1.5에서 2.2배 정도 높게 나타났다. ${\beta}$-Glycosyl hydrolase의 cellulase, linchenase 및 xylanase 최적 효소활성은 각각 pH 7.0, 7.0 및 6.0에서 이었다. 이러한 결과는, 아미노산 잔기인 Ala438이 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키는 중요한 역할을 한다고 추정할 수 있다. We previously reported that the truncated Cel44C-$Man26A_{P558}$ ${\beta}$-glycosyl hydrolase protein exhibits multifunctional activities, including cellulase, xylanase, and lichenase. DNA shuffling of the truncated Cel44C-$Man26A_{P558}$ enzyme was performed to enhance the enzymatic activity of the multifunctional ${\beta}$-glycosyl hydrolase. Two mutant enzymes, M2Cel44C-$Man26A_{P558}$ that carries one mutation (P438A) and M21Cel44C-$Man26A_{P558}$ that carries two mutations (A273T and P438A) were obtained. The enzymatic activity of the M21Cel44C-$Man26A_{P558}$ double mutant was lower than enzymatic activity of the single mutant (M2Cel44C-$Man26A_{P558}$). However, both mutants displayed the enhancements in their enzyme activities that were ${\approx}1.3$- to 2.2-fold higher than the original enzymatic activity in Cel44C-$Man26A_{P558}$. In particular, the mutant M2Cel44C-$Man26A_{P558}$ exhibited an approximate 1.5- to 2.2-fold increase in the cellulase, xylanase, and lichenase activities in comparison with the control (Cel44C-$Man26A_{P558}$). The optimum cellulase, linchenase, and xylanase activities of ${\beta}$-glycosyl hydrolase were observed at pH 7.0, pH 7.0 and pH 6.0, respectively. These results, therefore, suggest that the amino acid residue Ala438 plays important roles in the enhancement of the activity of multifunctional ${\beta}$-glycosyl hydrolase.

A Study on the Functionality of Halophytes from Coast area of Jeollanam-do

This study was undertaken to improve the perceived value of halophytes and contribute to the activity of the local economy by analyzing the antioxidant activity and mineral content of Suaeda japonica, Tetragonia tetragonoides, and Spergularia marina that grow wild on the mud flats and coastal areas. The total phenolic compound content found in each order is as follows: Suaeda japonica (3.674∼5.989 mg TAE/g dry wt.), Tetragonia tetragonoides (2.515∼4.471 mg TAE/g dry wt.), and Spergularia marina (2.450∼4.067 mg TAE/g dry wt.); DPPH radical scavenging and ABTS cation radical scavenging activities were also recorded in the same order. In the case of minerals, Iron(Fe) was relatively high in Suaeda japonica (960.1∼1,346.7 mg/kg), Manganese(Mn) in Tetragonia tetragonoides (197.1∼574.8 mg/kg), and Zinc(Zn) was relatively high in Spergularia marina (63.19∼216.56 mg/kg). It was confirmed that the mineral content of Suaeda japonica, Tetragonia tetragonoides, and Spergularia marina was significantly higher than that of Chinese cabbage, lettuce, leek, and spinach.

The purpose of this study was to examine effects of customers satisfaction on purchase behavior intention in commercial sports center. The subjects in this study was set up as 492 male and female who are above 20 years old in using sports club located in Busan city in the year of 2001, and sampling by using systematic stratified cluster random sampling. The means in this study is used to amend and complemented after confirming suitability of the contents in the questionnaire that is made on the basis of the questionnaire of Japan Fitness Industry Association(1997)and Cho, Man-Tae, Kim, Sang-dai(2000), Lee, Yong-ki, Shin, Doo-chul, Lryu-Cheol(2000). it is used as study means after being modified, whether the substance of question is right in fitness and applicability or not. Reliability of the question, the range of Cronbach`s a is showed from.7981 to.8886. The statistics analysis method adapted Pearson`s Correlation Analysis and Enter Multiple Regression. This conclusion of this study are as follows. First, perceived customer satisfaction factor effected on word-of-mouse intention. Specially, exercise effect, customer management, response in the front desk, facilities management. Second, perceived customer satisfaction factor effected on repurchase intention. Specially, teaching staffer, customer management, lives of club, response in the front desk.

Digital Converter로 구동되는 크레인용 직류전동기 속도제어 시스템의 안정도 해석

크레인 구동용으로 antiparallel converter의 반도체 전력회로에 마이크로 제어기를 사용한 직류전동기의 속도제어 시스템에 대한 안정영역을 해석한다. 먼저 불연속 전류인 경우 converter의 비선형특성으로 인하여 전동기 속도제어 능력이 저하되므로 이를 보상하는 방식을 제안한다. 이 converter와 직류전동기를 포함한 전체 속도제어 시스템을 이산형 상태방적식으로 모델링 한 후, 각 제어기 이득에 대한 안정영역을 해석한다. The stability regions for microcontroller-based dc motor speed control system driven by antiparallel converter, which has been used as the crane drive, are analyzed, first as the performance of motor speed control is deteriorated due to nonlinear characteristics of the converter in the discontinous current state, the method to compense for nonlinear characteristics of the converter is suggested. The overall dc motor speed control system including the converter and dc motor is represented by a discrete-time state equation, and then the stability regions for gains of controllers are dervied.

The purpose of this study was to investigate the Influence of Information searching path on Determination for Exercise Continue Opinion of Sports Center members. The subject of this study were 249 individuals who were selected using the systemic stratified cluster sampling the sports center member in the city of Pusan. The data were collected from a self-administrated questionnaires, statistics methods employed in this analysis were Cronbach's α, T-test, F-test, Pearson's Correlation Analysis, Stepwise Multiple Regression. The Conclusion of this study are as follows : First, Most common ways to get sport information search were by magazines dealing sports articles and articles written by prestigious sports analysts. Second, Determination of exercise continue opinion of sports center male participants is higher than for female participants. Third, Determination of exercise continue opinion in men's case was high on man who incomes bellow 1,000,000 a month, by watching magazine advertisement and who incomes over 4,000,001 a month, by consulting family or relative.

해양스포츠 참여자의 성격특성에 관한 연구

The purpose of this study was to investigate what difference is between the character of participants in marine leisure sports and that of normal non-participants. 332 persons including 84 male non-participants and 76 female adults as well as 90 male members and 82 female members over 20 in their age belonging to the marine leisure sports dub in Pusan as of 1999 are selected as purposive sampling in this study. Questionnaires concerning character factor edited by professors Tae-ho Yeom and Jung - kyou Kim in Korea Psychology Aptitude Research Institute are used as tool for test, statistics methods employed in this analysis were t-test, F-test. The findings of this study are as follows First, As a result of comparative analysis on their character of participants in marine leisure sports and non-participants, it can be seen that an impulse, the liability of the accused were found to have a significant different in the male. executive faculty and strain, the liability of the accused were found to have a significant different in the female. Second, As a result of comparative analysis on the character features of participants in marine leisure sports and non-participants in accordance with their level, it can be seen that boldness, the sense for reality and self-dependence were found to have a significant different.

중학교 남자 테니스 선수의 운동기능수준별 시합 전 경쟁상태불안

The purpose of this study was to investigate if there is any difference in the level of competitive state anxiety according to the level of motor skill before the game and to provide the players with basic data for the mental practice. The subjects of this study consist of 20 men's a middle school tennis players. The experimental group was divided into superior players group and inferior players group. And then by using a CSAI-2 questionnaire, three subfactors of competitive state anxiety, or cognitive state anxiety, somatic state anxiety and state self-confidence were measured. The method of measurement was carried out three times, or one week, one day and two hours before the game. The results of this study were as follows. First, About both cognitive state anxiety and somatic state anxiety, inferior players were significantly higher than superior players. Second, About state self-confidence, superior players were significantly higher than inferior players. Third, As the game was around, both cognitive state anxiety and somatic state anxiety which all the players including superior and inferior players had increased. But the state self-confidence of all players decreased.

Tracheobronchopathia osteochondroplastica (TO) is a rare benign disease of unknown etiology characterized by accumulation of calcium phosphate in the submucosa of large airways and benign proliferation of bone and cartilage resulting in nodular formation. We report a case of tracheobronchopathia osteochondroplastica diagnosed by Endobronchial ultrasonography in a 56-year-old man. Chest Computed Tomography revealed thickening of tracheal and bronchial wall, and multiple nodules through whole trachea. Endobronchial ultrasonography showed numerous submucosal nodules with hetero-echogenecity in the third and fourth layers. Histopathological examination revealed nonspecific bronchitis with squamous metaplasia and metaplastic ossification. We confirmed tracheobronchopathia osteochondroplastica. The patient's symptoms were successfully treated with antibiotics and oxygen supplyment. endobronchial ultrasonography can helpful diagnosis in tracheobronchopathia osteochondroplastica.
