Pepper mild mottle virus (PMMoV), one of the most prevalent viruses in chili pepper (Capsicum annuum L.) is a non-enveloped, rod-shaped, single-stranded positive-sense RNA virus classified in the genus Tobamovirus. The supernatants of five bacterial cultures (Pseudomonas putida [PP], Bacillus licheniformis [BLI], P. fluorescens [PF], Serratia marcescens [SER], and B. amyloliquifaciens [BA]) were analyzed to find novel antiviral agents to PMMoV in chili pepper. Foliar spraying with supernatants (1:1, v/v) obtained from Luria-Bertani broth cultures of PP, BLI, PF, SER, and BA inhibited PMMoV infection of chili pepper if applied before the PMMoV inoculation. Double-antibody sandwich enzyme-linked immunosorbent assay showed that treatments of five supernatants resulted in 51-66% reductions in PMMoV accumulation in the treated chili pepper. To identify key compounds in supernatants of PP, BLI, PF, SER, and BA, the supernatants were subjected to gas chromatography-mass spectrometry. The 24 different types of compounds were identified from the supernatants of PP, BLI, PF, SER, and BA. The compounds vary from supernatants of one bacterial culture to another which includes simple compounds— alkanes, ketones, alcohols, and an aromatic ring containing compounds. The compounds triggered the inhibitory effect on PMMoV propagation in chili pepper plants. In conclusion, the cultures could be used to further conduct tissue culture and field trial experiments as potential bio-control agents.
An Approach to Clone the Biosynthetic Genes of 2-deoxystreptamine Containing Aminoglycosides

Despite the most of the 2-deoxystreptamiue (2-DOS) containing aminoglycosids are produced from Streptomyces, very little information existe about their biosynthetic genrs; cloming and characterization. 2-deoxy-scyllo0inosose synthaes (2-DOIS) was osolated from S. kanamyceticus and expressed in heterologous host. We have isolated 2-DOS biosynthetic genes; 2-deoxy-scyllo-inosose synthase, 2-deoxy-scyllo-inosose aminotransferase and aminoglyccoside resistance ribosomal methylases, by PCR experiment using the primers designed from the conserved region of respective genes. The sequencing of PCR amplified DNA fragments reveled their good homology with btrC and btrB of Bacillus cirulans, the sole 2-DOIS gene exists in the database until now, and btrB of Bacillus circulans the sole 2-DOIS gene exists in the database until now, and with other ami noglycodide resistance ribosomal methylases. As a goal for isolation of biosynthetic gene cluster of tobramycin, a cosmid, pST51 was isolated and sequencing of this cosmid is under progress. The PCR primers are specific and reliable to isolate the biosymthetic genes of 2-DOS containing aminoglycosides or for screening the aminoglycoside producers. The use of 2-DOIS as probe can save both time and cost for cloning aminoglycoside biosynthetic genes.

<P>A cluster of genes for ribostamycin (Rbm) biosynthesis was isolated from Streptomyces ribosidificus ATCC 21294. Sequencing of 31.892 kb of the genomic DNA of S. ribosidificus revealed 26 open reading frames (ORFs) encoding putative Rbm biosynthetic genes as well as resistance and other genes. One of ten putative Rbm biosynthetic genes, rbmA, was expressed in S. lividans TK24, and shown to encode 2-deoxy-scyllo-inosose (DOI) synthase. Acetylation of various aminoglycoside-aminocyclitol (AmAcs) by RbmI confirmed it to be an aminoglycoside 3-N-acetyltransferase. Comparison of the genetic control of ribostamycin and butirosin biosynthesis pointed to a common biosynthetic route for these compounds, despite the considerable differences between them in genetic organization.</P>
Prediction Based Cost Estimation Model Using Clusters

On some parameters Software cost estimation based on development and effort on the project. Every line of code and every method are important to estimate the cost of project. To develop the software project using well defined scientific principles, methods, statements and functions, though there are many costs estimation models are available to estimate the cost of project, but still there is lack of efficient system to predict the cost estimation based on size of project, functionality of project, methods of project. In the existing system cost estimation is done on the basis of lines of code, resource management, in time process. Because of various reasons in the existing system we have identified the drawbacks. In this paper, cost prediction model is defined based on clusters, the number of clusters will form the centroids. Each centroid has a specific number of clusters. Each cluster will be called as rule. In this paper, results will show the prediction of cost estimation is defined.

Permeability is the ability of a material to transmit fluid/gases. It is an important material property and it depends on mould parameters such as grain fineness number, clay, moisture, mulling time, and hardness. Modeling the relationships among these variable and interactions by mathematical models is complex. Hence a biologically inspired artificial neural network technique with a back-propagation-learning algorithm was developed to estimate the permeability of green sand. The developed model was used to perform a sensitivity analysis to estimate permeability. The individual as well as the combined influence of mould parameters on permeability were simulated. The model was able to describe the complex relationships in the system. The optimum process window for maximum permeability was obtained as 8.75-10.5% clay and 3.9-9.5% moisture. The developed model is very useful in understanding various interactions between inputs and their effects on permeability.
Mesoporous silica (MCM-41) effect on (PEO+LiAsF<sub>6</sub>) solid polymer electrolyte

<P><B>Abstract</B></P><P>Composite polymer electrolyte films consisting of polyethylene oxide (PEO), LiAsF<SUB>6</SUB> and mesoporous silica (MCM-41) with fixed PEO/LiAsF<SUB>6</SUB>=90/10 but different weight percent ratios of MCM-41 were prepared using the solution casting method. The polymer electrolyte films were characterized using XRD, DSC, SEM and electrical impedance spectroscopy. In corporation of MCM-41 in a (PEO+LiAsF<SUB>6</SUB>) polymer electrolyte facilitates salt dissociation, enhances ion conductivity, and improves miscibility between organic and inorganic moieties. The scanning electron microscopy (SEM) photographs indicates the electrolytes are miscible and homogeneous up to 10wt.% of MCM-41, and an optimized conductivity is found at this composition (10wt.%). However, at higher weight ratios (>10wt.%), the Li/MCM-41-rich domain developed, and the conductivity decreased with increasing mesoporous material. The electrochemical performance of fabricated electrochemical cells of configuration Li/(PEO+LiAsF<SUB>6</SUB>+MCM-41)/(MoO<SUB>3</SUB>+C+PTFE) were investigated.</P>

Genomic libraries of S. ribosidificus and S. fradiae were constructed for sorting out the biosynthetic genes for 4, 5 disubstitued-2-deoxystreptamine (DOI) containing amino glycosides antibiotics. Screening over 10,000 colonies of each library by the conserved region-based probes revealed 6 positive cosmids. Sequencing about 40kb-region reveled 25 and 29 open reading frames (ORFs) putatively involved in ribostamycin and neomycin biosynthesis. The genetic organization of these gene clusters is in good agreement with the previously reported butirosin biosynthetic genes cluster. Furthermore acetylation by RbmJ of several amino glycosides was also conformed by in vitro enzyme assay of the purified protein.
