        Development and comparison of a porcine gelatin detection system targeting mitochondrial markers for Halal authentication

        <P><B>Abstract</B></P> <P>This study aimed to compare the sensitivity of porcine-specific primers targeting different mitochondrial DNA for the detection of porcine gelatin in ramen stock powder designated “Halal” using SYBR Green I real-time PCR. Four mitochondrial DNA markers (D-loop, ATP8, ND5, and 12S rRNA) were used to detect the porcine gelatin DNA. Extraction of DNA from porcine gelatin and spiked mixtures was successfully performed using a DNeasy Mericon Food Kit. Each primer had high porcine specificity for D-loop, ATP8, and ND5 genes, but not the 12S rRNA gene. Also, the detection limit for each primer was tested in a serial dilution of porcine gelatin DNA (10, 1, 0.1, 0.05, 0.01, and 0.005 ng) and ramen stock powder spiked with porcine gelatin (1, 0.1, 0.05, and 0.025%). Assays targeting both D-loop and ND5 genes were able to detect a level as low as 0.01 ng, while the detection limit for the ATP8 gene was 0.05 ng in pure porcine gelatin. The detection limits of D-loop, ATP8, and ND5 genes were 0.05%, 0.1%, and 1% (w/w) in ramen stock powder spiked with porcine gelatin, respectively. In conclusion, a novel detection system targeting the mitochondrial D-loop was superior to the others and could be an effective and simple method to authenticate Halal products.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A porcine gelatin detection system was developed based on mitochondrial genes. </LI> <LI> Among four assays, mitochondrial D-loop assay was the most reliable and sensitive. </LI> <LI> This assay will provide simple and efficient detection system. </LI> <LI> This assay will authenticate halal products including ramen stock powder. </LI> </UL> </P>

        노인부부가구 남편과 아내의 삶의 만족도에 영향을 미치는 특성들

        The purpose of this study was to investigate individual characteristics and spousal characteristics influencing elderly husbands`` and wives`` life satisfaction. The third wave of Korean longitudinal study of Aging in 2010 was used. The sample for this study was consisted of 696 elderly couples over 65 years old who lived by themselves. Hierarchical multiple regression analysis based on couple unit was performed for both husbands`` and wives’ life satisfaction respectively. Independent variables were participants’ and their spouse’s age, education level, job engagement, chronic disease, marital satisfaction, social participation as well as their spouse``s life satisfaction. Results indicated that common predictors of life satisfaction of both husbands and wives were individual marital satisfaction, social participation, and spouse``s life satisfaction. In contrast, there were also gender specific predictors. Husbands`` employment status was associated with only husbands`` life satisfaction while husbands`` chronic disease was associated with wives`` life satisfaction. The present results suggest a crossover effect in life satisfaction between elderly husbands and wives and imply that elderly couples have a interdependent relationship with each other in terms of life satisfaction.

        벨리댄스 지도자 신뢰와 무용몰입에 영향을 미치는 선행요인 : 변혁적 리더십, 비언어적커뮤니케이션, 코칭지식

        This study would empirically analyze the impacts of the belly dance instructor’s transformational leadership, nonverbal communication, and coaching knowledge on trust in the instructor and dance commitment perceived by belly dance participants. To do so, this study surveyed the participants who participated in a belly dance class and completed collecting a total of 345 effective responses. Based on the data collected, this study conducted a structural equation model analysis, using SmartPLS statistical analysis software. Accordingly, the specific statistical analysis results are as follows. First, the belly dance instructor’s transformational leadership and nonverbal communication had statistically significant positive (+) impacts on trust in the instructor perceived by belly dance students. Second, the belly dance instructor’s nonverbal communication did not have a statistically significant impact on trust in the instructor perceived by the belly dance students. Third, trust in the instructor perceived by the belly dance students had a statistically significant positive (+) impact on dance commitment. Fourth, in the path from transformational leadership through trust in the instructor to dance commitment, trust in the instructor had a mediating effect. Also, in the path from coaching knowledge through trust in the instructor to dance commitment, trust in the instructor had a mediating effect.

        공연예술기획자의 직무역량 척도개발 및 타당화

        This study was conducted by the following procedures to develop and validate a scale of job competences of performing art planners. First, with the valid samples of 109 persons of performing arts, a preliminary survey was conducted to draw the necessity of research. Second, a primary survey was conducted with 10 experts in performing arts, including professors of graduate schools of performing arts, doctors of performing arts studies, and the representatives of performance planning organizations to draw the results through the verification of content validity. Third, a secondary survey was conducted with the valid samples of 182 persons in the fields of performing arts, including theater, opera, musical, dance, music, and stage production arts to verify the validity and drew the following research results. Performing art planner’s job competencies were developed with a scale of a total 34 questions. As for the detailed areas, findings were presented with eight work profession questions, three qualification and character questions, five personnel management questions, six leadership questions, five fund-raising and financial management questions, and seven marketing area questions. This study was conducted with the following research methods. First, by descriptive statistics for questions, this study checked the normality with mean, standard deviation, skewness and kurtosis. Second, this study investigated the reliability, calculating question-total score correlation, Cronbach’s α and checked the factor structure of the scale with exploratory factor analysis. Third, this study drew up a measurement model for the scale and conducted confirmatory factor analysis to come up with the final model. Based on this, this study verified concept reliability, convergence validity, and discrimination validity to investigate the validity and checked cross validity. As for the analysis tools, SPSS 25 and AMOS 25 were employed.

        벨리댄스 참여자가 인식하는 지도자 주요 역량 탐색 및 요구도 분석 연구

        This study investigated 305 in-service belly dance participants and analyzed demands, aiming to explore the major indices of the instructor’s competencies perceived by the belly dance participants and investigate the prioritized competencies. For statistical processing, analysis was conducted, using SPSS WIN 21.0. As for research methods, to analyze the differences and order of priority between the instructor’s important competencies and retained competencies, a t-test and Borich demand analysis were conducted. The order of priority was drawn, using the Locus for Focus model. The results of the analysis are as follows: In all detailed competencies, including class planning ability and leadership in the professional competency cluster; attention and affection, and patience in the personality competency cluster; communication in the communication competency cluster; and appearance management and image management in the self-regulation competency cluster, there were statistically significant differences between importance level and retention level, and of the detailed competencies of motivation in the leadership competency cluster, the detailed competencies of participant management in the communication competency cluster and the detailed competencies of expressiveness in the artistic competency cluster, there were statistically significant differences between importance level and retention level. In addition, belly dance participants’ demands for the instructor’s teaching competencies were in the following order: personality, professional, communication, self-regulation, leadership, and artistic competency.

        벨리댄스 지도자의 이미지와 비언어적커뮤니케이션이 참여자 재등록 의사에 미치는 영향 : 참여만족의 매개효과

        This study would empirically analyze the impacts of a belly dance instructor’s image and nonverbal communication on participants-perceived re-registration intention. In addition, this study would test if the belly dance participants’ participation had a mediating effect on their re-registration intention. Concerning this, this study completed the collection of 342 valid participants in a class higher than the general class of agencies for training belly dance instructors nationwide. Based on the collected data, this study presents the following concrete statistical analysis results, using Statistical Analysis Software, SPSS 22. First, the belly dance instructor’s image had a statistically significant positive (+) impact on participants’ satisfaction. Second, the belly dance instructor’s nonverbal communication had a statistically significant positive (+) impact on participants’ satisfaction. Third, belly dance participants’ satisfaction had a statistically significant positive (+) impact on their re-registration intention. Fourth, satisfaction showed a mediating effect in the path from the belly dance instructor’s image through satisfaction with participation to re-registration intention. Fifth, satisfaction also showed a mediating effect in the path from the belly dance instructor’s nonverbal communication through satisfaction with participation to re-registration intention. In other words, the higher the image and nonverbal communication of the belly dance instructor, the more satisfied the participants become, and the higher their re-registration intention becomes.

        This study sought to ascertain and describe u-Health nurses (ubiquitous healthcare nurse, tele-nurse)` experiences working with low-income elders in Korea. Methods: Data were collected through semi-structured interview, focus group interviews of 6 nurses and participant observations in community places in public rental housing in the city of Seoul. The ethnographical analysis method, proposed by Spradley, was used to analyze interview data. Results: We identified two major clusters of themes on common issues and concerns experienced by nurses: "work environment" and "daily work life." In addition, ten themes and fourteen sub-themes were extracted from the field work data. Conclusion: The study provided a deeper understanding of the experiences and services of u-Health nurses in Korea, of which much remains to be understood, as the healthcare services they currently provide are limited. Moreover, its findings could empower novice nurses and expand nurse`s professional knowledge and competence.

      • Liver‐Specific and Echogenic Hyaluronic Acid Nanoparticles Facilitating Liver Cancer Discrimination

        <P><B>Abstract</B></P><P>With the increasing demand for instant real‐time ultrasound (US) imaging of a specific organ, target‐specific and long‐circulating ultrasound contrast agents are of special interest. A new species of echogenic hyaluronic acid nanoparticles is presented as an ultralong‐acting, liver‐specific, US contrast agent that is distinct from conventional gas‐filled microbubbles. Using an oil‐in‐water (O/W) emulsification method, bioinert and hydrophobic perfluoropentane (PFP) is encapsulated as an ultrasound gas precursor into hyaluronic acid nanoparticles (HANPs) using hydrophobic interactions. HANPs are formulated by self‐assembly, with amphiphilic hyaluronic acid‐5β‐cholanic acid (HA‐CA) conjugating in aqueous conditions. The resulting echogenic PFP‐encapsulated HANPs (Echo‐NPs) show solid nanostructures, differentiated from core‐empty conventional microbubbles, and exhibiting outstanding physical properties as an ultrasound contrast agent. They are more stable and robust echogenic solid bodies with an in vivo favorable hydrodynamic size and because PFPs vaporize gradually, their expansion process is very slow in body conditions. After several systemic circulations, echo‐NPs generated intense and ultralong echo signals for US imaging at the target site. The echogenic properties of Echo‐NPs show a significantly increased half‐life and echo persistence, compared with conventional microbubbles. The results clearly show that echo‐NPs outperform conventional microbubbles in terms of both physical and echogenic in vitro and in vivo properties.</P>

        스위칭 손실을 줄인 1700 V 4H-SiC Double Trench MOSFET 구조

        본 논문에서는 CDT(Conventional Double Trench) MOSFET보다 스위칭 시간과 손실이 적은 1700 V EPDT(Extended P+ shielding floating gate Double Trench) MOSFET 구조를 제안하였다. 제안한 EPDT MOSFET 구조는 CDT MOSFET 에서 소스 Trench의 P+ shielding 영역을 늘리고 게이트를 N+와 플로팅 P<SUP>-</SUP> 폴리실리콘 게이트로 나누었다. Sentaurus TCAD 시뮬레이션을 통해 두 구조를 비교한 결과 온 저항은 거의 차이가 없었으나 Crss(게이트-드레인 간 커패시턴스)는 게이트에 0 V 인가 시에는 CDT MOSFET 대비 32.54 % 줄었고 7 V 인가 시에는 65.5 % 감소하였다. 결과적으로 스위칭 시간 및 손실은 각각 45 %, 32.6 % 줄어 스위칭 특성이 크게 개선되었다. In this paper, 1700 V EPDT (Extended P+ shielding floating gate Double Trench) MOSFET structure, which has a smaller switching time and loss than CDT (Conventional Double Trench) MOSFET, is proposed. The proposed EPDT MOSFET structure extended the P+ shielding area of the source trench in the CDT MOSFET structure and divided the gate into N+ and floating P<SUP>-</SUP> polysilicon gate. By comparing the two structures through Sentaurus TCAD simulation, the on-resistance was almost unchanged, but Crss (Gate-Drain Capacitance) decreased by 32.54 % and 65.5 %, when 0 V and 7 V was applied to the gate respectively. Therefore, the switching time and loss were reduced by 45 %, 32.6 % respectively, which shows that switching performance was greatly improved.

      • Isolation and Identification of Anthocyanins contained Red Petals in Edible Rose

        The petals of edible rose contains diverse component, such as cratgolic acid, malic acid, vitamin C, B2, flavonoids, and lactons. The petals of edible rose are also known to have pharmaceutical effects. They have been used in the treatment of cardiac insufficiency, coronary heart disease, hyperpiesia and inflammatory disease only in folk remedies. Also, the petals of edible rose have various colors, such as red, pink, purple, blue, green, black, white, and yellow. Maybe, such color pigmentations are due to accumulation of anthocyanins, chlorophylls, flavonoids, and various combinations of breakdown products of these pigments. Despite many research on rose, report on the exact composition of anthocyanins in petal of edible rose is limited. Therefore, the objective of this study is to isolate and identify the anthocyanins in red petals of edible rose (Rosa hybrida cv. Noblered). Major anthocyanin pigments of the red petals in edible rose were extracted with 0.5% HCl - 20% MeOH for 24hrs. at room temperature, and purified using C18 open-column chromatography and prep-HPLC method. Two major anthocyanins were isolated, and their chemical structures were identified by spectroscopic methods (UV-VIS, LC-MS, and NMR). The λmax of anthocyanin 1 was 512nm, and m/z = 611[M+], while anthocyanin 2 had maximum absorbance at 501nm, and m/z = 595[M+], resulting in the two compounds are different. The complete structures of these anthocyanins were elucidated as cyanidin-3,5-diglucoside, and pelargonidin-3,5-diglucoside, respectively.
